Urinary microRNAs are the biomarker for the disease activity of petients with focal segmental glomerulosclerosis

Abstract:Objective: To examined the microRNA expression profile of urine supernatant in patients with focal segmental glomerulosclerosis(FSGS) and analyze the correlation between the level of miRNAs and the activity of disease.Methodology: Urine supernatant samples from patients with active FSGS(urinary protein excretion ≥3.5 g /24h),nonactive FSGS(urinary protein excretion ≤0.4 g /24h) and normal controls were tested by quantitative Real-time PCR.The correlation between the urinary miRNAs expression levels and clinical indexes were analyzed.In order to evaluate the specificity,urinary miRNAs level of patients with membranous nephropathy(MN) and diabetic nephropathy(DN) was also detected for validation.Results: The urinary expression levels of miRNA-196a,miRNA-30a,miRNA-155 and miRNA-490 were significantly elevated in active FSGS compared to the non-active FSGS patients and normal control(P < 0.001).The expression level of miRNA-196a,miRNA-30a,miRNA-155 and miRNA-490 was correlated with clinical indexes,such as proteinuria and serum albumin level(P < 0.05).However,the same change of miRNA-196a,miRNA-30a and miRNA-490 was not found in the urine of MN and DN patients.Conclusion: Urinary expression level of miRNA-196a,miRNA-30a and miRNA-490 correlated with disease activity suggested that miRNA-196a,miRNA-30a and miRNA-490 might be the new urinary biomarker for disease activity and therapeutic effects of FSGS.
