Abstract:The key step of tofu processing is the formation of tofu gel.Generally,after the thermal denaturation of soybean storage proteins (including glycinin and ~conglycinin) in soymilk,hydrophobic groups embedded in intemal protein molecules exposed and aggregation of these exposed protein occur.The combination of these aggregated proteins was promoted to form the compact,uniform,and stable network structure by appropriate coagulants,such as salts,acids,and enzymes.The formation of tofu gel was affected by vadous factors.In this paper,the influence factors and gelation mechanism of soybean curd had been reviewed,including constitution of soybean protein,heating process of soymilk and coagulants.The discussion of the gelation mechanism of tofu aimed to provide new ideas for the formation theory of gelation food,the parameter selection of tofu processing,and the exploitation of new tofu products.

Abstract:Soy protein gel can be developed into a variety of products, ranging from traditional food (e.g., tofu) to newly developed food (e.g., soy yogurt and meat analog). So far, efforts are still needed to be made on modifying the gel properties of soy protein for improving its sensory properties as animal protein-based food substitutes. Furthermore, there is always a need to regulate its gel properties for designing novel and tailored products of soy protein gels due to the fast-growing plant protein-based product market. This review gave an emphasis on the latest modification strategies and applications of gel properties of soy protein. The modifying methods of soy protein gel properties were reviewed from an aspect of composition or processing. Compositional modification included changing protein composition and gelling conditions and using additives, whereas processing strategies can be achieved through physical, chemical, and enzymatic treatments. Several compositional modification and processing strategies have been both proven to alter the gel properties of soy protein effectively. So far, soy protein gel has been applied in the field of food and biomedicine. In the future, more mechanistic studies on the modification methods are still needed to facilitate the full application of soy protein gel.

Abstract:Plant proteins have the advantages of low cost and high yield, but they are still not comparable to animal proteins in processing due to factors such as gelation and solubility. How to enhance the processing performance of plant proteins by simple and green modification means has become a hot research topic nowadays. Based on the above problems, we studied the effect of gel induction on its properties. In this study, a pea protein–zein complex was prepared by the pH cycle method, and the effects of different induced gel methods on the gel properties of the complex protein were studied. The conclusions are as follows: All three gel induction methods can make the complex protein form a gel system, among which the gel strength of heat treatment and the TG enzyme-inducted group is the highest (372.84 g). Through the observation of the gel microstructure, the gel double network structure disappears and the structure becomes denser, which leads to a stronger water-binding state of the gel sample in the collaborative treatment group. In the simulated digestion experiment, heat treatment and enzyme-induced samples showed the best slow-release effect. This study provides a new method for the preparation of multi-vegetable protein gels and lays a theoretical foundation for their application in food processing.

Abstract:Soybean protein isolate (SPI) is a high-quality plant protein that is primarily used to process various soybean products coagulated by transglutaminase (TGase). In this study, the degree of hydrolysis (DH), sulfhydryl content (SH), surface hydrophobicity (H0 ), secondary structural constitution, and microstructure of TGase-treated soybean protein (SPI, 7S, and 11S) were determined, as well as the effects of NaCl, urea, and SDS on the properties and intermolecular forces of SPI gel were analyzed. The results show that the H0 and SH content of SPI, 7S, and 11S decreased significantly with TGase treatment time (p < .05), while the DH gradually increased and reached its highest value (3.72%, 7.41%, and 1.27%, respectively) at 30 min. As the concentration of these two secondary structures exhibited an inverse relationship, the degradation of β-turns resulted in the increase in β-sheets. The microstructures of SPI and 11S gels were similar, being denser and more ordered than 7S gel. The low concentration of NaCl solution (0.2 mol/L) enhanced gel properties and intermolecular forces, promoting the formation of SPI gel, whereas a high concentration (0.4-0.8 mol/L) had a significant inhibitory effect. Urea and SDS solutions substantially inhibited the formation of SPI gel, leading to significant decreases in the water holding capacity and hardness as well as a considerable increase in the coagulation time (p < .05). The results revealed that hydrogen bonds and hydrophobic interactions were the main intermolecular forces responsible for the gel formation. This study provides adequate technical support and a theoretical basis for soybean protein gel products.

