The Protective Effects of Thiamine on Subventricular Zone Neurogenesis of MPTP-induced Parkinson's Disease Model in Adult Mice

Abstract:Aim: To examine the protective effects of thiamine on subventricular zone(SVZ) neurogenesis of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)-induced Parkinson's disease(PD) model in adult C57BL mice.Methods: PD mice were injected i.p.with MPTP(four times in one day at two hours intervals,20 mg·g-1 per time).PDT mice were made by i.p.injection with thiamine in PD mice(from one week before the injection of MPTP to one week after MPTP injection,100 mg·g-1·-1).The mice were administrated with BrdU by i.p.injection before they were killed.The locations and numbers of BrdU and Dcx at SVZ were observed by immunohistochemistry stain.Results: The number of BrdU-positive cells and the immunoreactivity of Dcx decreased significantly in PD mice [BrdU(89.3±8.89) pix/mm2;Dcx(1 507±105.67) pix/mm2] compared with the control group [BrdU(123.2±10.09) pix/mm2,Dcx(2 341±112.38) pix/mm2].Thiamine administration in PD mice(PDT) showed increased BrdU and Dcx-positive cells which were different dramatically compared with the PD group [PDT group BrdU(116.5±11.56) pix/mm2,Dcx(1 964±89.89) pix/mm2,PD group BrdU(89.3±8.89) pix/mm2,Dcx(1 507±105.67) pix/mm2].Conclusion: The present results suggest that SVZ neurogenesis of MPTP-induced acute PD model decrease and thiamine plays a protective effect on MPTP-damaged SVZ neurogenesis of C57BL mice.
