Abstract:Objective To study the chemical constituents of the marine sponge Spheciospongia sp. collected from the South China Sea. Methods The chemical constituents were isolated by various methods of isolation and their structures were elucidated by physicochemical properties and the analysis of spectral data compared with those reported in literatures. Results Six steroids including cholesterol (1), cholest-4-en-3-one (2), cholest-3,6-dione (3), 3β-hydroxy-cholest-5 -en-7-one ( 4 ), 6β-hydroxy-cholest-4 -en-3-one ( 5 ), cholest-3β, 5α,6β-triol (6)were isolated from the marine sponge Spheciospongia sp. collected from the South China Sea.Conclusions The compounds 2 -6 are isolated form the marine sponge for the first time.

Abstract:A new steroid, 24 xi, 25-dimethyl-3 alpha-hydroxyl-cholest-5-ene-2 beta-ol sodium sulfate (1), together with a known steroid, 24 xi, 25-dimethyl-cholest-5-ene-2 beta, 3 alpha-diol disodium sulfate (2), was isolated from the ethanol extract of marine sponge Halichondria rugosa. Their structures were elucidated on the base of spectroscopic analysis. Both compounds showed cytotoxicity to four human cancer cell lines (BEL-7402, HT-29, SPC-A1 and U-251) with IC50 values between 6.5 and 23.1 mu M.

Abstract:A detailed chemical investigation of two specimen of South China Sea sponges Halichondria sp. (No. 19-XD-47 and No. 17-XD-87) have resulted in the isolation of three new sterols, namely, halichsterols A (1), B (2) and C (3), together with seven related known ones (4–10). Their structures were determined by extensive spectroscopic analysis and by comparison with the spectral data reported in the literature. In bioassay, compound 2 displayed significantly anti-inflammatory activity in lipopolysaccharide (LPS)-stimulated BV-2 microglial cells.

Abstract:A new steroid, (3S,5R,9R,10S,13R,17R,20R,24S,22E)-ergosta-6,8,22-triene-3,25-diol (1), and its sulfonated analogue (2) together with a known one, 5α,8α-epidioxy-cholest-6-en-3β-ol (3) were isolated from the marine sponge Dactylospongia elegans collected from the South China Sea. The new structures including absolute configurations were established by the HRESIMS and 1D and 2D NMR analysis coupled with the X-ray crystal analysis. Both of 1 and 2 exhibited cytotoxicity against cancer cell line MCF-7 with IC50 values of 9.7 and 8.5 μM, respectively.

Abstract:Chemical investigation on CH2Cl2 extract of the marine sponge Leucetta chagosensis, collected from the South China Sea, afforded two new 5,6-epoxysterols, 5α,6α-epoxycholesta-8(14),22(E)-diene-3β,7α-diol (1) and (24R)-24-ethyl-5α,6α-epoxycholesta-8(14),22(E)-diene-3β,7α-diol (2) along with ten known related steroid analogs (3–12). Their structures were elucidated on the basis of NMR spectroscopic analyses, and comparison with the literature. All isolates were tested for cytotoxicity against three tumor cell lines only known compounds 11 and 12 exhibited notable cytotoxic activity against A549 (IC50: 3.0 and 5.6 μM), PC9 (2.0 and 15.6 μM), and MCF-7 (9.4 and 11.8 μM) cell lines, respectively.

Abstract:Five compounds with nitrogen have been isolated from Hyatella.sp collected from the Naozhou island of China South Sea and identified as:cyclo(Pro-Thr)(1),2-hydroxy-N-(1,3,4-trihydroxy-17-methyloctadecan-2-yl-18-methylicosanamide (2),2-hydroxy-N-(1,3,4-trihydroxy-17-methyloctadecan-2-yl)-19-methylheneicosanamide (3),2-hydroxy-N-(1,3,4-tri-hydroxy-17-methyl-octadecan-2-yl)-20-methylbehenamide (4) and 5-methyl-pryrimidine-2,4(1H,3H)-dione (5).Their structures were determined on the basis of chemical evidence and spectral data.Compound 1 was isolated from nature for the first time.

Abstract:Two polyhydroxylated steroids have been isolated from the South China Sea soft coral Sinularia microclavata, and their structures were established as 24-methylenecholestane-1 alpha,3 beta,5 alpha,6 beta-tetrol and 1 alpha,3 beta,5 alpha-trihydroxy-24-methylenecholestan-6-one from spectral evidence and from comparison with two reference compounds, numersterol A and 24-methylenecholestane-1 alpha,3 beta,5 alpha,6 beta, 25-pentol, which were isolated from the soft corals, Simularia numerosa and Sarcophyton glaucum, respectively.

