Abstract:With the enlargement of the cultivation scale, the kinds of turtle's disease become more and more, which results in lots of economic losses. A large number of researches were conducted in last few years.The present paper reviews the study of the cause, diagnosis and cure of the diseases. Some issues concerning the study are pointed out, and some relative suggestions are presented.
Effects of Cysteamine Hydrochloride on Growth Performance and Nonspecific Immunity of Chinese Soft-shelled Turtle

Abstract:The experiment was conducted to determine the effects of cysteamine hydrochloride on growth performance and nonspecific immunity of Chinese soft-shelled turtle.Seven hundreds and fifty healthy Chinese soft-shelled turtles (220±10 g of average weight) were randomly divided into five groups,each group included three replicates of 50 soft-shelled turtles.The turtles were given a special diet supplemented with different effective dosages of cysteamine hydrochloride. The control group was fed basal diet,groupⅠ was supplemented with 6 000 mg/kg CSH (cysteamine hydrochloride) which was fed once every 6 days,another three treatments were supplemented consecutively with 500 mg/kg CSH for group Ⅱ,1 000 mg/kg CSH for group Ⅲ,1 500 mg/kg CSH for group Ⅳ.The experiment lasted for 90 days.Chinese soft-shelled turtles fed with 1 000 mg/kg CSH had significantly increased weight gain rate and feed efficiency.Diets supplemented with CSH also improved the level of serum somatostatin and gastrin in Chinese soft-shelled turtles.The ratio of RNA/DNA increased when 1 000 mg/kg CSH were supplemented (P0.05),and the CSH had improved the nonspecific immunity of Chinese soft-shelled turtles.The diet of CSH supplementation can improve the growth and the level of nonspecific immunity.
Studies on the Pathogens of Hole Disease in Soft-Shelled Turtle (trionyx Sinensis)

Abstract:Strains of N 9910 and N 9911 were isolated from the naturally diseased soft shelled turtles. Experimental infection of the two strains caused the death of white mouses and brought on the same clinical symptoms in healthy soft shelled turtles. Two strains of N 9910 and N 9911 were identified as belonging to Morganella morganii on the basis of their morphological features and physio biochemical characteristics. Drug sensitivity tests showed that the two strains were highly sensitive to Chloramphenicol, Ceftriaxone, Norfloxacin and Amikacin.

Abstract:The present paper describes the characters of pathogenic bacteria of common disease in soft-shelled turtles from Zhejiang province.Of 20 bacteria isolated from the heart,liver and ascites of the turtles with caves disease,ulcerative skin diseased and hemolytic ascitic disease,5 were identified as Aeromonas sobria,3 A.caviae,2 A.veronii,2 Pseudomonas alcaligene,2 Proteus mirabilis,3 Achro sp,2 Acinetobacter sp,and 1 Flavobacterium sp.Aeromonas sobria and A.caviae were proved to be the pathogens of those common disease by infection experiment.The results of drug sensitivity test and therapeutic effect showed neomycin,ceftriaxon,amikacin,gentamycin and ciprofloxacin should be the chief drugs for treatment of these disease.

Abstract:To improve the light environment and welfare of the turtle cultured indoors, the effects of lighting mode on growth performance, cortisol level, and oxidative stress of juvenile Chinese three-keeled pond turtle, Chinemys reevesii, were investigated in this study. The experimental turtles with an initial weight of 5.61 +/- 0.09 g were reared in tanks under four different lighting modes: three groups with light (lighting the basking area and water area, LBW; lighting the water area only, LW; lighting the basking area only, LB) and control group (no light, NL). The experiment was conducted for more than six months, with each group having three replicates. After 203 d of the experiment, the turtle in the LW group exhibited higher weight gain rate (WGR) and a specific growth rate (SGR, %/d) compared to other treatments. Also, results showed that the final body weight of the turtle exposed to LW was higher than that exposed to other treatments. On the physiological level, serum cortisol level in turtles exposed to LW was significantly lower than that in other treatments. Regarding oxidative stress, the level of catalase (CAT) in turtles exposed to LW and LB was significantly lower than that exposed to LBW and NL. The malonaldehyde (MDA) activity in turtles exposed to LW was significantly lower than other treatments. Based on the growth performance and health status, it is suggested that lighting the water area only is the optimal lighting mode for the juvenile three-keeled pond turtle cultured indoors.

