Abstract:Objective:To optimize separation and purification technology of chlorogenic acid in leaves of Lonicera japonica.Method:With the content of chlorogenic acid as index,static and dynamic absorption,desorption properties of polyamide resin were observed by single factor test,and sample amount,the amount of washing water,type and dosage of eluent were investigated;With purity and retention rate as index,effects of elution flow rate,the concentration and volume of eluent on separation and purification technology was investigated by orthogonal test.Result:Optimized separation and purification technology was as following:sample amount 10 mg·g-1 resin,washed impurity with 2 BV water,and eluated with 3 BV the amount of 65% ethanol,flow rate of 0.4 BV·h-1,collected eluent.Conclusion:This optimized technology was stable and practicable,it could provide reference for industrial separation and purification of chlorogenic acid.

Abstract:The effects of extraction time, temperature, pressure and different concentration of ethanol and their interactions on the yields of amentoflavone, quercetin and ginkgetin extracted from Taxus chinensis by supercritical CO2 were investigated by using a central composite design (CCD). An CCD experimental design with four factors and five levels was used to optimize the extraction parameters. Ultra performance liquid chromatography (UPLC) was used to analyze the content of the tree components in the extracts. Experimental results show that the main effects of factors and their interactions are significant on the yields (p < 0.05). The optimal extraction conditions were established for the three compounds: yield of 4.47 mg/g for amentoflavone at 48 °C, 25 MPa, 2.02 h and 78.5% ethanol, 3.73 mg/g for quercetin at 46 °C, 24 MPa, 2.3 h, 82% ethanol and 3.47 mg/g for ginkgetin at 48 °C, 20 MPa, 2.38 h, 82% ethanol, respectively.
Purification of d-a-tocopheryl polyethylene glycol 1000 succinate (TPGS) by a temperature-modulated silica gel column chromatography: use of Taguchi method to optimize purification conditions.

Abstract:The demand for high purity d-a-tocopheryl polyethylene glycol 1000 succinate (TPGS) is increasing with the exploitation of TPGS-related products. Previously, we synthesized a TPGS mixture by esterifying vitamin E succinate with polyethyleneglycol 1000. In this study, a temperature-modulated silica gel chromatographic column was used to purify the synthesized TPGS. Taguchi method was used to optimize purification conditions associated with column temperature, loading amount, feedstock concentration and flow rate of mobile phases. Purification efficacy under the Taguchi optimized conditions was predicted theoretically and the predicted results were verified experimentally. High-performance liquid chromatography was used to quantify the unpurified and purified TPGS. The Taguchi-based analysis separately produced an optimum combination of purification conditions for TPGS purity and recovery. Under the optimized conditions, both the theoretical prediction and the confirmatory experiment yielded TPGS purity and recovery approximating to 98% each. Impressively, the study also found that column temperature had a considerable effect on purification efficacy, in particular on TPGS purity, although it was a less influential factor compared to loading amount and feedstock concentration.

Abstract:ObjectiveTo investigate the influences of different parameters on the process of alcohol precipitation and to assure the optimal conditions in this study. MethodsThe influences of the concentration of alcohol,the relative density of herb liquor,the speed of adding alcohol and setting temperature on the retenthor rate of active components in Cinobufacini alcohol precipitation process were investigated by single factor experiment.Then an orthogonal test was conducted by the four factors with greater influence.The optimization of technology was performed by using orthogonal design and multi-index compositive score method. ResultsThe optimal process parameters were the relative density of herb liquor 1.10,the final concentration of alcohol 82%,the concentration of alcohol 75% and the settling temperature 10℃. ConclusionThe method is stable and feasible,and has widely industrial application prospect.
Optimization of Alcohol Precipitation Process of Salviae Miltiorrhizae Water-extraction Solution by Orthogonal Design with Multi-index Comprehensive Evaluation

Abstract:An orthogonal design with multi-index comprehensive evaluation was adopted to optimize the alcohol precipitation process of Salviae miltiorrhizae water-extraction solution with impurity removal and retention rate of total salvianolic acids as indexes.The optimized process was to add 90% ethanol to 1.20 g/ml of Salviae miltiorrhizae water-extraction solution at a constant rate of 60.9 ml/min and stirring speed of 450 r/min until the final ethanol concentration of 80%.The impurity removal and retention rate of total salvianolic acids were 6.03 g and 67.2%.

