Abstract:A combination of immunomagnetic separation (IMS) and polymerase chain reaction (PCR) was used to detect Alicyclobacillus acidoterrestris (A. acidoterrestris) in apple juice. The optimum technological parameters of the IMS system were investigated. The results indicated that the immunocapture reactions could be finished in 60 min and the quantity of IMPs used for IMS was 2.5 mg/mL. Then the combined IMS-PCR procedure was assessed by detecting A. acidoterrestris in apple juice samples. The agarose gel electrophoresis results of 20 different strains showed that the IMS-PCR procedure presented high specificity to the A. acidoterrestris. The sensitivity of the IMS-PCR was 2x10(1) CFU/mL and the total detection time was 3 to 4 h. Of the 78 naturally contaminated apple juice samples examined, the sensitivity, specificity and accuracy of IMS-PCR compared with the standardized pour plate method were 90.9%, 97.0% and 96.2%, respectively. The results exhibited that the developed IMS-PCR method will be a valuable tool for detecting A. acidoterrestris and improving food quality in juice samples.
Optimization of DNA Extraction by Microwave Treatment and PCR Detection of Spoilage Alicyclobacillus acidoterrestris in Apple Juice

Abstract:A simple and rapid method for microwave-assisted DNA extraction from A.acidoterrestris in juice sample was developed in this study.A pair of primers was designed based on the gene sequence encoding squalene epoxidase from Alicyclobacillus acidoterrestris published in the Genbank database to establish a PCR assay for detecting Alicyclobacillus acidoterrestris in apple juice.The specificity and sensitivity of the method was investigated by DNA amplification from 11 Alicyclobacillus spp.and 7 other bacteria using the designed specific primers.The detection limit was determined using serially diluted extracts.The results of gel electrophoresis indicated that the DNA bands of microwave-assisted samples were the same as those obtained from ZR Genomic DNA Kit,implying effective bacterial cell lysis in juice samples.The limit of detection of this method was 200 CFU/mL.Only 2 hours were required to detect A.acidoterrestris in juice sample by this method.In conclusion,the microwave-assisted DNA extraction coupled to PCR detection method provides an effective and stable approach for the online detection of Alicyclobacillus spp.from apple juice.

Abstract:Alicyclobacillus acidoterrestris is the most important spoilage species within the Alicyclobacillus genus and has become a major issue in the pasteurized fruit juice industry. The aim of this study was to develop a method combining immunomagnetic separation (IMS) with real-time PCR system (IMS-PCR) for rapid and specific detection of A. acidoterrestris in fruit products. A real-time PCR with the TaqMan system was designed to target the 16S rDNA genes with specific primer and probe set. The specificity of the assay was confirmed using 9 A. acidoterrestris strains and 21 non-A. acidoterrestris strains. The results indicated that no combination of the designed primers and probe was found in any Alicyclobacillus genus except A. acidoterrestris. The detection limit of the established IMS-PCR was less than 10CFU/mL and the testing process was accomplished in 2–3h. For the three types of samples (sterile water, apple juice and kiwi juice), the correlation coefficient of standard curves was greater than 0.991, and the calculated PCR efficiencies were from 108% to 109%. As compared with the standard culture method performed concurrently on the same set of samples, the sensitivity, specificity and accuracy of IMS-PCR for 196 naturally contaminated fruit products were 90.0%, 98.3% and 97.5%, respectively. The results exhibited that the proposed IMS-PCR method was effective for the rapid detection of A. acidoterrestris in fruit products.
Establishment of quantitative PCR assays for the rapid detection of Alicyclobacillus spp. that can produce guaiacol in apple juice