Abstract:The Chinese turtle trade is the primary threat to endangered turtle populations throughout Asia, primarily because of the long tradition of consuming turtles in China. Practitioners of Traditional Chinese Medicine (TCM) promote nutritional and medicinal benefits from eating turtles, especially those made from hardshell species. We tested these claims by determining the nutritional value of turtle products (meat, fat and shell) in five species of geoemydid turtle, Cuora trifasciata, C. mouhotii, Mauremys mutica, M. sinensis and Geoemyda spengleri. Nutritional variables such as the composition of amino acids, fatty acids and mineral elements were analyzed to determine the relative nutritional quality of turtle products. Our study refutes TCM claims about products made from hardshell turtles. Alternative animal products should be substituted to obtain similar minerals, amino acids and fatty acids. Balancing the cultural use of turtles with their conservation status remains a major challenge.
Pathology and drug susceptibility study of an outbreak of bacterial pathogen infecting the Chinese soft-shelled turtle (Pelodiscus sinensis)

Abstract:The Chinese soft-shelled turtle (Pelodiscus sinensis) is a freshwater softshell turtle native to East Asia. It is a choice of food and tonic in China, Taiwan and other Asian countries and is commonly cultured in ponds, lakes and canals. This study aimed to study the aetiology of a deadly disease outbreak among these turtles in Jiangxi Province of China in 2020 and 2021. By performing an aseptic dissection of symptomatic and moribund P. sinensis, a dominant strain (No.: ZF2) was isolated from livers, and studies were carried out to ascertain the biological status, pathogenicity and pathological characteristics of the isolate. For isolate identification, a morphological observation, physiological and biochemical analysis, 16S rRNA and gyrB gene sequencing, and phylogenetic analysis were performed. Other experiments were also carried out simultaneously, such as infection experiments, histopathological observation, virulence gene detection and drug sensitivity. Through traditional and molecular biological methods study with the pathogenic strain, ZF2 was identified as Aeromonas veronii. The results of the virulence gene test showed that ZF2 carried three virulence genes, aer, ast and act. Histopathological observation revealed that various internal organs were severely damaged after infection and manifested as liver ischemia and whitening, intestinal chorion shedding, glomerular congestion and splenic lymphocyte proliferation. After the drug sensitivity test results, the ZF2 strain was identified to be sensitive to nine drugs, including cefradine, neomycin, minocycline and enrofloxacin. It was intermediate or resistant to other drugs.

Abstract:The Asian turtle crisis has caused a severe depletion of all native Chinese turtle species. During this time there have been many efforts, both within China and abroad, to slow and/or stop the trade of turtles. However, recent studies on the turtle trade in China show it is not slowing down and is becoming more sophisticated. The history of turtles and turtle trade on Hainan Island, China, has been well documented and therefore allows for comparisons with previous work. We conducted our study in Qiongzhong County and a portion of Danzhou County in central Hainan. We performed both village surveys and field surveys to determine which species are still present in what areas, in what abundance relative to other species, and what is the main use of turtles sold, and their prices. We surveyed 14 villages comprising five regions and collected nine turtles of seven species (five native, two introduced) in roughly 7,240 trap days. For most species, multiple age classes are being collected and there are certain areas where they can be found consistently. Prices have continued to increase for all species and they are now mainly sold for pets or for breeding by farmers. Lack of enforcement by the agriculture and forestry agencies enables the local villagers to freely collect and sell without sufficient fear of punishment.