Abstract:Objective: To study the optimum technical conditions of extracting Hydroxysafflor yellow A ( HSYA) from Carthmus tinctorius by multi-stage countercurrent extraction technology. Method: The effects of extraction time of each stage, extraction temperature, ethanol concentration and solid-liquid ratio (g • mL-1 ) on extraction yield of HSYA were studied by orthogonal test design and the comparison of other extraction methods were presented. Result: Extraction time and solid-liquid ratio had significant influence on the extraction yield, and the optimum parameters were as follows: Extraction time of each stage was 120 min, solid-liquid ratio was 1:10 (g • mL-1), ethanol concentration was 30% , and extracted at room temperature. Under the optimum conditions, the extraction yield of HSYA was 1.56% and the purity of the extract was 6.06%. Compared with the traditional extraction method and the ultrasonic extraction method of the pharmacopoeia, the extraction yield was increased by 6.12% and 9.09% , the purity of extract was increased by 42.9% and 27.0% , respectively. Conclusion: The multi-stage countercurrent extraction technology has many advantages such as simple operation, less solvent consumption, higher extraction yield and purity of extract and it has wide industrial application prospect.

Abstract:Ethanol precipitation is an important separation and purification process in the traditional Chinese medicines (TCMs) industry. In the present study, a membrane dispersion micromixer was applied to achieve good mixing for the ethanol precipitation process of Astragali radix concentrate. New experimental apparatus was set up to rapidly lower the temperature of ethanol solution before mixing with the concentrate. Ethanol precipitation process was optimized according to Quality by design concept. To identify critical material attributes (CMAs), ten batches of Astragali radix were used to prepare concentrates. Calycosin-7-O-β-D-glucoside content, the sucrose content, and the electrical conductivity were found to be CMAs after the correlation analysis and stepwise regression modelling. Definitive screening design was used to investigate the relationships among critical process parameters, CMAs, and process critical quality attributes (CQAs). Quadratic models were developed and design space was calculated according to the probability of attaining process CQA standards. A material quality control strategy was proposed. High quality and low quality Astragali radix concentrates can be discriminated by the inequalities. Low quality Astragali radix concentrates should not be released for ethanol precipitation process directly. Verification experiment results indicated accurate models and reliable design space. The temperature control method and control strategy are promising for ethanol precipitation process of other TCMs or foods.

Abstract:Ethanol precipitation is a widely applied separation technology in the production of botanical medicines, but the research on this technology is few. In this work, ethanol precipitation is used to purify the water extract of Danshen (Salvia miltiorrhiza Bge.) and Chuanxiong (Ligusticum chuanxiong Hort.) as a sample to describe its separation characteristics and mechanism. The effects of refrigeration temperature, ethanol addition, and supernatant pH value were investigated. Total saccharide removal and dry matter removal increased as refrigeration temperature decreased or ethanol addition increased. Danshensu (DSS) recovery and salvianolic acid B (SaB) recovery increased as supernatant pH value decreased or ethanol addition decreased. The purity of phenolic compounds increased after ethanol precipitation. The losses of caffeic acid (CA) and ferulic acid (FA) in ethanol precipitation were caused by chemical transformation. The losses of DSS and SaB were both caused by chemical transformation and precipitation. By using a simple model, the pK(a) values of DSS and SaB in supernatant can be calibrated, which were 3.63 and 3.56, respectively. The solubilities of DSS salt and SaB salt can also be calibrated. Most of DSS and SaB existed in supernatant in their ionic forms. The results of this work will contribute to the implementation of Quality by design (QbD) concept in the manufacture of botanical medicines. (C) 2013 Elsevier B.V. All rights reserved.

