Abstract:Polychlorinated biphenyls (PCBs) are the typical persistent organic pollutants (POPs) in the environment. As a ubiquitous attenuation course of chlorinated organic compounds in anoxic environment, the microorganism-mediated reductive dechlorination process plays an important role in PCBs transformation, especially the transformation of higher chlorinated PCBs. The higher chlorinated PCBs can be dechlorinated in anaerobic condition, and thus, their persistence and toxicity can be decreased. The resultant lower chlorinated PCBs from the dechlorination can be further degraded and completely mineralized in aerobic condition. This paper summarized the research advances of PCBs microbial reductive dechlorination, introduced the mechanisms and characteristics of the dechlorination and the related specific microorganisms, and approached the affecting factors of PCBs bio-dechlorination, as well as the significances of anaerobic dechlorination coupling with aerobic degradation. The future research directions, including the complex metabolic networks of dechlorinating microbial populations, the screening of novel specific dechlorinators and their practical applications in the remediation of PCBs contaminated sites were also prospected.

Abstract:Anaerobic microbial transformation is a key pathway in the natural attenuation of polychlorinated biphenyls (PCBs). Much less is known about the transformation behaviors induced by pure organohalide-respiring bacteria, especially kinetic isotope effects. Therefore, the kinetics, pathways, enantioselectivity, and carbon and chlorine isotope fractionation of PCBs transformation by Dehalococcoides mccartyi CG1 were comprehensively explored. The results indicated that the PCBs were mainly dechlorinated via removing their double-flanked meta -chlorine, with their first-order kinetic constants following the order of PCB132 > PCB174 > PCB85 > PCB183 > PCB138. However, PCBs occurred great loss of stoichiometric mass balance during microbial transformation, suggesting the generation of other non-dehalogenation products and/or stable intermediates. The preferential transformation of (−)-atropisomers and generation of (+)-atropisomers were observed during PCB132 and PCB174 biotransformation with the enantiomeric enrichment factors of −0.8609 ± 0.1077 and −0.4503 ± 0.1334 (first half incubation times)/−0.1888 ± 0.1354 (second half incubation times), respectively, whereas no enantioselectivity occurred during PCB183 biotransformation. More importantly, although there was no carbon and chlorine isotope fractionation occurring for studied substrates, the δ 13 C values of dechlorination products, including PCB47 (−28.15 ± 0.35‰ ∼ −27.77 ± 0.20‰), PCB91 (−36.36 ± 0.09‰ ∼ −34.71 ± 0.49‰), and PCB149 (−28.08 ± 0.26‰ ∼ −26.83 ± 0.10‰), were all significantly different from those of their corresponding substrates (PCB85: −30.81 ± 0.02‰ ∼ −30.22 ± 0.21‰, PCB132: −33.57 ± 0.15‰ ∼ −33.13 ± 0.14‰, and PCB174: −26.30 ± 0.09‰ ∼ −26.01 ± 0.07‰), which further supported the generation of other non-dehalogenation products and/or stable intermediates with enrichment or depletion of 13 C. These findings provide deeper insights into the anaerobic microbial transformation behaviors of PCBs.

Abstract:Polychlorinated biphenyls (PCBs) are a class of synthetic organic compounds that are widely distributed in the environment and have significant adverse effects on humans as well as the ecosystem. Many microorganisms that can degrade PCBs have been reported, including monocultures of bacteria and fungi, as well as natural and artificial microbial consortia. Generally, anaerobic dechlorination can degrade highly chlorinated PCBs to reduce their toxicity, while aerobic oxidation degrades lowly chlorinated PCBs (four or fewer chlorine atoms) and realizes their complete mineralization into CO 2 by disrupting the benzene ring structure. Due to the need for the metabolism of a wide range of PCB congeners in the environment, microbial consortia have become a research hotspot, and their species diversity facilitates metabolic cooperation, enabling them to undertake more complex tasks. In this review, we will discuss the metabolic mechanisms of PCB degradation by monocultures as well as microbial consortia, and focus on the recent progress in the development and optimization of PCB-degrading microbial consortia, the community composition analysis methods, the prevailing optimization strategies such as biostimulation, bioaugmentation, metagenomics, and the combination of other methods for a better remediation effect, which will provide theoretical and practical bases for the rational design, construction and further optimization of efficient microbial consortia for the high-efficiency degradation of PCBs.

