Comparison of three different fixative methods of mouse retinal paraffin section

Abstract:Objective To search for an effective and cheap fixative method of mouse retinal paraffin section.Methods About 30 BALB/C mice were randomly divided into 3 groups,10 rats in each group.All rats were killed to take the eyeballs.Rats in group A were treated with dehydration for 1 hour in absolute ethyl alcohol at twice times.Rats in group B were treated with the same dehydration as group A,but not removed cornea,iris and lens.Rats in group C were treated with dehydration for 30 minutes with 55%,65%,75%,85% and 95% alcohol,the cornea,iris and lens were removed as group A,and then treated with dehydration for 30 minutes in absolute ethyl alcohol for twice times.The other procedures were same with group A.after staining of slice,completeness of retinal structure in each group were compared and rates of retinal detachment were analyzed.Results In group B,part of outer nuclear layer broke,IS and OS layers were with extensive dissection.In group A,the lens was removed,outer nuclear layer was complete,but IS and OS layers were with partly broken.In group C,lens was removed and time of dehydration was shortened,retinal structure was complete,staining was obvious and with clear levels.Obscission of retinal pigment epithelial layer as the condition of retinal detachment showed that rate of retinal detachment were 70% in group A with mainly at peripheral 1/2 part,65% in group B with uncertain place and various degree of fragmentation and rupture,10% in group C with mainly at peripheral 1/4 part.Conclusion In filmmaking of rat eyeball,remove of lens,improvement of alcohol concentration,shortening of dehydration time can effectively decrease the deformation of retinal tissues,and help for exact location of retina.