Abstract:Chinese soft-shelled turtle is a traditional food in China.In this paper,present research status at home and abroad about the nutrition,flavor and functional properties of Chinese soft-shelled turtle were introduced.Chinese soft-shelled turtle had unique flavour,which was a high-protein low-fat food.It contained various functional factors,such as polysaccharide,collagen,taurine and Vitamin B-17.Additionally,Chinese soft-shelled could prevent and cure a lot of dieases including cancer and heart and blood vessel disease,also it could boost up the immunity function of body and resist caducity.

Abstract:Sea turtles are an important umbrella species in marine ecosystems. The populations of all five species of sea turtles in China have dropped sharply due to massive illegal trade and habitat loss. The fast-growing demand for sea turtle displays from Chinese aquariums and private individuals has led to a large-scale illegal trade domestically and internationally. Captive sea turtles are also frequently kept in harsh environments with severe injuries and high mortality rates. Sea turtles have only recently been upgraded from level II to level I on the "List of Wildlife under Special State Protection", this protection level has therefore not matched the real status of sea turtles over the past three decades. The additional collusion between the government and business corporations encourages illegal trade. We argue that the commercial use of sea turtles must be completely prohibited to guarantee their future survival in Chinese waters.

Improved immune function of Chinese soft-shelled turtles (<i>Pelodiscus sinensis</i>) through oral probiotics via the TLR signaling pathway

Abstract:In recent years, large-scale farming of Chinese soft-shelled turtles (Pelodiscus sinensis) has been developing rapidly. However, bacterial infections and environmental stresses limit its healthy development and probiotics have the potential to improve turtles' immunity and reduce disease occurrences. The objective of this study was to evaluate the effects of Bacillus subtilis B10 on intestinal health and immune function of Chinese soft-shelled turtles and explore its mechanism. The results showed that the dietary administration of B. subtilis B10 remarkably increased the levels of urea nitrogen and IL-1 beta, the activities of alkaline phosphatase, lactic dehydrogenase and antioxidant enzymes (including superoxide dismutase and catalase) in serum of Chinese softshelled turtles. Furthermore, B. subtilis B10 up-regulated the expression of genes related to intestinal tight junction protein and TLR8, which improved the intestinal health status and immune function of Chinese softshelled turtle. Finally, RNA-Seq results showed that TLR5 expression was significantly increased in liver. It could be concluded that probiotics B. subtilis B10 may improve the immune function of Chinese soft-shelled turtles via TLR5 in the liver.

Abstract:The immune response of soft-shelled turles(Trionyx sinensis) to oral controlled release microencapsulated vaccine of Aeromonas sobria was described in this study.The formalin-inactivated cultures of Aeromonas sobria Z-1 strain was encapsulated in the micro-particles prepared from the biodegradable materials and the encapsulated vaccine was orally used for immunizing soft-shelled turtles.The agglutinating antibody titers in the serum,bactericidal percent of the blood leucocytes and relative percent survival post challenged with virulent live bacteria of control and immunized mice were detected.After being orally immunized with the microencapsulated vaccine,the agglutinating antibody titers in the serum and bactericidal percent of the blood leucocytes of soft-shelled turtles reached a level equivalent to that of injection with formalin-inactivated cultures and was significantly higher than that of control group.The relative survival percentage of mice immunized with above-mentioned immunization method was 94.7% and 89.5% respectively,while 95% of the control mice had died.The biodegradable microparticle can be used as a candidate vector system of oral vaccine of Aeromonas.
Advancing sea turtle conservation in the South China Sea via U.S.‐China diplomacy

