Dynamic observations of glycosaminoglycan synthesis of primary and passaged rabbit articular chondrocytes

Abstract:BACKGROUND: Chondrocytes always dedifferentiate during in vitro culture. The dedifferentiation of passages is important to determine the chondrocyte’s in vitro culture time, and is one of the key factors to affect the cartilage tissue engineered result. OBJECTIVE: To study the capacity of glycosaminoglycan (GAG) synthesis of cultured rabbit articular chondrocytes at different incubation densities. DESIGN, TIME AND SETTING: A single-sample in vitro experiment of cells was performed at the Laboratory of Orthopeadics of the Second Hospital of Shanxi Medical University between January 2007 and May 2007. MATERIALS: Five New Zealand white rabbit aged 1 month, irrespective of genders. METHODS: Chondrocytes were isolated from the knee joints of five rabbits using 0.4% Pronase enzyme and 0.25 g/L type Ⅱ collagenase digestion under sterile conditions, and then cultured in vitro. The serum-free culture solution was renewed when the cells were confluent. The supernatants were sampled at 12, 24, 36, 48, 60 hours after the culture medium renewal to measure GAG concentration. The passages of 0, 1, 2 cells underwent the same protocols. MAIN OUTCOME MEASURES: The morphology of the cells under inverted microscope. The grow speed of the chondrocyte. The GAG concentrations of supernatants of P0, P1 and P2 cells. RESULTS: The morphology of chondrocytes was not changed within passage 2, but more vacuole-shaped particles were seen in P2 cells. After the passage, the time of cell confluence was shortened, but the GAG concentrations of the supernatants reduced with the increase of passage (P < 0.001). There were significant differences for the two-two comparison between P0, P1 and P2 cells (P < 0.001). 60 hours after the medium renewal, the GAG concentration in P1 chondrocytes was reduced by 24%, and P2 cells by 74% compared with P0 cells. The GAG concentration was shown to increase with time (P < 0.001). It decreased with the increase of passage (P < 0.001). The significant difference of GAG concentration became greater between the primary cells and passage cells (P < 0.001). CONCLUSION: The GAG synthesis of primary chondrocytes is the strongest and that of in vitro cultured chondrocytes decreases with the passages. It is an effective method to continuously detect the GAG concentration of the supernatant following cell confluence.
