The Regulation Effects of TGF-β1 Secreted by Transfected Corneal Endothelial Cells on the Expression of Cytokines in Mice Helper Lymphocytes

Abstract:Objective To investigate the regulation effects of TGF-β1 secreted by transfected corneal endothelial cells on the expression of cytokines in mice helper lymphocytes. Methods Corneal endothelial cells were transfected with pSecTag2-TGF-β1MP using a Lipofectamine reagent. The resistant clones secreting the recombinant proteins were established with Hygromycin B screening. The stable expression of TGF-β1 protein was detected with immunohistochemical SABC examination. The concentration of TGF-β1 protein secreted in the corneal endothelial cell supernatant was assayed by ELISA. Expressions of IL-2、IFN-γ、TNF-α、IL-4、and IL-6mRNA in mice helper lymphocytes were detected by RT-PCR. Results SABC examination displayed all stable transfected cells that were stained to brown. The concentration of TGF-β1 in the transfected cell supernatant was (597±7) pg/ml,which was higher than the concentration in the controls. Compared to the untransfected group, the expression of IL-2、IFN-γ、and TNF-α decreased, but the expression of IL-4 and IL-6 increased significantly in mice spleen lymphocytes(P0.05). Conclusion The high expression of TGF-β1 from the transfected corneal endothelial cells could induce lymphocyte immune deviation from Th1 to Th2 and showed negative immunological activity, which may be a benefit in suppressing the immune rejection induced by corneal transplants.