Abstract:The South China Sea is an important oceanic habitat for endangered sea turtles, which are facing anthropogenic pressures from the illegal wildlife trade, fisheries by‐catch, debris pollution, habitat loss, and climate change. Compounding these threats are international disputes over territorial claims in the South China Sea, exerting a general disinclination towards regional collaborations to solve environmental issues. Since sea turtles are migratory animals that move through aquatic and terrestrial habitats, feeding in waters and nesting on beaches under the jurisdiction of multiple countries, the species is an ideal ambassador to raise public support for international collaborations. Transnational communications to coordinate conservation efforts across borders provide a positive agenda of cooperation to build trust, creating a politically neutral platform to facilitate opportunities for diplomacy to reset and improve foreign relations, underscoring the value of sea turtles as a flagship species to reduce international tensions and bring nations together. For this reason, the U.S.‐China EcoPartnership between Sea Turtles 911 and Hainan Normal University was formed to empower local communities to protect the ocean through sea turtle rescue, public education, and scientific research projects. Genetic research determined the geographic origins of illegally traded sea turtles, validating the uniqueness of the Paracel (Xisha) Islands rookery, and urging policymakers to establish a large‐scale sea turtle nature reserve in the South China Sea. To define boundaries of the proposed marine protected area, sea turtles were tracked with satellite telemetry delineating their migratory routes and habitats across multiple countries, underscoring the importance of international collaborations. Debris pollution survey at a major turtle nesting beach in the Paracel Islands demonstrated that pollution is negatively impacting newborn turtle hatchlings and that regional cooperation in the South China Sea should promote the reduction and cleanup of marine debris. Laparoscopic surgery was performed on sea turtles for the first time in China to assess the gender ratio of the local population, which revealed a female biased ratio, indicative of the effects of climate change. A sea turtle rescue center was jointly established in which more than one hundred sea turtles were successfully rescued and released by the international and local community, including students who won national awards for their volunteer service. Implementing grassroots initiatives for sea turtle conservation have become an ideal diplomatic channel to strengthen U.S.‐China relations as several diplomats, including U.S. Ambassador Max Baucus, have engaged in people‐to‐people exchanges during public educational events. This international partnership serves as a case example in which current political disputes can be set aside to prepare future generations for the existential threat of biodiversity loss and climate change, while increasing peace, stability, and turtles in the region.This article is protected by copyright. Abstract:Five species of sea turtles, including leatherback (Dermochelys coriacea), loggerhead (Caretta caretta), green (Chelonia mydas), hawksbill (Eretmochelys imbricata), and olive ridley turtles (Lepidochelys olivacea), were found in the waters east and south of mainland China and the associated islands. Decades of harvesting and habitat degradation have led to a drastic decline in the sea turtle population in the last century. Many foraging grounds and nesting sites have been degraded or have disappeared as a result of human activities. Nowadays, there are only a few nesting sites and even fewer foraging grounds for sea turtles in China. Fishery bycatch is also believed to have been a major cause of sea turtle mortality in recent years. There is an urgent need for more efforts to be focused on the conservation of sea turtles in the region. However, limited information is available, which has impeded the formulation of effective conservation measures. The goal of this paper is to provide an overview on the current population and conservation status of sea turtles, as well as recommendations for their effective management in China.
Transcriptomic and Proteomic Analyses of the Immune Mechanism in Pathogenetic and Resistant Chinese Soft-Shelled Turtle (Pelodiscus sinensis) Infected with Aeromonas hydrophila

Abstract:Background: As intensive aquaculture practices have progressed, the prevalence of bacterial diseases in the Chinese soft-shell turtle (Pelodiscus sinensis) has escalated, particularly infections caused by Aeromonas hydrophila, such as ulcerative dermatitis and abscess disease. Despite this, little is known about their immune defenses against this pathogen. Methods: Our study pioneers an integrated analysis of transcriptomics and proteomics to investigate the immune responses of Chinese soft-shelled turtles to A. hydrophila infection. Results: The investigation revealed significant differences in immune-related pathways between groups susceptible and resistant to A. hydrophila infection after 4 days. A total of 4667 and 3417 differentially expressed genes (DEGs), 763 and 568 differentially expressed proteins (DEPs), and 13 and 5 correlated differentially expressed genes and proteins (cor-DEGs-DEPs) were identified in susceptible and resistant Chinese soft-shelled turtles, respectively. In the resistant group, upregulation of immune-related genes, such as CD3ε and CD45, enhanced T-cell activation and the immune response. The proteomic analysis indicated that immune proteins, such as NF-κB1, were significantly upregulated in the resistant group. The correlation analysis between transcriptomics and proteomics demonstrated that the CD40 gene and protein, differentially expressed in the resistant group compared to the control group, were commonly upregulated within the Toll-like receptor signaling pathway. Conclusions: The transcriptomic and proteomic data obtained from this study provide a scientific foundation for understanding the immune mechanisms that enable the Chinese soft-shelled turtle to resist A. hydrophila infection.

