Abstract:Two new steroidal glycosides, named dioseptemlosides I (1) and J (2), along with two known trihydroxy fatty acids, (12Z,15Z)-9,10,11-trihydroxy-12,15-octadecadienoic acid (3) and (12Z)-9,10,11-trihydroxy-12-octadecenoic acid (4), were isolated from the rhizomes of Dioscorea septemloba. Their structures were determined by HRESIMS, 1D and 2D NMR experiments, physical data, and chemical methods. The antitumor activity of compounds 1-4 was evaluated against three tumor cell lines and all of them were inactive at a concentration of 10 mu M.

Abstract:A new steroidal glycoside (1), (25R)-14α, 17α-hydroxyspirost-5-en-3β-yl 3-O-α-L-rhamnpyranosyl-(1 → 2)-β-D-glucopyranosyl-(1 → 3)-β-D-glucopyranoside, together with three known steroidal glycosides, (25R)-3β-hydroxyspirost-5-en-1β-yl-3-O-α-L-rhamnopyranosyl-(1 → 2)-O-β-D-xylopyranosyl-(1 → 3)-α-L-arabinopyranoside (2), Cixi-ophiopogon B (3) and Cixi-ophiopogon A (4), were obtained from the tuberous roots of Ophiopogon japonicus (Liliaceae). Compound 2 was isolated from the Ophiopogon genus for the first time. Their structures were identified on the basis of extensive mass and nuclear magnetic resonance spectroscopic analysis.

Abstract:A new acetylated spirostanol saponin (7), dioscin-6′-O-acetate, was isolated from the rhizomes of Dioscorea althaeoides R. Knuth (Dioscoreaceae), together with fourteen known compounds, including three spirostanol saponins (1–2, 6), three furostanol saponins (10–12), one diarylheptanoid (3), three sterols (4–5, 9), three aliphatic acids (8, 13–14) and one aliphatic alkane (15). Their structures were established by extensive 1D- and 2D- NMR (DEPT, 1H–1H COSY, HSQC, HMBC and NOESY) experiments in conjunction with IR and HRESI/TOF-MS data, as well as the results of acid hydrolysis. The compounds (3–8, 10–15) were isolated from this species for the first time. The chemotaxonomic significance of these compounds was also discussed

Abstract:Four new steroidal sapogenins (1-4), named diosbulbisins A-D, two new spirostane glycosides, diosbulbisides A (5) and B (6), one new cholestane glycoside, diosbulbiside C (7), and the known compounds 8-10 were isolated from rhizomes of Dioscorea bulbifera. Their structures were elucidated by ID and 2D NMR techniques, HRFTMS, and chemical methods. The unusual furospirostanol sapogenin skeletons, as found in compounds 3 and 4, are reported in the family Dioscoreaceae for the first time. Cytotoxicity of compounds 1-10 was evaluated using two human hepatocellular carcinoma cell lines (Bel-7402 and SMMC7721).

Abstract:The 70% ethanol extract of Dioscorea septemloba rhizomes was subjected to chromatographic columns including silica gel, ODS, D101 resin, Sephadex LH-20 and HPLC, to yield three new diarylheptanoids (1-3) and two novel stilbenes (4 and 5), together with six known ones (6-11). Their structures were elucidated on the basis of extensive 1D NMR (1H and 13C), 2D NMR (COSY, HSQC, HMBC, NOESY), IR, and MS spectroscopic data analyses. Moreover, the in vitro bioactivity screening experiment suggested that compounds 6-8 and 10 could significantly increase the glucose consumption in differentiated L6 myotubes, and that 2, 9-11 displayed triglyceride inhibitory effects in HepG2 cells.

Abstract:Much attention has been paid to cholestane-type steroidal glycosides because of their importance from the perspectives of both chemical diversity and significant biological activities. A phytochemical investigation of the rhizomes of Polygonatum odoratum (Liliaceae) resulted in the isolation of three novel cholestane-type steroidal glycosides (1-3) with unique Δ(14,16)-unsaturated D-ring structures as well as two novel spirostane-type steroidal saponins (4 and 5) and three known steroidal glycosides (6-8). Their structures were determined by various spectroscopic methods and chemical reactions. Steroidal saponin 7 showed significant antifungal activity against Candida albicans JCM1542 (MIC 3.1 μg/mL) and Aspergillus fumigatus JCM1738 (MIC 6.3 μg/mL).

Abstract:Two new steroidal saponins, diosbulbisides D (1) and E (2), along with five known saponins (3-7) were isolated from the rhizomes of Dioscorea bulbifera L. Their structures were elucidated on the basis of spectral data. Compounds 6 and 7, two 3-O-trisaccharides of diosgenin spirostanes, showed moderate cytotoxic activity against human hepatocellular carcinoma cells, with IC(50) values of 3.89 mu M and 7.47 mu M on SMMC7721, and 10.87 mu M and 19.10 mu M on Bel-7402 cell lines, respectively.

Abstract:Eight new compounds, dioscorosides G (1), H1 (2), H2 (3), dioscorol B (4), dioscorosides I (5), J (6), K1 (7), and K2 (8), together with twelve known ones (9–20) were obtained from the rhizomes of Dioscorea septemloba. Their structures were elucidated by chemical and spectroscopic methods. Among the known isolates, 12–14, 18, and 20 were isolated from the Dioscoreae genus for the first time. While, 9–11, 15, and 16 were firstly obtained from the plant. Moreover, all the isolates were evaluated for in vitro anti-inflammatory potential using LPS-stimulated RAW 264.7 murine macrophages, and compounds 7, 11, 15, and 16 were found to display significant inhibition of nitrite production.