Abstract:Alicyclobacillus species are one of the most significant qualities and safety factors in fruit juice and beverages. The growth of some Alicyclobacillus genus can lead to sour spoilage with the off-odor of medicinal, phenolic or antiseptic, which is mainly caused by the metabolites of guaiacol, dihalophenol and dibromophenol. Especially, guaiacol is regarded as the predominant taint in Alicyclobacillus-spoiled products. In this study, quantitative PCR (qPCR) assays were proposed for the detection of A. acidoterrestris, A. acidiphilus, A. cycloheptanicus and A. herbarius that can produce guaiacol in fruit juice. The 16S rDNA sequences of these four kinds of Alicyclobacillus species were identified and the primers suitable for the qPCR assay were obtained. The sensitivity and specificity of the established methods were evaluated. The results indicated that the developed qPCR approaches were distinctive enough to detect A. acidoterrestris, A. acidiphilus, A. cycloheptanicus and A. herbarius with the sensitivity of 2.6 × 102 CFU/mL, 74 CFU/mL, 2.8 × 102 CFU/mL and 3.1 × 102 CFU/mL, respectively. The correlation coefficients of standard curves were from 0.9807 to 0.9985. Based on the pretreatment of filtration-culture, these bacteria with the initial concentration of 10−1 CFU/mL, 100 CFU/mL and 101 CFU/mL can be effectively detected in 2–20 h, which depended on the target bacteria and their initial concentration. The results displayed that the proposed procedures were effective for the rapid detection of Alicyclobacillus species that can produce guaiacol in apple juice.

Abstract:Apple juice beverage spoilage spawned by Alicyclobacillus acidoterrestris can cause sensory defect by production of medicinal off-flavor, which could hardly be noticed in the early stage and would lead to the consumer complaints and, consequently, the economic and image loss of the manufacturers. Thus, it is necessary to detect the contamination at early stage in case of serious converse effect. The aim of this study was to evaluate the performance of an electronic nose (e-nose) in tandem with chemometric analysis as online, rapid screening technique for detection of the spoilage in apple juice beverage, using test panel evaluation as reference, as well as to predict the spoilage status of apple juice beverage based on the response to the specific volatiles in it. Coupled to linear discriminant analysis (LDA), e-nose could discern the contaminated beverage after 4 h, corresponding to the bacterial number of 200 cfu/mL, the level at which the test panel could not yet diagnose the spoilage, indicating that the signals of e-nose could be applied as early indicators for the onset of spoilage. Sensors 2, 6, 7, and 8 were determined to be quite sensitive to the alteration of flavor in apple juice beverage and had played the most important part in discrimination. Furthermore, based on the response of e-nose, A. acidoterrestris counts in unknown samples could be well predicted by established partial least squares (PLS) regression model with a high relation of 0.95. Based on these results, e-nose can be employed as a powerful tool for quick and reliable analysis of the flavor in apple juice beverages during processing or on the shelf to implement rapid routine inspection as well as realize the early detection of potential spoilage caused by A. acidoterrestris.

Abstract:An approach based on immunomagnetic separation (IMS) and SYBR Green I real-time PCR (real-time PCR) with species-specific primers and melting curve analysis was proposed as a rapid and effective method for detecting Alicyclobacillus spp. in fruit juices. Specific primers targeting the 16S rDNA sequences of Alicyclobacillus spp. were designed and then confirmed by the amplification of DNA extracted from standard strains and isolates. Spiked samples containing known amounts of target bacteria were used to obtain standard curves; the correlation coefficient was greater than 0.986 and the real-time PCR amplification efficiencies were 98.9%- 101.8%. The detection limit of the testing system was 2.8×101 CFU/mL. The coefficient of variation for intra-assay and inter-assay variability were all within the acceptable limit of 5%. Besides, the performance of the IMS-real-time PCR assay was further investigated by detecting naturally contaminated kiwi fruit juice; the sensitivity, specificity and accuracy were 91.7%, 95.9% and 95.3%, respectively. The established IMS-real-time PCR procedure provides a new method for identification and quantitative detection of Alicyclobacillus spp. in fruit juice.