Abstract:Soy protein isolate(SPI) were preheated at 80℃ for 10 hours with sodium chloride as a coagulant.protein was denatured at 95℃ for 30 minutes.The different concentration,ionic strength and pH conditions of denatured protein were studied.The soybean protein texture and gel properties were analyzed.The results showed that the solubility of the protein with pH7.0 and preheated at 80℃ for 10 hours was less than untreated SPI.;the protein gel formation with pH2 and preheated at 80℃ for 10 hours is much better than untreated SPI;in a certain range,with the increase of protein concentration and ionic strength,the gel formation is getting better.

Abstract:Objective: To examine the effectiveness of transglutaminase as a catalyst for the interaction of chicken and soy proteins and investigate the contribution of enzyme-induced cross-linking to the gel-forming ability and emulsifying properties of the protein mixtures. Methods: Sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) ,differential scanning calorimetry(DSC) ,fluorescence spectra were applied to study protein-protein interactions. Results: TG could be able to catalyze the covalent cross-linking reaction of chicken and soy proteins effectively,producing large protein molecules to form a stable structure with three-dimensional network,which made the water holding capacity and emulsifying capacity of the protein mixtures be improved very well,respectively to 95% and 13 m2/g. Conclusion: Enzyme-catalyzed covalent cross-linking improved the protein's properties,such as gelation and emulsification,which could be to maximize the functional characteristics of soy protein in chicken products.

Abstract:The soybean glycinin (11S)-chitosan (CS) complex gels with various textural properties were successfully constructed. The process involved the initial formation of 11S-CS coacervates through electrostatic interactions, followed by a heating treatment to obtain the final complex gels. The impacts of pH, heating temperature, and centrifugation on 11S-CS complex gel properties were investigated. The results indicated that the pore arrangement of the gel formed at pH 7.3 was more tightly and uniformly packed than those formed at pH 6.8 and 7.8. Centrifugation facilitated denser and more ordered gel structures at the three pH values, while increasing the heating temperature exhibited the opposite trend at pH 6.8 and 7.8. These structural differences were also reflected in the rheological and textural properties of the gel. The 11S-CS complex gels exhibited an elasticity-based gel property. The textural properties of gels formed at pH 6.8 were stronger compared to those formed at pH 7.3 and 7.8. However, when the 11S-CS coacervates were heated without centrifugation, the resulting gels were weak. This study emphasizes the potential of using protein/polysaccharide associative interactions during gel formation to alter the microstructure of the gel, meeting various production requirements.
Enzymatic-induced Gelation Properties of Protein Isolates from Phaseolus

Abstract:Microbial transglutaminase-induced(MTGase) gelation properties of protein isolates from red kidney bean,red bean,mung bean and soybean(designated as KPI,RPI,MPI and SPI) were investigated.SDS-PAGE analysis and dynamic oscillatory measurement indicated that the time needed to gelation onset(tgel) exhibited a following order,KPIMPISPIRPI,whereas the order of G' value after MTGase incubation was RPI MPI KPI SPI.Protein solubility analysis of MTGase-induced gels suggested that electrostatic interaction,hydrophobic interaction and hydrogen bonds except covalent bonds were the primary forces responsible for the formation of gels.In addition,disulfide bonds were also involved in gelation.Therefore,compared to SPI,the effects of protein isolates from Phaseolus on gelation stability were less obvious due to the devoid of disulfide bonds among individual subunits in protein isolates from Phaseolus.
Mechanism of ultrasound-induced soybean/egg white composite gelation: Gel properties, morphological structure and co-aggregation kinetics

Abstract:This study aims to develop ultrasound-assisted acid-induced egg white protein (EWP)-soy protein isolate (SPI) composite gels and to investigate the mechanistic relationship between the co-aggregation behavior of composite proteins and gel properties through aggregation kinetics monitored continuously by turbidity. The results showed that the inclusion of EWP caused the attenuation of gel properties and maximum aggregation (Amax) because EWP could aggregate with SPI at a higher rate (Kapp), which impeded the interaction between SPI and the formation of a continuous gelling network. In the EWP-dominated system, SPI with higher molecular weights also increased the fractal dimension of gels. Ultrasound improved properties of composite gels, especially the SPI-dominated system. After ultrasound treatment, the small, uniform size of co-aggregates and the decrease in potential led to an increase in the aggregation rate and formation of dense particles, consistent with an increase in gelation rate and texture properties. Excessively fast aggregation generated coarse chains and more pores. Still, the exposure of free sulfhydryl groups assisted the gel structure units to form a compact network through disulfide bonding. On the whole, the study could provide theoretical support for a deeper understanding on the interaction mechanism and gelation of composite proteins.