The structures of new compounds were established as 25(R)-spirost-5-en-21 beta-methyl-3 beta-ol-3-O-alpha-L-rhamnopyranosyl(1 -> 4)-beta-D-glucopyranoside (1) and (25R)-spirost-5-en-3 beta-ol-7-one 3-O-alpha-L-rhamnopyranosyl-(1 -> 4)-beta-D-glucopyranoside (2) by detailed spectroscopic analyses including 1D and 2D NMR spectral data (H-1-H-1 COSY, HSQC, and HMBC) and MS spectrometry.

Abstract:AIM:To investigate the chemical constituents of the rhizomes of Dioscorea panthaica and to search for new active compounds. RESULT:Three C 27 steroidal saponins were isolated from the fresh rhizomes of Dioscorea panthaica. Their structures were elucidated as 26 O β D glucopyranosyl (25 R ) furost 5 en 3β,22ξ,and 26 triol 3 O β L rhamnopyranosyl (1→2) β D glucopyranoside(protobioside),26 O β D glucopyranosyl (25 R ) furost 5 en 3β,22ξ,26 triol 3 O β D glucopyranosyl (1→4) [α L rhamnopyranosyl (1→2)] β D glucopyr anoside(deltosid),26 O β D glucopyranosyl (25 R ) furost 5 en 3β,22ξ,26 triol 3 O βD glucopyrannosyl (1→3) β D glucopyranosyl (1→4) [α L rhamnopyranosyl (1→2)] β D glucopyranoside respectively. CONCLUSION All the above compounds were found in this species for the first time. The isolated compounds by other researchers were enzymatic decomposed or artifact compounds.

Abstract:Two new pregnane glycosides (1, 2) together with two known saponins were isolated from the rhizomes of Dioscorea futschauensis R. KUNTH. The structures of 1 and 2 were established as 16alpha-methoxyl-3beta-[(O-alpha-L-rhamnopyranosyl-(1-->2)-O-[alpha-L-rhamnopyranosyl-(1-->4)]-beta-D-glueopyranosyl)oxy]pregn-5-en-20-one and 21-methoxyl-3beta-[(O-alpha-L-rhamnopyranosyl-(1-->2)-O-[alpha-L-rhamnopyranosyl-(1-->4)]-beta-D-glucopyranosyl)oxy]pregn- 5,16-en-20-one, respectively, on the basis of two-dimension NMR (2D NMR) and other spectral analysis. Their in vitro bioactivity against plant pathogenic fungus Pyricularia oryzae and osteoblastic proliferation stimulatory activity in the UMR106 cell line were evaluated.

Abstract:Objective To study the chemical constituents of Dioscorea septemloba.Methods The compounds were isolated by GC-MS and chromatography on silica gel column and identified on the basis of physico-chemical constants and spectral analysis.Results Thirteen compounds were isolated and their structures were identified as 4,8-dimethyl-1,7-nonadiene(Ⅰ),5-isopropyl-2,8-dimethylcyclodeca(Ⅱ),3,7-dimethyl-6-octen-1-ol-formate(Ⅲ),dodecanoic acid methyl easter(Ⅳ),N,N'-dinitro-1,2 cyclohexanddiamine(Ⅴ),1-octyn-4-ol(Ⅵ),β-sitosterol(Ⅶ),palmitic acid(Ⅷ),stigmasterol(Ⅸ),6,7-dihydroxy-2-methoxy-1,4-phenanthrenedione(Ⅹ),diosgenin(Ⅺ),ruscogenin(ⅩⅡ),and stigmasterol-3-O-β-D-glucoside(ⅩⅢ).Conclusion Compounds Ⅰ—Ⅵ,Ⅸ,Ⅹ,ⅩⅡ,and ⅩⅢ are obtained from this plant for the first time.

Among them, protoneodioscin, protodioscin, and protoneogracillin are first reported from the title plant. The structures of the compounds were established on the basis of chemical evidence and spectral analysis (H-1-NMR, C-13-NMR, H-1-H-1 COSY, HMQC, HMBC, and FAB-MS). These eight compounds all caused morphological abnormality of Pyricularia oryzae mycelia. They also showed cytotoxic activities against the cancer cell line of K562 in vitro as antineoplastic agents.

Kunth. Their structures were elucidated on the basis of spectroscopic analysis (NMR and FABMS). Their anti-fungal activity against the plant pathogenic fungus Pyricularia oryzae and cytotoxic activity on K562 cancer cell line were evaluated in vitro .

Abstract:Two new cholestanol glycosides (1 and 2) were obtained from the roots and rhizomes of Smilacina henryi. Their structures were determined as 5 alpha a-cholest-9(11)-ene-3 beta, 26-dihydroxy-16, 22-dione 3-O-beta-D-glucopyranosyl-(1 -> 2)-beta-D-galactopyrano side (1), 5 alpha-cholest-9(11)-ene-3 beta, 26-dihydroxy-16, 22-dione 3-O-beta-D-glucopyranosyl-(1 -> 2)-[beta-D-glucopyranosyl-(1 -> 3)]-beta-D -galactopyrano side (2), by physicochemical properties and spectroscopic methods. In addition, the isolated glycosides were tested for their cytotoxic activity against human HepG2 tumor cells and compound 2 showed moderate activity with IC50 value of 59.32 mu M.
