Relationship between the polymorphisms in the cholesterol ester transfer protein gene and the effect of fluvastatin

Abstract:Objective:To examine the relationship between single nucleotide polymorphisms in the cholesteryl-ester transfer protein(CETP) and variable lipid response to fluvastatin in Shanghai Han population.Method:Lipid levels were determined at baseline and following 8 weeks of treatment with 40 mg dose of fluvastatin in 121 hyperlipidemia patients,(male 49,female 72),aged(61.13±11.05) years.CETP SNP genotyping was performed to examine the associations between common SNPs and lipid response.Result:In Shanghai Han people,there are 10 SNPs was detected in CETP gene in our reseach,3 of them has never been reported before.(1) SNP1:HDL-C levels have increased 16.65% after treatment with fluvastatin in T homozygotes,but dcreased 2.49% in CC orCT subjects(P value=0.012).(2) rs 289741:G homozygotes had lower TC levels than AA or GA subjects(P value=0.003).HDL-C levels have increased 10.25% after treatment with fluvastatin in G homozygotes,but dcreased 2.22% in AA orGA subjects(P value=0.036).Conclusion:CETP has independent effects on lipid changes following fluvastatin treatment.The results of this study may lead to an improved understanding of the genetic determinants of lipids response to treatment.
