Abstract:The first isolation of benzimidazoline ring substituted ketene aminals 7a-c and/or their amidine tautomers 7'b-d, which were synthesized by the reactions: (1) benzoyl substituted ketene mercaptals 3 with N-methyl-o-phenylenediamine 4, and (2) 1,2-dimethylbenzimidazole 5 with ethyl benzoates 6, were disclosed. The tautomeric equilibrium between 7 and 7', along with benzothiazoline and benzoxazoline ring substituted ketene N,S-acetals 8 and 8' and ketene N,O-acetals 9 and 9', were discussed.

Abstract:2-Cyclopenten-1-one-5-carboxylic ester derivatives 14 are synthesized in a four-step-reaction sequence starting from alkynyl aldehydes 9 via 4Z-β-vinyl-α-diazo β-ketoesters intermediate 8. The synthetic method for 8 is described. When the δ substituent is an alkyl group, Rh(II)-mediated decomposition of the diazo compounds 8 led to an intramolecular C–H insertion to afford 2-cyclopenten-1-one-5-carboxylic ester derivatives 14 in high yields. When the δ substituent is an aryl group, 2-hydroxynaphthoate 15 is obtained exclusively. In both cases, no Wolff rearrangement product was observed.

Abstract:The homologation of ketones with diazo compounds is a useful strategy to synthesize one-carbon chain-extended acyclic ketones or ring-expanded cyclic ketones. However, the asymmetric homologation of acyclic ketones with α-diazo esters remains a challenge due to the lower reactivity and complicated selectivity. Herein, we report the enantioselective catalytic homologation of acetophenone and related derivatives with α-alkyl α-diazo esters utilizing a chiral scandium(III) N,N'-dioxide as the Lewis acid catalyst. This reaction supplies a highly chemo-, regio-, and enantioselective pathway for the synthesis of optically active β-keto esters with an all-carbon quaternary center through highly selective alkyl-group migration of the ketones. Moreover, the ring expansion of cyclic ketones was accomplished under slightly modified conditions, affording a series of enantioenriched cyclic β-keto esters. Density functional theory calculations have been carried out to elucidate the reaction pathway and possible working models that can explain the observed regio- and enantioselectivity.

Abstract:The fused heterocyclic compounds 2 : imidazo [1,2-a] pyridine 2a-c and pyrido [1,2-a] pyrimidine 2d were obtained from the reaction of alpha-cinnamoyl ketene dibenzylthio acetals 1 with diamine. When alpha-cinnamoyl -alpha'-benzoyl ketene N, N-acetals 3a-b were treated by t-BuONa/t-BuOH solution. 8-benzoyl-pyrido[1,2-a] pyrimidine 4 was produced.

Abstract:Two epimeric pairs of N‐Ts‐protected cyclohexylamines that incorporate an unsaturated side‐chain at the C2 position have been prepared. Contrary to expectations, none of these underwent clean, photochemically‐induced intramolecular hydroamination reactions to give the corresponding perhydro‐indole or ‐quinolines. However, they did participate in efficient olefin cross metathesis (OCM) reactions with methyl crotonate. Three of the four acrylates so‐formed engaged in base‐promoted and completely diastereoselective, intramolecular aza‐Michael addition (cyclisation) reactions to give the isomeric and C2‐substituted perhydro‐indoles or ‐quinolines with the structures of these being confirmed by single‐crystal X‐ray analyses. DFT calculations generated results consistent with the observed diastereoselectivities and the proposed transition states. Manipulation of the ester groups associated with the cyclization products allowed for their conversion, inter alia, into the corresponding n‐propyl residues and thus providing novel analogues of neurotoxic alkaloids such as pumiliotoxin C.

Abstract:Active methylene compounds 3 react with CS2, 2-bromoethanol in the presence of K2CO3 to give two products 4 and 5 through one-pot reaction. Acylketene cyclic O,S-ethylene acetals 10 have been synthesized by the displacement of 2 with 2-mercaptoethanol using sodium tert-butoxide as base. Configurations of the compounds 4 and 10 have been confirmed by X-ray results and H-1 NMR data. The mechanism has been discussed.

Abstract:N,O-acetals are part of many synthetic intermediates and important skeletons of numerous natural products and pharmaceutical drugs. The most straightforward method of the synthesis of N,O-acetals is the enantioselective addition of O-nucleophiles to imines. However, using this method for the synthesis of linear chiral N,O-ketals still remains challenging due to the instability of raw materials under acidic or basic conditions. Herein, we developed a Cu-catalyzed asymmetric addition of alcohols to β,γ-alkynyl-α-imino esters under mild conditions, providing the corresponding linear chiral N,O-ketals with up to 96% ee. The method tolerates some variation in the β,γ-alkynyl-α-imino ester and alcohol scope, including some glucose and natural amino acid derivatives. Computational results indicate that the Boc group of the substrates assist in the extraction of hydrogen atoms from the alcohols to promote the addition reactions. These products could be synthesized on a gram-scale and can be used in several transformations. This asymmetric addition system provides an efficient, mild, gram-scale, and transition-metal-catalyzed synthesis of linear chiral N,O-ketals.

Abstract:An effective and novel synthesis of β-keto amides had been developed by NaOH promoted hydrolysis of α-oxo ketene-N, S-acetals. It was found that the hydrolysis of α-oxo ketene-N, S-acetals in the presence of 2 equivalent of NaOH in refluxing EtOH (95%) gave β-keto amides in excellent yields. The uses of toxic and harmful amines as well as the prepurification of these nucleophilic reagents were not required in the hydrolysis reaction. The method can extend to gram scale.

Abstract:A FeCl3·6H2O-catalyzed synthesis of β-ketothioesters from the hydrolysis reaction of chain α-oxoketene dithioacetals was carried out. Subsequently, we explored the FeCl3·6H2O-catalyzed Friedel-Crafts alkylation of the synthesized β-ketothioesters with alcohols in CH3CN at 25 °C, affording the desired α-alkylated β-ketothioesters in excellent yields. Wide scope of the substrates, use of inexpensive reagents, high yields under mild reaction conditions are notable features of these reactions.

Abstract:The laboratory-abundant and low-toxic ethyl acetate was explored successfully for the first time as acyl group donor for the Minisci-type acylation of quinolines. In this approach, TBAC/K 2 S 2 O 8 (TBAC: tert-butylacetyl chloride) system played a important role, and the introduced acyl group was derived from the alkoxyl group moiety of ester acetates. Most N -heteroarenes, such as quinoxaline and isoquinoline, etc. were also compatible to this synthetic strategy affording acylated products in high yields. Simultaneously, the C2—H alkylation was realized by accident for 4-quinazolinone. Furthermore, the acylation mechanism was proposed through the chlorine radical abstracting the inactive α -hydrogen of the alkoxyl group.