Abstract:The reaction process and corresponding mechanism of coagulation and gelation of native soy protein isolates (SPIs) induced by microbial transglutaminase (MTGase) were investigated. The protein constituents of SPIs, including a majority of subunits of beta-conglycinin and acidic subunits of glycinin, could be polymerized by MTGase to form high weight molecular (WM) biopolymers. Both the coagulation and gelation reactions of native SPI solutions induced by MTGase were dependent upon the initial protein substrate concentration ([C](0)). In the coagulating reactions, the turbidity of SPI solutions continually increased with increasing [C](0) in the range from 0.25 to 3.0%. As for the gelation reactions, with the concentration increasing from 3 to 8% (w/v), the onset time of gelation of native SPIs induced by 0.8 units/mL of MTGase at 37C shortened by similar to 5-fold, and the storage modulus (G') of finally formed gels (after 4 h) increased from similar to 1 to 1300 Pa. Both the coagulation and gelation reactions of SPI solutions were promoted remarkably by increasing the enzyme concentration. Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis analysis showed that the protein constituents of MTGase-induced aggregates of SPI (2% w/v) were mainly composed of basic subunits of glycinin and some of newly cross-linked high MW biopolymers. The solubility analysis of protein constituents indicated that the covalent cross-linkage, hydrophobic and H bindings and disulfide bonds were mainly involved in the coagulation of SPI induced by MTGase.

Abstract:EDTAD modified soyprotein gel was prepared by acylation with ethylenediaminetetraacetic dianhydride (EDTAD), followed by crosslinked with glutaraldehyde. The variation of functional groups content of acylated soy protein isolate(SPI)was analysed, the tensile strength of dry gels was investigated, and the state of water in gels was studied. The results indicate that the carboxyl content increases after acylation reaction in SPI molecules;plastic deformation appears in gels, and toughness of gels increases;the analytical results of differential scanning calorimetry (DSC)and thermogravimetry(TG)show that the content of non-freezing water in gels increases and that of freezing water decreases with increase of acylation degree of EDTAD, whearas the equilibrium water content shows less variation. With proper acylation degree of EDTAD the hydrophilie carboxyl content in protein gels also the repulsive force between protein molecule chains increases, which produces a looser polymer network structure and strengthens a bound of the protein molecule chains to the non-freezing water.

Abstract:In order to improve the nutritional and gelling properties of soy protein isolate (SPI) gel system, potato protein (PP), egg white (EW) and PP-EW mixtures were added. The texture properties, intermolecular forces, and sec-ondary structures of the gels were examined. Then the microstructure observation, rheological measurement and digestibility assay were conducted. The results showed that the gel of 3PP-3EW had the highest proportion of beta-sheet (47.04%), the most ordered gel structure, and the highest proportion of immobilized water (96.03%). The digestibility of SPI-PP-EW gels was higher than that of SPI-PP gel or SPI-EW gel. With the increase of EW pro-portion, the gel hardness increased along with the increased proportion of covalent bonds. This study is conducive to the development of meat analogues with better amino acid pattern.

Abstract:High pressure processing is a recently developing food processing technology, which can avoid the defects caused by heating, and keep the original color, flavour, taste and nutrition of food. Soybean protein is an economical and nutritious food and has promising application prospect. The Isolated Soybean protein is studied. The gel strength and appearance of high pressure induced gel and heat induced gel are analysed. The gel strength of high pressure induced isolated soybean protein gel is affected by temperature, pressure, time and the concentration of isolated soybean protein. The gel strength of high pressure induced isolated soybean protein gel was higher than that of the heat induced one. The appearance of the high pressure induced gel is more smooth and finer than that of heat induced gel. High quality gel can be obtained by high pressure than by heating.

Abstract:Soy protein isolate (SPI) and plaster were used to mimic the gelation of traditional tofu to investigate the effects of intermolecular forces on formation and texture properties of soy protein gels. The results showed that the formation of soy protein gel is influenced by multifarious intermolecular forces and electrostatic forces, and hydrophobic interactions and hydrogen bonds make predominant contributions to the gelation of SPI induced by plaster addition.