Abstract:The soft-shelled turtles are often exposed to high ambient ammonia levels in intensive rearing conditions.The effects of chronic ammonia exposure on growth and nonspecific immune parameters in juvenile Chinese soft-shelled turtle,Pelodiscus sinensis are evaluated.64 healthy turtles with identical body weight(Body weight:(90.5±20.5) g) are submitted to three ammonia concentrations,1.5(C1),2.6(C2) and 4.1(C3) mg·L~(-1) unionized ammoniaN(UIA-N) for 42 days.A control group(C0) is also set in which the animals are reared in tap water with no extrinsic NH_(4)Cl added.Consistent ambient temperature((29.5±0.5)()℃) and pH(7.75±0.10) in each treatment are maintained throughout the experiment.Over the ammonia range set in the present study,no significant differences of specific growth rate(SGR),serum hemolytic activities,and serum bactericidal activity are observed.But serum bacteriolytic activity and whole blood phagocytic rate are significantly enhanced in group C1 and C2 respectively.The present results indicate that the turtle has adapted to ambient ammonia set in the present study and is fairly resistant to high ambient ammonia levels.
A Comprehensive Review of Disease-Causing Agents in Freshwater Turtles: Implications for Conservation and Public Health

Abstract:Freshwater turtles comprise 81% of all chelonian species despite freshwater systems only occupying 1% of the earth's surface, and they are commonly exploited as pets and food resources. This contact between humans and turtles may put both sides at risk of disease transmission. Additionally, human impact on ecosystems can cause disease outbreaks in turtle populations. In this review, we focused on disease agents affecting freshwater turtles, intending to contribute to conservation and public health efforts. We analysed 423 articles and noted a post-SARS-COVID-19 peak, with most research originating from Asia, North America, and Europe. Emydidae was the most frequently studied family, and there was also a bias towards adults, live specimens, and native species. Since most of the studied turtles were wild-caught, we recommend that captive turtles should also be thoroughly studied since they can transmit diseases to other turtles and humans. We registered 2104 potential disease-causing agents, with Platyhelminthes dominating within Animalia, while Proteobacteria dominated bacterial agents. Viruses' representation was low, highlighting gaps in reptile virology. Fungi, Chromista, and Protozoa were also underrepresented, but this is changing with the development of molecular tools. This synthesis serves as a foundation for targeted health assessments, conservation strategies, and future research, essential to mitigate ecosystem and public health threats.

Abstract:China has one of the world's most diverse freshwater turtle faunas but is also a major consumer of freshwater turtles. In the 1980s over-collecting, illegal trade and habitat destruction increased dramatically, pushing almost all Chinese turtles towards extinction. Despite the critical status of Chinese turtles little has been done to quantify and evaluate the impact of illegal activities on wild populations. We used a combination of market surveys and questionnaires to examine the illegal trade and conservation status of native freshwater turtles in Nanmao, Hainan Province, from February to August 2003. We found a total of 245 collected freshwater turtles comprising eight species. These data indicate that Nanmao has a rich freshwater turtle fauna that is being affected heavily by illegal poaching. No effective measures have been taken to protect the few remaining wild turtle populations. The main factors that lead to illegal trade include the cultural belief that turtles are a viable food/medicine product, lack of conservation awareness, poaching of turtles as a supplement to income, and poor law enforcement. Possible conservation strategies are discussed.
