Zyxin is Involved in Thrombin Signalingviainteraction with PAR‐1 Receptor

Abstract:Thrombin‐induced increase in endothelial monolayer permeability depends on reorganization of the cytoskeletal structures. Protease‐activated receptor‐1 (PAR‐1) is the main receptor responsible for thrombin signaling in endothelial cells. Here we showed that PAR‐1 receptor interacted with zyxin, a LIM domain containing phosphoprotein, both in vitro and in vivo. We have shown that LIM domain 1 and 2 of zyxin interacted with C‐terminal domain of PAR‐1 receptor. In human umbilical vein endothelial cells (HUVECs), thrombin induced recruitment of the endogenous zyxin to the plasma membrane, promoted zyxin targeting to the focal adhesion, and promoted zyxin association with the actin stress fibers. Downregulation of zyxin using siRNA technique inhibited thrombin‐induced recruitment of FAK to focal adhesions. Similarly, disruption of the interaction between endogenous zyxin of PAR‐1 receptor by overexpression of the C‐terminal domain of zyxin inhibited thrombin‐induced recruitment of FAK to focal adhesions. In addition, downregulation of zyxin using siRNA resulted in reduced cellular responses to thrombin stimulation, such as cell rounding, actin stress fiber formation and serum response factor‐dependent gene expression. These results suggest that zyxin is involved in thrombin signaling by direct interaction with PAR‐1 receptor.
