Protection of Kidney by Ischemic Postconditioning Through Inhibition of TLR4/NF-κB Pathway

Abstract:Objective To investigate the protective effect of renal ischemic postconditioning(IPC) on ischemia-reperfusion injury(IRI) and its correlation with toll-like receptor(TLR) 4/nuclear factor(NF)-κB pathway in mice. Methods Forty-two BALB/c mice were randomly divided into three groups: sham operation group(n=6), IRI group(n=18) and IPR group(n=18). IRI and IPC groups were further divided into three subgroups on day 1, 3, 5 after operation with 6 mice in each subgroup. The model of IRI and IPC was established. The levels of serum creatinine(Scr) and blood urea nitrogen(BUN) and renal histopathological examination were detected on day 1, 3, 5 post-operation. Score of Jablonski Grade in acute renal tubular injury was calculated on day 1 post-operation. Expressions of TLR4 and NF-κB were detected by Wes-tern blot on day 1, 3, 5 post-operation. Results The Scr and BUN levels of IRI group were significantly higher than that of sham group at each time point post-operation (all in P0.05). On day 1 post-operation, Scr and BUN levels of IPC group were significantly lower than those of IRI group(all in P0.05). And on day 5 post-operation, the two parameters decreased significantly and recovered to normal. Compared with the sham group, renal tubular injury was more serious in IRI group and IPC group (all in P0.05). Compared with the IRI group, renal tubular injury was significantly alleviated in IPC group(P0.05). On day 1 and day 3 post-operation, the expression of TLR4 in IPC group was significantly lower than that in the IRI group (all in P0.05). On day 1, 3, 5 post-operation, the expression of NF-κB in IPC group was significantly lower than that in IRI group(all in P0.05). Conclusion IPC can reduce IRI in kidneys by inhibiting TLR4/NF-κB signaling pathway.