Abstract:As one type of the persistent organic pollutants, polychlorinated biphenyls (PCBs) are tremendously harmful to organisms. These compounds are easily absorbed onto soil particles and able to accumulate in soil after they are released into the environment. Bioremediation technology of PCBs-contaminated soils has become a research hotspot in recent years, and immobilized microorganism technique has high developing and applying value because of its unique advantages in environmental remediation. This paper reviewed the chief remediation technology of PCBs-contaminated soils and then analyzed the characteristics of immobilized microorganism technique and its research progress in remediation of organic polluted soil. Finally, the feasibility and problems of this technique in remediation of PCBs-contaminated soil were also discussed.

Abstract:Microbial remediation is preferred as a clean and cost-effective method for restoring environments polluted by organics. But the biodegradation rates of hydrophobic organic contaminants (HOCs) are usually extremely restricted by their low bioavailability, especially in soil. Here, a physical method (mechanical mixing) and a chemical method (randomly methylated-P-cyclodextrins, RAMEB) were adopted to improve the bioavailability and biodegradation of polychlorinated biphenyls (PCBs) of an aged soil. The bioavailability of tri-CBs was increased by adding RAMEB in soil/slurry or assisting mechanical mixing in slurry, but these methods had no effects on the bioavailability of tetra-CBs and high chlorinated PCBs (Cl > 4). The degradation rate of tri-CBs could be obviously enhanced by adding RAMEB in soil or assisting mechanical mixing in slurry. The highest removal amount of tri-CBs reached 43.8% in 100 d with a first-order decay kinetics constant of 0.0059 d(-1). But the removal of tetra-CBs and high chlorinated PCBs (Cl > 4) were not significant in all mesocosms, possibly due to the lack or weakness of the native degrading microflora. Based on the analysis of the richness and diversity of bacterial communities, the characteristics of the heatmap and the variation of bphC copy numbers in the soil/slurry mesocosms, it could be inferred that there was no obvious corresponding relationship between the variation of the bacterial communities and the physical/chemical measures. Unravelling the Processes Involved in Biodegradation of Chlorinated Organic Pollutant: from Microbial Community to Isolated Organohalide Degraders

Abstract:Hundreds of studies have demonstrated the bioremediation of chlorinated organic pollutants (COPs) in flooded environments. However, the role of specific functional strains in degrading COPs under complex media such as wetlands is still unclear. Here, we focused on the microbial characteristics of COP-polluted sediments, identified the bacteria responsible for degradation and conducted a genomic analysis of these bacteria. Four strains were obtained and identified as Petrimonas sulfuriphila PET, Robertmurraya sp. CYTO, Hungatella sp. CloS1 and Enterococcus avium PseS3, respectively. They were capable of degrading a typical COP, γ-hexachlorocyclohexane (γ-HCH). The residual γ-HCH concentrations were 58.8 % (PET), 45.6 % (CYTO), 60.2 % (CloS1), and 69.3 % (PseS3) of its initial value, respectively. Strain PET, CYTO and CloS1 could degrade γ-HCH to its dehalogenation product chlorobenzene. Each strain harbors genes annotated to the pathway of halogenated organic matter degradation (e.g. 2-haloacid dehalogenase) and cobalamin biosynthesis, which are involved in the degradation of COPs. Comparative genomic analysis of the four strains and other classical organohalide-respiring bacteria (e.g. Dehalococcoides mccartyi and Sulfurospirillum multivorans DSM 12446) showed that they share orthologous clusters related to the cobalamin biosynthetic process (GO:0009236). VB12 was also detected in the culture systems of the four strains, further highlighting the importance of cobalamin in COPs degradation. In the genome of the four strains, some genes were annotated to the halogenated organic matter degradation and cobalamin biosynthesis pathway within horizontal gene transfer (HGT) regions. This further indicated that microorganisms carrying these genes can adapt faster to pollution stress through HGT. Together, these findings revealed the co-evolution mechanism of functional strains and may provide novel insights into improved bioremediation strategies for COP-polluted complex media based on generalist organochlorine-degrading bacteria.

Abstract:The polychlorinated technologies of polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzo-furans (PCDFs) were reviewed in the paper, including (1) photodegradation and photocatalytic degradation of standard PCDD/Fs compounds in different matrixes (such as organic solvents, soils and surface of vegetation); (2) photodegradation of PCDD/Fs produced in experiments; (3) photodegradation of PCDD/Fs existed in nature (such as fly ash from municipal waste incineration). Finally, the future research direction on photodegration of PCDD/Fs was proposed.

