Abstract:A miniaturized multi-wavelength modulation surface plasmon resonance (SPR) analyzer was developed by our laboratory. The incident angle of light was fixed and the change in the resonant wavelength was real-time monitored. The multi-wavelength measurement can be performed. The minor optic fiber spectrometer was designed and developed. The test light was input using optical fiber; the holographic diffraction grating was fixed for spectrophotometric; linear CCD optical sensor was used for detection. The wavelength range that can be detected is 500 - 900 nm and the resolution is 0.23 nm. The computer software of control sensor data acquisition and processing were self-developed. The experimental data were collected with the signal strength to the wavelength (the actual spectrum of horizontal and vertical coordinates). The SPR analyzer is not only small, but also has good analytical performance for quantitative analysis of biomolecular interaction studies. The human immunoglobulin (IgG) was detected in real-time. With three different methods, the lowest concentration of 0.15 mgm/L of human IgG that can be detected. Meanwhile, the stability of the instrument, the effect on sample biological activity and other factors were also studied.