Abstract:Alicyclobacillus acidoterrestris is the main hazardous factor leading to deterioration of commercialized fruit juices, as it can result in serious economic loss to the industry. Currently detection methods require high-end equipment with low specificity and sensitivity. To address these problems, we describe herein a monoclonal antibody with high specificity to A. acidoterrestris using a cell wall protein as antigen. A colorimetric and fluorescence dual-mode immunoassay system employing the prepared antibody was then developed based on the quenching effect of oxidized p-phenylenediamine on fluorescein. This approach could accurately differentiate between the target of interest and other bacteria species with enhanced limit of detection of 4.8 x 10(2) CFU/mL. Apple juice artificially contaminated with A. acidoterrestris at the concentration of 1 CFU/mL could be determined as positive after 24 h enrichment. The resulting immunoassay platform provided a rapid, convenient, sensitive, and selective way for detecting A. acidoterrestris, holding great potential as a robust tool for preventing spoilage in the fruit juice industry.

Abstract:Alicyclobacillus spp. has recently received much attention due to its implication in the spoilage of pasteurized fruit juices, which is characterized by the formation of guaiacol. Previous researches indicate that not all Alicyclobacillus spp. are able to produce guaiacol. The aim of this study was to identify possible differences in the vanillic acid decarboxylase gene involved in guaiacol biosynthesis and then develop specific detection methods for guaiacol producing Alicyclobacillus. Agarose gel electrophoresis results showed that the partial vdcC gene was present in all the guaiacol producing Alicyclobacillus, but absent in non-guaicaol producing strains apart from A. fastidiosus DSM 17978. On the basis of the vdcC gene sequence, a primer pair specific to A. acidoterrestris was designed; then a SYBR Green I real time PCR was established for the direct quantification of A. acidoterrestris in apple juice, and the detection limit was 2.6 x 10(1) CFU/mL. The developed real time PCR system was used to detect A. acidoterrestris in 36 artificially contaminated apple juice samples and guaiacol production in the sample was also analyzed by GC-MS. The Gompertz model was employed to describe the relationship between A. acidoterrestris cell concentration and guaiacol content, and the value of R-2 was 0.854. This work provides an alternative to conventional methods of guaiacol quantification and A. acidoterrestris detection and could be very useful for the early recognition of A. acidoterrestris contamination in fruit juices.

Abstract:The immunomagnetic separation (IMS) technique was used in combination with an enzyme-linked immunosorbent assay (ELISA) procedure to shorten the total analysis time and improve the sensitivity for the detection of Alicyclobacillus spp. in apple juice samples. The specificity of IMS–ELISA for twenty strains of Alicyclobacillus spp. and eighteen strains of non-Alicyclobacillus spp. was determined and there was little cross-reaction with non-Alicyclobacillus strains. Artificially contaminated apple juice with different concentrations of Alicyclobacillus acidoterrestris was detected by IMS–ELISA, and the detection limit of the assay in apple juice was 103CFU/mL. Furthermore, the sample inoculated with 1CFU/mL of A. acidoterrestris could be detected as positive after incubation for 24h. The IMS–ELISA described, allows for the identification of suspect positive samples within 3h of testing versus 3–5days required by standard culture methods while significantly reducing the materials and labor required for the detection of Alicyclobacillus spp. in apple juice samples. As compared with the standard culture method performed concurrently on the same set of samples, the sensitivity, specificity and accuracy of IMS–ELISA for 102 naturally contaminated apple juice samples were 91.3%, 96.02% and 95.09%, respectively. These results demonstrated that the newly proposed IMS–ELISA procedure can be a potentially useful analytical method for the detection of Alicyclobacillus spp. in apple juice.
A G-quadruplex DNAzyme-based colorimetric method for facile detection of Alicyclobacillus acidoterrestris.