Abstract:Ethanol precipitation is an important purification process affecting the safety and efficacy of botanical injections. In this paper, the second ethanol precipitation of Guanxinning injection was optimized with response surface methodology as a sample to represent the principles of indicator selection. The influences of three factors, including dry matter content of concentrated supernatant (DMCCS), refrigeration temperature, and the mass ratio of ethanol and concentrated supernatant (E/CS ratio) were explained based on the precipitation mechanism. To characterize the effects of ethanol precipitation on drug safety, drug efficacy, and drug quality control, phenolic compound recovery and dry matter removal were selected as optimization criteria. In the optimization, E/CS ratio was set to 3.60. The optimum operating conditions were found at refrigeration temperature of 18.66 degrees C and DMCCS of 0.58. Most of the experimental results at optimum operating conditions agreed well with the predicted results.
The Influences of Concentrate Extract Properties and Ethanol Addition Amount on the Ethanol Precipitation Process of Salvia Miltiorrhiza

Abstract:In this work, 10 batches of Salvia miltiorrhiza concentrate were prepared and purified with ethanol precipitation process.Dry matter content, pH value, conductivity and water content of the concentrates and supernatants were all determined.When more ethanol was used in ethanol precipitation, the pH value of the supernatant generally increased, but dry matter content, water content, and the conductivity decreased.Multivariate linear models were built with the most determination coefficient values greater than 0.7.More than 80% of stachyose was removed in the ethanol precipitation process.The removal rate of fructose, raffinose and sucrose were all higher than 30%.When ethanol addition amount increased, the purity of phenolic acids in the supernatant increased, but the retention of lithosperimic acid and salvianolic acid B decreased.The conductivity and pH value of concentrated extract show relatively small influences on ethanol precipitation indices.When fructose, raffinose, or stachyose contents in the concentrated extract were high, the retention rate of phenolic acids tends to be low on most occasions.The purity and retention rate of phenolic acids in the supernatants were also affected by the purity of phenolic acids in the concentrated.The sugar contents in the concentrate are suggested to be monitored in industry because they significantly affect ethanol precipitation process indices.

Abstract:The optimization of fermentation process,extraction and separation of chaetoglobosin A were studied with HPLC analytical techniques to establish high yield,low cost,pollution-free scale fermentation process.[Results] The fermentation process of chaetoglobosin A was optimized by Uniform Design Method.The fermentation medium in the optimal combination of carbon and nitrogen source were determined sucrose 17 g/L,starch concentration of 6 g/L respectively,peptone concentration was 0.5 g/L and fermented for 13 d.The results showed that eluent ethyl acetate was the suitable solvent for the preparation of test samples while considering both extract ratio and fungicidal activity.Found eluant ethyl acetate,petroleum ether(1∶3),developing solvent system ethyl acetate,petroleum ether,formic acid(37∶63∶1) as the best separation conditions.[Conclusions] The ultimate production of 0.17 g/L under the optimum conditions could be reached,which was increased by 13.3% compared to 0.15 g/L under the original fermentation conditions.

Abstract:OBJECTIVETo suggest a method for discriminating quality control indicators of concentrated solution before traditional Chinese medicine ethanol precipitation.METHODThe second ethanol precipitation (SEP) of Guanxinning was taken as an example, with the concentrated supernatant of first ethanol precipitation (CSFEP) prepared with same dry matter content and the same amount of ethanol as raw materials, to conduct the ethanol precipitation test under the same conditions. The experimental data was analyzed by stepwise regression and partial least squares regression. Both of them selected the parameters which affect phenolic compounds retention ratios in SEP supernatant as the key indicators.RESULTPhenolic compounds contents in SEP supernatant were mainly affected by the contents of danshensu, caffeic acid and salvianolic acid B in CSFEP.CONCLUSIONThe discrimination method can be used to discover key quality control indicators of concentrated solution before ethanol precipitation.

Abstract:Ganmaoling granule is the first brand of domestic cold medicine sales, but its preparation method and process control parameters are relatively rough. Therefore it is urgent to upgrade the technologies of large varieties of traditional Chinese medicine (TCM). This paper focused on the balance between the remove of impurity and the retention of linarin during the process of alcohol precipitation of Ganmaoling granules. The effects of four factors on the process were investigated via single factor experiments. The results showed that the precipitating period, the initial ethanol concentration and the final ethanol concentration had a great effect on retention of linarin while the initial density of the extract has not. Similarly, the initial ethanol concentration, the final ethanol concentration and the initial extract density have a great effect on the yield of dry extract while the time of alcohol precipitation has not. The parameters of alcohol precipitation of Ganmaoling granules were optimized as 16 h of precipitating period, 95% ethanol as the initial reagent, 70% of the final ethanol concentration, and 1.10 of the initial extract density.