Abstract:The environmental pollution and health risks caused by the improper disposal of electric and electronic waste (e-waste) have become urgent issues for the developing countries. One of the typical pollutants, polychlorinated biphenyls (PCBs), is commonly found in farmland in Taizhou, a major hotspot of e-waste recycling in China. This study investigated the amount of PCB residue in local farmlands. Biotransformation of PCBs was further studied under different water management conditions in paddy field with or without rice cultivation, with a special focus on the alternating flooded and drying processes. It was found that paddy field improved the attenuation of PCBs, especially for highly chlorinated congeners. In the microcosm experiment, 40% or more of the initial total PCBs was removed after sequential flood-drying treatments, compared to less than 10% in the sterilized control and 20% in the constant-drying system. Variation in the quantity of PCBs degrading and dechlorinating bacterial groups were closely related to the alteration of anaerobic-aerobic conditions. These results suggested that alternating anoxic-oxic environment in paddy field led to the sequential aerobic-anaerobic transformation of PCBs, which provided a favorable environment for natural PCB attenuation.
The roles of organic amendments and plant treatments in soil polychlorinated biphenyl dissipation under oxic and sequential anoxic-oxic conditions

Abstract:Understanding polychlorinated biphenyl (PCB) degradation in sequential anaerobic-aerobic remediation is crucial for effective remediation strategies. In this study, microcosm and greenhouse experiments were conducted to dissect the effects of organic amendments (carbon-based) and plant treatments (ryegrass) on soil PCB dissipation under oxic and sequential anoxic-oxic conditions. We analyzed the soil bacterial community in greenhouse experiments using high-throughput sequencing to explore plant-pollutant-microbe interactions. Microcosm results showed that organic amendments alone did not facilitate aerobic PCB removal, but significantly accelerated PCB dechlorination under anoxic conditions altering the profiles of PCB congeners. In standard greenhouses, plant treatments substantially increased PCB dissipation to 50.8 ± 3.9%, while organic amendments aided phytoremediation by promoting plant growth, increasing PCB removal to 65.9 ± 3.2%. In sequential anaerobic-aerobic greenhouses, plant growth was inhibited by flooding treatment while flooding stress was markedly alleviated by organic amendments. Plant treatments alone during sequential treatments did not lead to PCB dissipation; however, dissipation was significantly promoted following organic amendments, achieving a removal of 41.2 ± 5.7%. This PCB removal was primarily due to anaerobic dechlorination during flooding (27.8 ± 0.5% removal), rather than from plant growth stimulation in subsequent planting phase. Co-occurrence network and functional prediction analyses revealed that organic amendments recruited specific bacterial clusters with distinct functions under different conditions, especially stimulating plant-microbe interactions and xenobiotics biodegradation pathways in planted systems. The findings provide valuable guidance for the design of practical remediation strategies under various remedy scenarios, such as in arable or paddy fields.

Abstract:Bacterial degradation mechanism for high chlorinated pentachlorobiphenyl (PentaCB) with worse biodegradability has not been fully elucidated, which could limit the full remediation of environments afflicted by the complex pollution of polychlorinated biphenyls (PCBs). In this research, a new PentaCB-degrading bacterium Microbacterium paraoxydans that has not been reported was obtained using enzymatic screening method. The characteristics of its intracellular enzymes, proteome and metabolome variation during PentaCB degradation were investigated systematically compared to non-PentaCB conditions. The findings indicate that the degradation rate of PentaCB (1 mg/L) could reach 23.9% within 4 hours and achieve complete degradation within 12 hours, with the mixture of intracellular enzymes being most effective at a pH of 6.0. During the biodegradation of PentaCB, the 12 up-regulated proteins characterized included ABC transporter PentaCB-binding protein, translocase protein TatA, and signal peptidase I (SPase I), indicating the presence of functional proteins for PentaCB degradation in both the cytoplasm and the outer surface of the cytoplasmic membrane. Furthermore, five differentially enriched metabolites were strongly associated with the aforementioned proteins, especially the up-regulated 1, 2, 4-benzenetriol which feeds into multiple degradation pathways of benzoate, chlorocyclohexane, chlorobenzene and aminobenzoate. These relevant results help to understand and speculate the complex mechanisms regarding PentaCB degradation by M. paraoxydans , which have both theoretical and practical implications for PCB bioremediation.

