Abstract:Herein, an atom-economical and eco-friendly electrochemical oxidation/cyclization of glycine derivatives through intramolecular Shono-type oxidative coupling is disclosed, leading to a variety of 2-substituted benzoxazoles in 51-85% yields. This oxidative cyclization proceeded in transition metal- and oxidant-free conditions and generated H-2 as only a byproduct. Additionally, gram-scale reactions and a broad substrate scope demonstrated the synthetic usefulness of this protocol.

Abstract:The 1,3,4‐oxadiazole is a widely encountered motif in the areas of pharmaceuticals, materials, and agrochemicals. This work has established a mild, mediated electrochemical synthesis of 2,5‐disubstituted 1,3,4‐oxadiazoles from N‐acyl hydrazones. Using DABCO as the optimal redox mediator has enabled a mild oxidative cyclisation, without recourse to stoichiometric oxidants. In contrast to previous methods, this indirect electrochemical oxidation has enabled a broad range of substrates to be accessed, with yields of up to 83%, and on gram scale. The simplicity of the method has been further demonstrated by the development of a one‐pot procedure, directly transforming readily available aldehydes and hydrazides into valuable heterocycles.

Abstract:An effective method for the synthesis of benzazoles through the electrochemical oxidative cyclization of o‐aminophenol/o‐aminothiophenol/o‐phenylenediamine and aldehydes was developed. A series of benzazoles were efficiently synthesized by using this method in good yields. Furthermore, this method was applied to the synthesis of drug thiabendazole in a gram‐scale, which proved the practicality of the method. This approach avoids using catalysts and oxidants, and has the advantages of wide substrate range, mild reaction conditions and good tolerance of functional groups. Finally, the possible reaction mechanism was proposed and supported by cyclic voltammetry (CV) and the control experiments.

Abstract:An electro-oxidative cyclization pathway in which hydrazones are selected as starting materials to generate amphiphiles by reacting with benzylamines and benzamides was reported. This strategy successfully prepared a series of 1,2,4-triazoles in satisfactory yields. Moreover, the use of cheap stainless steel as the anode, the feasibility to conduct the transformation as a one-pot reaction and the proof that scaling-up these reactions is possible make this transformation attractive for potential application in industry.

Abstract:With the help of the in‐situ electrogenerated iodine radicals and I+ cations, aromatic ketones with benzylamines could be smoothly converted to polysubstituted oxazoles. This electrosynthetic route has the advantages of easily available substrates, cheap electrode materials and mild reaction conditions. With KI as the supporting electrolyte and a mediator, DMSO as the solvent and KHCO3 as the base, a convenient and efficient electrochemical route was developed for the synthesis of polysubstituted oxazoles from aromatic ketones and benzylamines. The electrosynthesis was smoothly carried out in an undivided cell at room temperature, affording the target products in moderate to high yields.

Abstract:An efficient N-H and C(sp3)-H functionalization of aryl ketones with benzylamines/amino acids was developed under mild conditions by virtue of anodic oxidation. A variety of functionalized 2,5-diaryloxazoles were obtained with good to excellent yields. Moreover, some important natural products can be prepared by this method. Abstract:An electrochemically promoted coupling of benzoxazoles and amines has been developed, leading directly to the formation of 2-aminobenzoxazoles. The chemistry utilizes catalytic quantities of a tetraalkylammonium halide redox catalyst and is carried out under constant current conditions in a simple undivided cell. The use of excess chemical oxidant or large amounts of supporting electrolyte is avoided. This greatly simplifies the workup and isolation process and leads to a reduction in waste.

Abstract:A metal- and oxidizing reagent-free electrochemical dehydrogenative annulation of enamides with O -nucleophiles has been developed, leading to a series of 4-oxo-oxazolines under environmentally friendly conditions. The present method demonstrates broad substrate scope and wide functional group tolerance. In addition to water, alcohols and carboxylic acids were also competent reaction partners. The merit of this electrochemical protocol has also been demonstrated by the gram-scale reaction and the synthesis of 2-(3,4-dimethoxyphenyl)-5-methyl-4,5-dihydrooxazol-4-ol (an antitumor nature product precursor). The mechanistic results suggest that the formation and homolytic cleavage of the N–Br bond is key to the success of this electrochemical dehydrogenative annulation reaction.

Abstract:A novel electrochemical route has been developed for the synthesis of 1,5-disubstituted and 1-aryl 1,2,4-triazoles from aryl hydrazines, paraformaldehyde, NH4OAc, and alcohols. In this multicomponent reaction system, alcohols act as solvents as well as reactants and NH4OAc is used as the nitrogen source. With the assistance of reactive iodide radical or I2 and NH3 electrogenerated in situ, this process could effectively avoid the use of strong oxidants and transition-metal catalysts and be smoothly carried out at room temperature to give a wide array of 1,2,4-triazole derivatives in good to high yields. Preliminary studies reveal that the reaction mechanism involves a radical process.

Abstract:Herein we report an electrochemical strategy for the construction of polysubstituted oxazoles from easily available ketones and acetonitrile under room temperature. The method shows high efficiency, a broad substrate scope, and requires no external chemical oxidant. Mechanistic investigations suggest that the reaction proceeds through a Ritter-type reaction/oxidative cyclization using acetonitrile as the reactant and solvent. X-ray crystallographic analysis reveals that the reaction involves the attack of acetonitrile on the carbonyl carbon instead of the α-carbon.

Abstract:Benzoxathiazine dioxide, as a bioisostere of the clinically widely used diazoxide, exhibits interesting biological activity. However, limited success has been achieved in terms of its concise and direct synthesis. We report herein a facile electrochemical migratory cyclization of N-acylsulfonamides to access a diverse array of benzoxathiazine dioxides. The inclusion of electrochemistry is crucial for realizing such a novel transformation, which is substantiated both by the experiments and density-functional-theory calculations.

Abstract:With formaldehyde as an atom-economical carbonyl synthon, the EtOH-catalyzed electrochemical multicomponent synthesis of various imidazolidine-fused sulfamidates was developed. A wide range of products were obtained in good to excellent yields under organic oxidant-free, energy-saving and mild conditions. The mechanistic study revealed that the in situ generated ethoxide played a crucial role in producing aminomethylene radicals at a lower voltage.

Abstract:Diazo compounds are privileged carbene precursors in synthetic organic chemistry. Despite their versatility in both chemo- and biocatalytic synthesis, their preparation typically requires the use of reagents that are expensive, toxic, and unsustainable. Herein, we describe a chemoenzymatic strategy for the preparation of stabilized diazo compounds enabled by enzymatic halide recycling by vanadium-dependent haloperoxidase (VHPO) enzymes. The process involves the conversion of a carbonyl-containing compound to an intermediate hydrazone that is subjected directly to a VHPO-catalyzed nitrogen-nitrogen (N-N) bond oxidation to the corresponding diazo compound. The protocol is applied to a broad range of benzoylformate and isatin derivatives. Molecular docking experiments provide insight into reactivity rate differences between (E)- and (Z)-configured hydrazones in the VHPO-mediated oxidation process. Finally, the developed method is interfaced with lipase-mediated transacylation to produce a collection of diazo derivatives starting from a single benzoylformate starting material.