Abstract:The self-assembly fibrillar aggregations of soy glycinin and β-conglycinin at pH 2.0,85 ℃ with various protein concentrations were investigated using dynamic light scattering(DLS) and Thioflavin T fluorescence techniques.The heat-induced gelation properties including rheology,hardness and network structure of different fibrillar aggregations were characterized simultaneously.The results showed that protein concentration played an important role in the self-assembly aggregate formation at low pH and low ionic strength.The data suggested fibrillar aggregation progressively increased with the increasing of protein concentration.β-Conglycinin exhibited a higher ability to form thermally fibrillar aggregates than glycinin.The increasing degree of fibrillar aggregate was favorable for enhancing the network structure of heat-set gels.The hardness of glycinin fibril gel was stronger than that of β-conglycinin.Scanning electron microscopy observation indicated that the network of β-conglycinin fibril gel was more compact than glycinin,while the latter one was more in order.
Analysis of influence mechanism of grinding on the gel properties of soybean protein isolate based on the changes of its structure and solubility

Abstract:Abstract The compact spherical structure of soy protein isolate results in low protein flexibility and poor functional properties. In this study, the influence mechanism of grinding on its gel properties was analyzed according to the changes of soy protein isolate structure and solubility after grinding. The results indicated that at 15 min of grinding, β-sheet of the soy protein isolate decreased from 38.54% to 29.15%, the particle size became smaller, and the original solid sphere became a hollow sphere, the protein had a more disordered secondary structure, and the solubility increased to 28.24%. The glucono-δ-lactone-induced cold-set gel of this protein exhibited higher water holding capacity (76.37%), storage modulus (G') and loss modulus (G″). Creep recovery test showed that this gel had better elasticity and rigid structure at 15 min of grinding. This study could lay a theoretical foundation for research on improving the gel properties of soy protein isolate by mechanical modification in future.

Abstract:Gelation property, thermal property, protein subunits distribution, and molecular forces involved in gelation of alcohol-extracted soy protein isolate were investigated using texture analyzer, differential scanning calorimeter, sodium dodecyl sulfate polyacrylamide gel electrophoresis, and various reagents. Results showed that salts and pH played important roles in gel firmness, and a power law relationship between gel firmness and protein concentration was observed. The effects of various reagents revealed that disulfide bonds play a major role in soy protein gel formation, while the involvement of electrostatic interactions, hydrogen bonds, and/or hydrophobic interactions also occurred in gel networks. Thermal analysis indicated that both alcohol-extracted soy protein isolate and commercial soy protein isolate (isoelectric precipitation) have undergone serious denaturation, while the gel firmness of alcohol-extracted soy protein isolate was significantly greater than that of commercial soy protein isolate. Sodium dodecyl sulfate polyacrylamide gel electrophoresis image showed that there was almost no difference for protein subunits among alcohol-extracted soy protein isolate, commercial soy protein isolate, and soy powder. Hence, as an alternative environmental friendly extraction method, alcohol-extraction of soy protein isolate has a great prospect to replace presently applied isoelectric precipitation method.
Influence of Processing Parameters on Transglutaminase-Induced Gelation of Soy Protein Isolate.

Abstract:The effects of processing parameters,including incubation temperature,substrate protein and enzyme concentrations,thermal pretreatment and the addition of salts on the microbial transglutaminase(MTGase)-induced gelation of soy protein isolate(SPI) were investigated by dynamic viscoelastic measurements.Some characteristics of the gelation,including on-set time of gelation(tgel) and mechanical moduli(storage modulus,G′ and loss modulus,G″) of formed gels were evaluated.The tgel gradually decreased with increasing incubation temperature in the range from 15 to 60 °C,while highest G′ and G″ of formed gels(at 180 min) were obtained at 37 °C.At this optimal temperature,the tgel progressively decreased,and the G′ and G″ increased,with increasing both protein(4.0~8.0%,m/V) and enzyme(0.2~0.8 U/mL) concentrations.The moderate thermal pre-treatments significantly shortened the tgel,and improved the development of mechanical moduli of formed gels.However,the presence of salts(such as NaCl and CaCl2) markedly weakened the gelation.