Abstract:The rapid and sensitive detection of Alicyclobacillus acidoterrestris (AA) has become very important due to the frequent occurrence of fruit juice spoilage by AA. In the present study, using guaiacol, both as the metabolic product of AA related to its concentration and as a green colorimetric substrate of G-quadruplex DNAzyme, a novel G-quadruplex DNAzyme-based colorimetric method for a rapid detection of AA has been developed for the first time. Under optimal conditions, AA has been successfully detected in the concentration range of 10(2)-10(5) cfu mL(-1) with a detection limit of 85 cfu mL(-1). The recoveries ranging from 71.8% to 115.7% with relative standard deviation from 1.2% to 6.6% in spiked apple and orange juice samples were obtained. Results demonstrate that the sensitivity and precision of the developed method is comparable with most other analytical methods and is prominently rapid than them. We believe that the work provides a novel and effective approach and is beneficial for monitoring and reducing the risk of AA contaminations during the process of fruit juice production.

Abstract:Alicyclobacillus spp. are heat-resistant, acid-tolerant spore-forming bacteria that could survive in commercial pasteurized fruit juice and cause spoilage along with the production of medical, phenolic, or antiseptic off-flavor, with or without visible sediment. As spoilage poses a great challenge to fruit juice industry, the aim of the present paper has been twofold. Firstly, to explore various identification, detection, and control approaches that have been applied in order to prevent possible contamination, thus summarizing the characteristics of spoilage spawned by Alicyclobacillus spp. since the first relevant juice spoilage was reported. Secondly, to provide useful information to the juice producers by critically reviewing both conventional and novel detection technologies targeting either the organisms of Alicyclobacillus spp. or the off-flavors caused and by encouraging the study and application of novel preservation procedures of fruit juices, such as high pressure, multiple hurdles, and alternative natural antibiotics.
Spoilage Effect of Alicyclobacillus on the Quality of Apple Juice

Abstract:In order to investigate the spoilage effect of Alicyclobacillus separated from productive line of concentrated apple juice on the quality of apple juice. In this paper, DSM3922 and the other eighteen Alicyclobacillus strains through purification and primary identification have been inoculated into 100% apple juice and cultivated at 37 ℃. Samples were taken and OD360nm, turbidity, appearance, and flavors have been detected and recorded. From these data, we can tell that all the tested bacteria could make apple juice turbid, deposited, bad smelled, and spoiled. The longer the juice cultivated, the worse quality it can be. Among these strains, DSMZ3922, 7- 2, 2- 1 and 3- 5 have more spoilage effect on apple juice.

Abstract:Alicyclobacillus acidoterrestris greatly affects the quality of fruit juice products. In this study, we propose a new strategy that combines an aptamer and antibody with magnetic nanoparticles and fluorescence detection to effectively detect A. acidoterrestris in fruit juices. The aptamer-coated magnetic nanoparticles (MNP-APTs) were prepared for specific separation of target bacteria, and the antibody-stabilized quantum dots (QDs-Abs) were synthesized for fluorescence detection. For reverse detection of the target bacteria, we measured the residual quantum dot fluorescence signal in the system. The detection limit of the established method was 103 CFU/mL, and the entire detection process was completed within 90 min. The results demonstrate that the constructed MNP-APTs capture probe and QDs-Abs recognition probe could be successfully applied to the rapid separation and detection of A. acidoterrestris in apple juice, which provide a new idea for the detection of contaminated substances in juice production.

Abstract:A sort of specific polyclonal anti-Alicyclobacillus antibody was generated by immunizing New Zealand white rabbits, and a sensitive indirect enzyme-linked immunosorbent assay (ELISA) was developed for Alicyclobacillus detection in apple juice. A set of experimental parameters such as concentration of antigen, dilutions of the antibody and goat anti-rabbit IgG-horseradish peroxidase conjugate, selection of the blocking reagent, incubation time, and temperature was optimized. The cross-reactivity of the antibody was evaluated by ELISA and the result was consistent with Western blot analysis. The detection limit of the ELISA was about 10(5) colony forming units (CFU)/mL in apple juice samples. Samples were detected by ELISA and conventional culture method, and the ELISA results gave a good agreement with the results obtained by plating on Alicyclobacillus acidoterrestris medium agar. ELISA takes a total detection time of 6 to 7 h, which is less than the time of conventional techniques requiring more than 24 to 48 h. These results indicated that the established ELISA was a potential useful analytical method for detection of Alicyclobacillus in apple juice.
Early Detection of Zygosaccharomyces Rouxii—spawned Spoilage in Apple Juice by Electronic Nose Combined with Chemometrics

