Comparison of Surface Marker of Monocyte-Derived Dendritic Cells Between Cord Blood and Adult Peripheral Blood

Abstract:Objective To compare the phenotype changes during the differentiation and maturation between cord blood monocyte(CBMC)-derived dendritic cells(DC) and adult peripheral blood monocyte(PBMC),and to explore the possible mechanism of immature immunity in neonates. Methods The expression of CD11c,CD1a,CD80,CD86,CD40,MHCⅠ and MHCⅡ molecules on the surface of DC co-cultured with different combinations of granulocyte-macrophage colony-stimulating factor(GM-CSF),recombinant human interleukin-4(rhIL-4) and lipopolysaccharide(LPS) were detected by immunofluorescence staining and flow cytometry. Results The expression levels of CD11c and MHCⅠ molecules were similar between CBMC-derived DC and PBMC-derived DC(P0.05).The percentage of CD40 positive cells in CBMC-derived DC was lower than that in PBMC-derived DC[(34.80±7.77)% vs(54.37±9.57)%,P0.01].When co-cultured with LPS,the expression levels of CD80 and CD86 in CBMC-derived DC were lower than those in PBMC-derived DC[(3.72±1.63)% vs(7.82±0.81)% and(28.53±4.67)% vs(45.62±5.16)%,respectively,P0.01 for both].Compared with PBMC-derived DC,CBMC-derived DC had lower relative fluorescence intensity in the expression of MHCⅡ molecules(P0.01). Conclusion The significantly reduced expression of MHCⅡ molecules and costimulatory molecules of CBMC-derived DC may result in impaired neonatal T-cell function,accounting for the increased susceptibility to different infections in the neonates,as well as the lower incidence of graft-versus-host disease in cord blood transplantation.
