Tolerance to Lipopolysaccharide and Aspergillus Fumigatus Suppress the Inflammatory Responses in Cornea

Abstract:To determine whether lipopolysaccharide (LPS) and Aspergillus fumigatus (AF) can induce tolerance responses in cornea cells and improve the defensive function of cornea to the pathogen stimulation, the telomerase-immortalized human cor-neal epithelial cells (THCE) and the telomerase-immunortalized human stroma fibroblasts (THSF) were pre-treated with small amount and then re-stimulated with larger amount of of LPS and AF respectively.The cellular supernatants were collected 4 hours after stimulation and tested with ELISA to detect the expression of cytokine IL-8, and the polymorphonuclear leukocytes (PMN) migration was assayed using 24 well Transwell filters.Meanwhile, the mRNA expressions of antimicrobial peptides CC chemokine-ligand CCL20 and LL-37 were detected 20 hours after stimulation with real-time RT-PCR assay.The experimen-tal results demonstrated that the secretion of IL-8 from THCE and THSF were markedly inhibited when these cells were pre-treated with LPS and AF.and the pre-treatment of THSF with low-dose LPS resulted in diminished production of IL-8 and ele-vated expression of CCL20 upon subsequent a high dose of LPS.Furthermore, LPS pretreatment resulted in suppression of PMN infiltration.This may induce protective mechanisms during keratitis to prevent an excessive inflammatory response and to controlinfection of the cornea.
