Isolation,culture and identification of the cells derived from fetal heart

Abstract:Aim: Fetal heart is in the phase of organ formation and development. Cell amount and proliferation ability are higher than these of adult organs. However, there are few reports on the culture of cells from fetal heart and its biological features. This study established a method of culture the cells derived from fetal heart in vitro and analyzed their basically biological features and sorts. Methods: Experiments were performed at the Medical School & State Key Laboratory of Pharmaceutical Biotechnology, Nanjing University from October 2005 to May 2007. 1 Ten heart samples obtained from 12-18 weeks of fetus aborted with water bag were provided by Gulou Hospital Affiliated to Medical College of Nanjing University. The parturients and their family members signed the informed consent. Experimental procedures were approved by Hospital Medical Ethical Committee. 2 The heart samples were obtained from 12-18 weeks of fetus aborted with water bag. The samples were digested and cell suspension was prepared. The obtained cells were put into the DMEM/F12 medium including epidermal growth factor, basic fibroblast growth factor, stem cells growth factor, cardiotrophin and 10% fetal bovine serum. 3 The morphology of the cells was observed with a microscope. Their gene expressions were detected by RT-PCR. The immunophenotypes of the cells were analyzed with the flow cytometer before and 19 days after culture. Results: 1 The cells derived from the fetus heart could survive and proliferate. The cells also could form clones after 16 days, and at the central region of clone the cells were round, smaller, and the nuclear was bigger. Surrounding cells were fusiform, and nuclei were polygonal and smaller. The dividing cells could be found under the microscope. 2 RT-PCR showed the expression of c-Kit, KDR, GATA-4, Nkx-2.5 and MEF-2c, other than c-Tnl in the fresh isolated cells. After 19 days, the expressions of GATA-4, MEF-2c, KDR was low, and there was not the expression of c-Kit, Nkx-2.5 and c-Tnl. 3 Flow cytometry showed CD29 and CD44 expressed highly, but the expression of CD45, GATA-4 and c-Tnt was low. Conclusion: A method for culturing human fetal heart-derived cells is successfully established. Mesenchymal stem cells and cardiac progenitor cells exist in the cell populations derived from fetal heart.