Abstract:Spoilage spawned by Zygosaccharomyces rouxii can cause sensory defect in apple juice, which could hardly be perceived in the early stage and therefore would lead to the serious economic loss. Thus, it is essential to detect the contamination in early stage to avoid costly waste of products or recalls. In this work the performance of an electronic nose (e-nose) coupled with chemometric analysis was evaluated for diagnosis of the contamination in apple juice, using test panel evaluation as reference. The feasibility of using e-nose responses to predict the spoilage level of apple juice was also evaluated. Coupled with linear discriminant analysis (LDA), detection of the contamination was achieved after 12h, corresponding to the cell concentration of less than 2.0log10CFU/mL, the level at which the test panelists could not yet identify the contamination, indicating that the signals of e-nose could be utilized as early indicators for the onset of contamination. Loading analysis indicated that sensors 2, 6, 7 and 8 were the most important in the detection of Z. rouxii-contaminated apple juice. Moreover, Z. rouxii counts in unknown samples could be well predicted by the established models using partial least squares (PLS) algorithm with high correlation coefficient (R) of 0.98 (Z. rouxii strain ATCC 2623 and ATCC 8383) and 0.97 (Z. rouxii strain B-WHX-12-53). Based on these results, e-nose appears to be promising for rapid analysis of the odor in apple juice during processing or on the shelf to realize the early detection of potential contamination caused by Z. rouxii strains.

Abstract:Apples are produced using a high-density plantation system using M.9 rootstock in Korea. However, the virus infection rate of apples tends to be high because the rootstocks produced from virus-infected mother stocks were widely used and vegetative propagation based on the grafting is common with scion cultivars. The most effective way to control plant virus and viroid depends on conducting a massively rapid and precise diagnostic inspection. Therefore, this study aims to develop a real-time PCR assay that can be precisely and inexpensively tested for ACLSV and ASSVd. To expand the detection range, we developed a singleplex real-time PCR method targeting the conserved region of ACLSV and ASSVd, respectively. The detection methods using two kinds of fluorescent chemical dye, SYBR Green I and TaqMan probe, have been developed in this study. Comparing these methods developed with two dyes, the assay using SYBR Green I was efficient for detection as much as the method using TaqMan probe and faster. The field test demonstrated real-time detecting methods developed using SYBR Green I dye were applicable to reliable diagnosis and quantification for virus and viroid in quarantine fields.

Abstract:An approach based on multifunctional fluorescent magnetic nanoparticles was proposed for the enrichment and identification of Alicyclobacillus spp. in apple juice simultaneously. The prepared Fe3O4 magnetic particles (MNPs) were modified by the sol-gel process and a silica shell was formed to improve the reactivity, and then the obtained MNPs@SiO2-SH nanoparticles were conjugated with Thioglycolic acid functionalized CdTe/CdS QDs via thiols chemistry. The characteristic evaluation results indicated that the MNPs-QD nanocomposites exhibited good magnetic properties and optical characterization. The polyclonal anti-Alicyclobacillus IgG antibody was immobilized onto the surface of MNPs-QD materials via esterification reactions. The maximum antibody immobilization capacity was 119.62 μg/mg and the adsorption reaction could be accomplished in 60 min. The adsorption process could be represented by Langmuir model and pseudo-second order kinetics equation, respectively. Based on the high immunocapture efficiency and sensitive fluorescence characteristics, the obtained MNPs-QDs-antibody conjugates could be applied to recognize the contamination of Alicyclobacillus spp. and a quantitative analysis method was established for target cells detection. The minimum quantitative limit was 104 CFU (colony forming unit)/mL and the testing process could be completed in 90 min. The results indicated that the MNPs-QDs-antibody conjugates can be successfully applied for immunocapture and detection of Alicyclobacillus spp. in apple juice. That is to say, the developed MNPs-QDs-antibody conjugates have exhibit more attractive and great potential for the immunocapture and recognition of target bacteria, fully demonstrated a new method for enrichment and rapid detection of Alicyclobacillus spp. in fruit juices.
Rapid and visual detection of Listeria monocytogenes based on polymerase spiral reaction in fresh-cut fruit