Abstract:This paper reports the steroids from the marine sponge, Sigmadocia cymiformis, collected around Weizhou Island of Guangxi Province, and Linshui County of Hainan Province respectively. Ten sterols, cholesta-4, 6-dien-3beta-ol(1), 27-norergosta-5, 22-dien-3beta-ol(2), cholesterol(3), (24S)-ergosta-5, 22-dien-3beta-ol(4), ergosta-5-en-3beta-ol(5), ergosta-7, 22-dien-3beta-ol(6), stigmasterol(7), beta-sitosterol(8), ergosta-4, 24(28)-dien-3-one(9) and gorgost-5-en-3beta-ol(10), were identified by GC-MS from the sponge collected from Linshui County waters and seven sterols, (3), (4), (5), (6), (8), cholesta-5, 22-dien-3beta-ol(11) and cholesta-3, 5-dien-7-one(12) from the sponge from Weizhou Island waters. This is the first report for the steroids from the marine sponge Sigmadocia cymiformis.

Abstract:The marine fungus Chondrostereum sp. was collected from a soft coral Sarcophyton tortuosum from the South China Sea. This fungus was cultured in glucose-peptone-yeast (GPY) medium and the culture broth was extracted with EtOAc. By the method of 1H NMR pre-screening and tracing the diagnostic proton signals of the methyl groups, two new hirsutane-type sesquiterpenoids, chondrosterins N and O (1 and 2) were isolated from the metabolite extracts. Their structures were elucidated on the basis of MS, 1D and 2D NMR data.

Abstract:A new steroid, 2a-oxa-2-oxo-5 beta-hydroxy-3,4-dinor-24-methylcholesta-22E-ene (1), together with 10 known ones (2-11), was isolated from the marine sponge Cliona sp. The structures of these compounds were determined by the spectroscopic methods (UV, IR, MS, and NMR) and X-ray diffraction analysis. Compound 1 was the third example of 3,4-dinorsteroid with a hemiketal at C-5 that was isolated from the natural source. In addition, the antibacterial activities of these compounds were also evaluated. However, none of them exhibited significant inhibition effects.

Abstract:Five sterols with a nucleus skeleton of 6-hydroxy-4-en-3-one, namely cholesta-6beta-hydroxy-4-en-3-one (1), ergosta-6beta-hydroxy-4,24(28)-dien-3-one (2), ergosta-6alpha-hydroxy-4,24(28)-dien-3-one (3), ergosta-6alpha-hydroxy-4,22-E-dien-3-one (4), and ergosta-28-methyl-6beta-hydroxy-4,24(28)-dien-3-one (5) have been isolated from the marine sponge Iotrochoto birotulata, collected from the southern China sea. Sterols 2-4 are new compounds, and 1 has been isolated from marine organisms for the first time. The structures have been elucidated on the basis of extensive spectroscopic and chemical properties.

Abstract:Fifteen steroids were isolated from the soft coral Dendronephthya sp., of which five are determined as new compounds, namely (22E)-3-O-β-formylcholest-5,22-diene (1), (22E)-3-O-β-formyl-24-methyl-cholest-5,22-diene (2), 2-ethoxycarbonyl-2-β-hydroxy-A-nor-cholest-5-ene-4-one (3), (22E)-2-ethoxycarbonyl-2-β-hydroxy-A-nor-cholest-5,22-diene-4-one (4), and (22E)-2-ethoxycarbonyl-2-β-hydroxy-24-mthyl-A-nor-cholest-5,22-diene-4- one (5). 1 and 2 belonged to 3-O-formylated cholesterol analogues, and 3 to 5 are unique ring A-contracted steroids. Their structures were elucidated by extensive 2D NMR in association with IR, MS analysis.

Abstract:O bjective:To study the chemical constituents from the South China Sea sponge Haliclona sp..Methods:The isolation and purification of the compounds were performed by column chromatography techniques with silica gel, PTLC, Toyopearl HW-40 and semi-preparative HPLC. Their structures were determined mainly by spectral data analysis and comparison with literatures. Results: Five compounds had been isolated and their structures were identified as pregnenolone (1), 24-methylenecholesterol (2), cholesterol (3), phenylpropionic acid (4), dibutylphthalat (5). Conclusion:Compounds 4, 5 have been isolated for the first time from the genus Haliclona.
