Effects of Phosphorus Supply on Copper Tolerance and Ascorbate-Glutathione Cycle in Maize (zea Mays L.) Seedlings

Abstract:Maize (Zea mays L.) seedlings were exposed to normal Cu (CuL, 1 mu m) and excess Cu (CuM, 100 mu m and CuH, 1 mM) under two P levels: normal (1 mM, +P) and excess (10 mM, +HP). During 72 h treatment, the effects of P supply were investigated on Cu accumulation, growth of plant, the contents of malondialdehyde (MDA) and pigment, ascorbate (AsA)-glutathione (GSH) cycle. Compared with the treatment of normal Cu, excessive Cu significantly increased lipid peroxidation indicated by MDA. The leaves showed apparent symptoms of Cu toxicity and the plant growth was significantly inhibited by excess Cu. +HP plants experienced lower oxidative stress owing to a decrease Cu accumulation. +HP supply inhibited the appearance of Cu toxicity symptoms, and improved plant growth. And +HP supply had no significant modulation on AsA-GSH cycle. The result also suggested the nonenzymatic system, including pigment, AsA and GSH, played a main role in antioxidate process at early stress stage, while the enzymatic system was crucial to the delay of oxidative damage at later stress stage. Therefore, adequate P supply to the plants may inhibit Cu accumulation and enhance their Cu tolerance capacity.
