Abstract:Objective To investigate the effects of leptin on proliferation and differentiation of human dental pulp stem cells (hDPSCs).Methods MTT assay and flow cytometric (FCM) were used to evaluate the proliferative effects of leptin in different concentrations. The activity of alkaline phophatase (ALP), Von Kossa and dentin sialoprotein (DSP) and osteocalcin (OCN) were detected to assess the effects of leptin on differentiation of hDPSCs.Results No significant change was observed in the proliferation of hDPSCs after incubation with different concentrations of Lepin compared with the control group. Leptin increased ALP activity of hDPSCs in dose dependently manner. Von Kossa staining showed the formation of mineralized nodules and SABC showed expression of DSP and OCN in hDPSCs after incubation with leptin.Conclusion Leptin did no effects on hDPSCs proliferation, but it can promote hDPSCs to differentiate into odontoblasts.