Abstract:The utilization of bio-derived solvents holds immense promise in the extraction of natural products. In the present study, a systematic evaluation was conducted to assess the performance of bio-derived solvents in simultaneously extracting polysaccharides and phenolic substances from Carthamus tinctorius L. flower. Among them, ethyl lactate (24.7%) exhibited superior efficacy. A comprehensive investigation was also conducted on the salt response behavior of ethyl lactate and its in-situ enrichment efficiency for polysaccharides and phenolics, demonstrating that the recovery yield, enrichment factor, and separation factor can be finely tuned by manipulating the system composition. Based on these findings, we have developed a highly efficient ultrasound-assisted method that is responsive to salt and enables simultaneous extraction and in situ enrichment of polysaccharides and phenolics. In comparison with the method in Chinese Pharmacopoeia (2020), the extraction yield of Carthamus tinctorius L. flower quality markers is increased by more than 1.14 times, while the extraction time is shortened by over 4 times. Additionally, the amount of solvent used is reduced by a factor of 5.73, eliminating the need for methanol and corrosive HCl that are harmful to the environment. The findings of this study will provide crucial theoretical guidance for the application of bio-derived solvents, demonstrating significant potential in terms of quality control and large-scale preparation of natural medicines.

Abstract:Sinomenine hydrochloride is generally produced from Caulis Sinomenii. At present, the purification process in industrial production suffers from large amount of solid waste, high solvent toxicity, and low sinomenine hydrochloride yield. In this study, a new purification process for sinomenine hydrochloride was proposed by using the extract obtained from acid extraction of Caulis Sinomenii as the starting material. The process included the following steps: alkalization, extraction, water washing, acid–water stripping, drying, and crystallization. 1-Heptanol was used as the extractant. The distribution coefficients of sinomenine and sinomenine hydrochloride in 1-heptanol–water system were 27.4 and 0.0167, respectively. The dissociation constants of sinomenine hydrochloride were 8.27 and 11.24, respectively. Process parameters of the new purification process were optimized with experimental design. The extractant 1-heptanol and sinomenine hydrochloride in the crystallization mother solution can be recycled in the new process. The purity of the obtained sinomenine hydrochloride crystals exceeded 85%, and the yield was about 70%. Compared with current industrial processes, safer extractant, less solid waste, and higher sinomenine hydrochloride yield can be achieved using the new purification process of sinomenine hydrochloride provided in this study.
Optimizing the Alcohol Precipitation of Danshen by Response Surface Methodology

Abstract:Alcohol precipitation (AP) is a common method to achieve separation and purification in the process of food and herbal preparations. The response surface methodology (RSM) was applied to study and optimize the AP of Danshen, regarding the recovery of danshensu (RD) and the removal of saccharides (RS). A fractional factorial design was initially performed and solid content, ethanol consumption, acid consumption, and temperature showed significant influence on RD and RS. A Box-Behnken design with 28 experimental runs was then applied and two polynomial equations were established, based on which the effects of significant factors and their interactions were discussed. The optimum operating conditions were solid content at 66.0%, ethanol consumption at 4.3mL/g, acid consumption at 0g/g, and temperature at 10.7 degrees C. The average experimental RD and RS under the optimum conditions were obtained to be 66.25 +/- 1.01% and 61.64 +/- 1.84%, respectively, which agreed well with the predicted values (68.85% for RD and 61.92% for RS).

Abstract:The current methods used to industrially produce sinomenine hydrochloride involve several issues, including high solvent toxicity, long process flow, and low atomic utilization efficiency, and the greenness scores of the processes are below 65 points. To solve these problems, a new process using anisole as the extractant was proposed. Anisole exhibits high selectivity for sinomenine and can be connected to the subsequent water-washing steps. After alkalization of the medicinal material, heating extraction, water washing, and acidification crystallization were carried out. The process was modeled and optimized. The design space was constructed. The recommended operating ranges for the critical process parameters were 3.0-4.0 h for alkalization time, 60.0 degrees C-80.0 degrees C for extraction temperature, 2.0-3.0 (volume ratio) for washing solution amount, and 2.0-2.4 mol.L-1 for hydrochloric acid concentration. The new process shows good robustness because different batches of medicinal materials did not greatly impact crystal purity or sinomenine transfer rate. The sinomenine transfer rate was about 20% higher than that of industrial processes. The greenness score increased to 90 points since the novel process proposed in this research solves the problems of long process flow, high solvent toxicity, and poor atomic economy, better aligning with the concept of green chemistry. [Fast Measurement Method Based on Near Infrared Spectroscopy in Purifying Process of Carthamus Tinctorius Extracts].

