Abstract:Although pro-UK is a single-chain (sc) zymogen, it has an intrinsic catalytic activity substantially greater than most other proenzymes. The structural basis for this unusual activity has not been previously reported. A 3-d homology model was calculated from the crystallographic structure of chymotrypsinogen. The model revealed a region (aa297-313), adjacent to the active site, called "flexible loop" because of its very high isotropic temperature factors. Lys300 within this structure was found to be critical to the intrinsic activity of pro-UK, since a Lys300-Ala mutant had a 40-fold lower sc activity. The model indicated that this was related to disruption of the weak salt bridge between Lys300 and Asp355, which transiently (depending on the loop position) induces an active site conformation. By contrast, a Lys300His mutant had only a 3-5-fold lower intrinsic activity, probably because the salt bridge was weakened rather than broken. This mutant had a 2-fold higher tc activity than UK, whereas that of the Lys300-Ala mutant was 3-fold lower. To increase the flexibility of the loop, Asp305-Ala and Tyr306-Gly mutants were made. In both, a 510-ibld reduction in sc and tc-activities (decrease in Kcal) were found. Lys3'3 was another potential surrogate for the neo-terminal a-amino of tc-UK according to studies of t-PA. However, a Lys313-Ala mutation caused no functional changes, showing that there are important structure-function differences between these two activators. Plasmin activation of pro-UK was not significantly affected by any of these mutations despite spatial proximity between the flexible loop and the activation site. In conclusion, the observed effects of these mutations within the flexible loop demonstrate its causal relationship to the unusually high intrinsic activity of pro-UK. The flexible loop also had some influence on tc enzymatic activity, which has not been described in other serine proteases.

Mutagenesis at Pro309 of Single-Chain Urokinase-Type Plasminogen Activator Alters Its Catalytic Properties.

Abstract:The charge of Lys(300(c143)) located within a flexible loop((297-313)) of sc-uPA has been identified as an important determinant for its high intrinsic activity. Mutations affecting the flexibility of the loop also modulate the intrinsic activity. Glu-plasminogen activation by sc-uPA is strongly promoted by fibrin fragment E but not fibrin fragment D-dimer, whereas plasminogen activation by t-PA is strongly promoted by fragment D-dimer but not fragment E. To further investigate the effect of conformation changes in the flexible loop on catalytic properties of sc-uPA, cassette mutations at Pro(309(c152)) were made and characterized. It was found that the activation of Pro(309(c152)) mutants by Lys-plasmin was only moderately affected. In contrast, the intrinsic and two-chain activities of Pro(309(c152)) mutants against S2444 were both significantly decreased. The two-chain activities of these mutants against Glu-plasminogen were also reduced in a range of 1.1- to 127-fold. The mutations of Pro(309(c152)) to Trp/Phe and Arg/Asp more significantly affected both intrinsic and two-chain activities, while only a moderate decrease in activity was found with mutations to Ala/Ser/Thr. In contrast to wild-type sc-uPA, plasminogen activation by Pro(309(c152)) mutants was found to be promoted by both fibrin fragment E and D-dimer. In the presence of 2.0 mu M D-dimer, plasminogen activation by mutant Pro(309(c152)) -> His was promoted by 22-fold, while only 2.0-fold promotion was found with mutant Pro(309(c152)) -> Gly. In conclusion, these findings demonstrated that conformation changes in the flexible loop of sc-uPA not only affect its intrinsic and two-chain activity, but also extend its promotion of plasminogen activation by fragment E to D-dimer.

Abstract:Single-chain urokinase-type plasminogen activator or pro-urokinase is a zymogen with an intrinsic catalytic activity which is greater than that of most other zymogens. To study the structural basis for this activity, a three-dimensional homology model was calculated using the crystallographic structure of chymotrypsinogen, and the structure-function relationship was studied using site-directed mutagenesis and kinetic analysis. This model revealed a unique Lys(300) in pro-urokinase which could form a weak interaction with Asp(355), adjacent to the active site Ser(356). It was postulated that this lysine, by its epsilon-amino group, may serve to pull Ser(356) close to the active position, thereby inducing the higher intrinsic activity of pro-urokinase. This was consistent with the published finding that a homologous lysine (Lys(416)) in single chain tissue plasminogen activator when mutated to serine induced some reduction in activity, To test this hypothesis, a site-directed mutant with a neutral residue (Lys(300) --> Ala) was produced and characterized. The Ala(300)-pro-urokinase had a 40-fold lower amidolytic activity than that of pro-urokinase. It was also stable in plasma at much higher concentrations than pro-urokinase, reflecting much attenuated plasminogen activation, Plasmin activatability was comparable to that of pro-urokinase. but the resultant two-chain derivative (Ala(300)-urokinase) had a lower enzymatic activity (approximate to 33% that of urokinase) due to a reduction of k(cat). Interestingly, the K-M Of two-chain Ala(300)-urokinase against plasminogen was 5.8-fold lower than that of urokinase, being similar to that of pro-urokinase which has a K-M about 5-fold lower than urokinase. In conclusion, the hypothesis that Lys(300) is a key structural determinant of the high intrinsic activity of pro-urokinase was confirmed by these studies. This residue also appears to be important for the full expression of the enzymatic activity of urokinase.

Abstract:The Kringle-1 structure of plasminogen (PGK-1), the Kringle-2 structure of tissue plasminogen activator (PAK-2) and the Kringle structure of prourokinase (UKK) has been modeled on the basis of the three-dimensional structure of Kringle-1 of prothrombin (PTK-1) at 2.8 A resolution. The predicted three-dimensional structure of these Kringles shows that the binding site of PGK-1 is characterized by an apparent dipolar site, the polar parts of which are separated by a hydrophobic region. PAK-2 possesses the anionic center but has not a cationic binding center which might be provided by a guanidinium group from Arg-69 located adjacent to the Arg-71 position. UKK possesses neither the anionic binding center nor the cationic center which are probably the main reason for the poor fibrin specificity of urokinase.

Abstract:In view of the similarity of the charge distribution between fibrin A alpha 148-161 and A chain 149-157 of urokinase, the latter might compete with fibrin A alpha 148-161 when single chain pro-urokinase is converted to double chain urokinase. To test this, the stretch of urokinase A chain 135-157 was separated from the low molecular weight urokinase, a competitive binding between this stretch and fibrin to tPA kringle-2 was shown by radio-binding assay. The inhibition of the stretch on the fibrin stimulated activation of plasminogen was demonstrated in the caseinolytic system. The synthesized novapeptide urokinase A chain 149-157 (R-peptide) showed a significant inhibition on the activation of plasminogen in the presence of fibrin. By contrasting finely with R-peptide, a synthesized novapeptide in which Arg154 and Arg156 were replaced by Asp (D-peptide) did not show any inhibition effect on the fibrin stimulated activation of plasminogen by tPA. These results suggest that the positively charged residues in the stretch 149-157 of urokinase are crucial for the inhibition of fibrin binding with the kringle domain of urokinase.