Abstract:Widespread foodborne pathogen Listeria monocytogenes exhibits a pronounced fatality rate among individuals with compromised immune systems, the elderly, and pregnant women. A simple and user-friendly approach is required to identify L. monocytogenes . A distinctive thermostatic nucleic acid amplification technology called polymerase spiral reaction (PSR) has been extensively employed in the identification of foodborne pathogens. In the study, a straightforward and intuitive approach for identifying L. monocytogenes in fresh-cut fruit was established using PSR in conjunction with hydroxy naphthol blue (HNB). Following optimization, the system's primary constitutes-betaine, dNTP, and MgSO 4 were found to be 0.5 M, 1.0 mM, and 6 mM, respectively. Six strains of non- L. monocytogenes were used to test the assay's specificity. The PSR assay performed at 65 °C for 50 min demonstrated the capability to detect L. monocytogenes at level as low as 1×10 −4 ng/μL DNA per tube and 5.1×10 1 CFU/g of freshly cut fruit. Notably, the sensitivity of this approach surpassed that of PCR by10-fold (1×10 −3 ng/μL). In order to detect L. monocytogenes , the PSR assay was designed to be rapid, accurate, effective, and apparatus free. This innovative assay has provided researchers a fresh perspective on identification harmful microorganisms.
Condition Optimization of Real Time PCR Used in Rapid Detection of Penicillium Expansum in Apple Juice Concentrate.

Abstract:In order to to detect Penicillium expansum rapidly in apple juice concentration,the real time PCR method was used to detect Penicillium expansum and the conditions were also optimized.Based on the primers from polygatacturonase gene,the effect of annealing temperature,primer concentration and template concentration of real time PCR on the amplification was evaluated.The result showed that the amplification temperature of Penicillium expansum DNA was 61 ℃ when using Lightcycler,primer concentration was between 0.20-0.40 μmol/L,and the template concentration had little effect on amplification.Using above mentioned parameters,the best amplification,product melting curve and clear bands could be obtained.The parameters to detect Penicillium expansum obtained in this study can be used in the detection of Penicillium expansum in apple juice production,it can be also used as a reference for real time PCR detection for other microbes.
Isolation and Characterization of Thermo-Acidophilic Endospore-Forming Bacteria from the Concentrated Apple Juice-Processing Environment

Abstract:Forty-five thermo-acidophilic, spore-forming bacteria were isolated from a concentrated apple juice-processing environment. All of them were Gram-positive, rod shaped, and strictly aerobic that most likely belong to the genus of Alicyclobacillus. A fast identification method-16S rDNA PCR-RFLP was used to identify them. The results indicated that at the similarity level of 87%, apple juice isolates strains of 1-4 and 1-2-4 clustered with the reference strain of A. acidoterrstris DSM 3922T, and 4-2-1, S-22 and 5-1 with A. cycloheptanicus DSM 4006T, respectively. The other tested strains were different from all the reference strains in this study and may be new species of Alicyclobacillus genus or the other. In order to confirm this conclusion, we selected 7 16S rDNA PCR-RFLP identified strains and 5 type strains of Alicyclobacillus genus, carried on 51 kinds of phenotypic characteristics and analysis the data by unweighted pair group method with arithmetic mean (UPGMA). The results showed that the similarity degree between every two strains was lower than 80%. It also suggested that they may be different from each other and the unidentified strains may be new species. In addition, spoilage effects of them on 12 Brix apple juice were also studied. The result suggested that all 19 tested bacterial strains caused apple juice to become turbid, form a precipitate and off odor at varying rates when incubated at 37 degrees C up to 12 days. It suggested that these bacteria are associated with the spoilage of apple juice during storage.