Abstract:OBJECTIVE:To really realize quality control of Chinese herb purifying process, near-infrared spectroscopy (NIRS) was used not only for fast monitoring quality-control index of the process, but also for fast judgment of absorption endpoint.METHOD:The purification process of Carthamus tinctorius extracts with nonionic macroreticular resin was selected as an example. HPLC was used as the reference method to determine the content of HSYA. Quantitative and qualitative detection modes of purification were developed by NIRS combined with partial least squares (PLS) and moving block of standard deviation (MBSD).RESULT:The correlation coefficient of the calibration model was 0.999, and the RPD for calibration and validation were above 5, of 5.54 and 5.22, respectively. Based on acquisition spectra, absorption endpoint calculated by MBSD was close to that by HPLC, Only 1 min deviation.CONCLUSION:The method mentioned above is proved to be convenient, rapid and nondestructive, and is applicable for fast monitoring the content of HSYA and fast judgment of absorption endpoint in purifying process of C. tinctorius extracts.

Abstract:Objective:To really realize quality control of Chinese herb concentrating process, near-infrared spectroscopy (NIRS) was used for fast monitoring multiple quality-control indexes of the process simultaneously, which could real-timely reflect its state.Methods: Taking the concentrating process of Carthamus tinctorius L.(CT)alcohol sedimentation solution as an example, samples were composed of two parts, one collected in the process and another prepared by diluting concentrated CT alcohol sedimentation solution.Models of HSYA content and density were established based on PLS algorithm, which were applied to predict the samples in the process of concentration which didn't participate in modeling to test the performance of the models.Results: The correlation coefficients of both calibration models achieved 0.990.And the predicted coefficients were up to 0.980, RSEP for them were less than 10%.Moreover, the fitting curve of the density and alcohol concentration were developed, the correlation coefficients of which reached 0.998.Therefore, the alcohol concentration could be quickly attained by the curve with known density so that the complicated operations of gas chromatography were avoided and analytical time was cut.Conclusion: It is concluded that NIRS can be used as a rapid method to determinate three key quality control indexes (the density, concentration of HSYA and ethanol) at one time in the process of concentrating process of Carthamus tinctorius L.alcohol sedimentation solution.
Preliminary Study on the Particle Sedimentation in the Process of Alcohol Precipitation of Traditional Chinese Medicine

Abstract:Objective To study the process of particle sedimentation of alcohol precipitation process of traditional Chinese medicine and its characteristics,and to preliminarily understand the basic characteristics and key technological parameters of the settling particle in the alcohol precipitation process of different kinds of traditional Chinese medicines,and hold the regulation of sedimentation and providing guidance for technology design and industrial application.Methods Salvia Miltiorrhiza,Radix Sophorae Flavescentis and Fructus Aurantii were used as the experimental materials and UV spectrophotometry was used to determine the effective components of the solution in the process of alcohol precipitation,parameters like the content of settling particulate,the particle size and the sedimentation velocity were also determined.Results The content of the effective components was lower than the original in different levels during the precipitation process.The content of the sedimentation particle increased with the extension of the settling time.The average diameters of particles in each material solution were 36.272 μm,131.820 μm and 0.684 μm and the minimum diameters were 2.000 μm,6.325 μm and 0.142 μm respectively,the average sedimentation velocity were 5.04×10-4m·s-1,1.95×10-4m·s-1 and 1.63×10-7m·s-1.Conclusion There are two ways for the particulate to sediment in the alcohol precipitation process in traditional Chinese medicine,one is free sedimentation,the other is flocculation sedimentation.Process should be designed according to the particle sedimentation process and its characteristics of different traditional Chinese medicine.