Abstract:2,3',4,4',5-pentachlorodiphenyl (PCB 118), a highly representative PCB congener, has been frequently detected in various environments, garnering much attention across the scientific community. The degradation of highly chlorinated PCBs by aerobic microorganisms is challenging due to their hydrophobicity and persistence. Herein, the biodegradation and adaptation mechanisms of Methylorubrum sp. ZY-1 to PCB 118 were comprehensively investigated using an integrative approach that combined degradation performance, product identification, metabolomic and transcriptomic analyses. The results indicated that the highest degradation efficiency of 0.5 mg L-1 PCB 118 reached 75.66% after seven days of inoculation when the bacteria dosage was 1.0 g L-1 at pH 7.0. A total of eleven products were identified during the degradation process, including low chlorinated PCBs, hydroxylated PCBs, and ring-opening products, suggesting that strain ZY-1 degraded PCB 118 through dechlorination, hydroxylation, and ring-opening pathways. Metabolomic analysis demonstrated that the energy supply and redox metabolism of strain ZY-1 was disturbed with exposure to PCB 118. To counteract this environmental stress, strain ZY-1 adjusted both the fatty acid synthesis and purine metabolism. The analysis of transcriptomics disclosed that multiple intracellular and extracellular oxidoreductases (e.g., monooxygenase, alpha/beta hydrolase and cytochrome P450) participated in the degradation of PCB 118. Besides, active efflux of PCB 118 and its degradation intermediates mediated by multiple transporters (e.g., MFS transporter and ABC transporter ATP-binding protein) might enhance bacterial resistance against these substances. These discoveries provided the inaugural insights into the biotransformation of strain ZY-1 to PCB 118 stress, illustrating its potential in the remediation of contaminated environments.
Influence of Redox Conditions on the Microbial Degradation of Polychlorinated Biphenyls in Different Niches of Rice Paddy Fields

Abstract:The biotransformation of PCBs in three niches with contrasting redox conditions (water–soil interface, rhizosphere and non-rhizosphere subsoil) was studied using rhizoboxes subjected to the sequential flooding and draining conditions associated with rice cultivation in paddy fields. Rice cultivation favored PCB dechlorination and further transformation of the degradation products. Microbial dechlorination activity not only began earlier but was also 50% greater in the rhizosphere in comparison with bulk soil. Distinct profiles of both phospholipid fatty acids (PLFAs) and PCB congeners arose in each niche. Signature PLFAs attributed to Gram-positive (G+) bacteria were significantly correlated with the dissipation of PCB congeners in paddy field soils, especially in the rhizosphere.
Remediation of Polychlorinated Biphenyls (PCBs) through Bio and Phytoremediation in the Environment: A Review Study

Abstract:One of the persistent organic pollutants (POPs) utilized globally between the 1930s and 1980s was polychlorinated biphenyls (PCBs). Even though their usage has been severely constrained, a lot of PCBs are still present in the environment, including soils and air. This local, national, international and governmental organizations regarding polychlorinated biphenyls and how can resolve and treat the problem sustainably. This can be achieved if seriously working on degrading persistent organic pollutants, especially polychlorinated biphenyls biologically via thorough research and sound implementation. The methods opted for remediation by the use of choosing suitable types of microorganisms and plants. These methods are practically active for the identification of the source, concentration level and effects on the ecosystem. It would help in developing appropriate strategy and data generation in formulating the inventories for the management of polychlorinated biphenyls in the future. It is also noted through numerous research that polychlorinated biphenyls are also found in human blood commonly analyzed by indicator congeners such as PCB 138,153, 180, and in air and soil markers of PCBs pollution include PCB congeners 28, 52,101,138,153 and 180 as well. The methods and mechanisms described for degrading the persistent pollutant in this research review the solutions for mitigating PCB pollution in an environment. The best option is to strictly ban it in Pakistan or weightage its usage with its multiple benefits and disadvantages. Promote biological treatment if the payback is healthy and environmentally sustainable.

Abstract:Polychlorinated biphenyls (PCBs) are manmade organic compounds, and pollution due to PCBs has been a global environmental problem because of their persistence, long-range atmospheric transport and bioaccumulation. Many physical, chemical and biological technologies have been utilized to remediate PCBs contaminated soils and sediments, and there are some emerging new technologies and combined methods that may provide cost-effective alternatives to the existing remediation practice. This review provides a general overview on the recent developments in chemical treatment and electrokinetic remediation (EK) technologies related to PCBs remediation. In particular, four technologies including photocatalytic degradation of PCBs combined with soil washing, Fe-based reductive dechlorination, advanced oxidation process, and EK/integrated EK technology (e.g., EK coupled with chemical oxidation, nanotechnology and bioremediation) are reviewed in detail. We focus on the fundamental principles and governing factors of chemical technologies, and EK/integrated EK technologies. Comparative analysis of these technologies including their major advantages and disadvantages is summarized. The existing problems and future prospects of these technologies regarding PCBs remediation are further highlighted.
