Abstract:1. In the present study, we investigated the immunosuppressive effects and mechanisms of action of emodin on acute graft rejection following liver transplantation in a rat model of orthotopic liver transplantation. 2. Rats were divided into three groups: Group A, syngenic control (Brown Norway-to-Brown Norway); Group B, acute rejection group (Lewis-to-Brown Norway); and Group C, emodin-treated group (Lewis-to-Brown Norway treated with 50 mg/kg emodin, 50 mg/kgxd, injected intraperitoneally once a day from days 1 to 5 posttransplantation). The survival time of the recipients in each group was recorded. Histopathological changes in the liver, hepatocellular apoptosis, serum concentrations of interleukin (IL)-2, interferon (IFN)-gamma and IL-4 and their expression in liver tissue were determined. 3. Emodin treatment prolonged liver allograft survival time from 10.9 days in Group B to 25.6 days in Group C. The rejection activity index (calculated according to the Banff Schema) in Groups A, B and C was 1.29 +/- 0.47, 7.58 +/- 0.85 and 4.72 +/- 0.79, respectively (P < 0.01 for Groups A and B vs Group C), whereas the apoptosis index in the three groups was 15.51 +/- 1.47, 39.50 +/- 1.65 and 16.72 +/- 1.73, respectively (P < 0.01 for Groups A and C vs Group B). Serum levels of IL-2 and IFN-gamma were higher, whereas levels of IL-4 were lower, in the acute rejection group (Group B) than in the emodin-treated group (Group C; P < 0.05). Changes in the expression of these cytokines in transplanted liver tissue were consistent with changes in serum concentrations. 4. In conclusion, emodin effectively suppresses acute graft rejection in vivo to prolong the survival of recipient rats. The mechanism underlying this effect may be associated with the prevention of hepatocyte apoptosis and with a changing in the balance of Th1/Th2 cytokines towards Th2.
Immunosuppressive effect of emodin on acute rejection after orthotopic liver transplantation in rats

Abstract:AIM: A few reports have shown that emodin has immunosuppressive effect. This study was aimed to verify the effect of emodin on acute rejection in the early stage after hepatic allograft operation. METHODS:The experiment was performed at the Laboratory of Hepatobiliary Surgery, First Affiliated Hospital, Xi'an Jiaotong University from January 2004 to February 2005. SD→Wistar rats were selected to establish orthotopic liver transplantation models (n =80). These rats were randomized into 4 groups with 20 in each group. Rats in the model control group, emodin group, ciclosporin A group and ciclosporin A plus emodin group were intraperitoneally treated with 9 g/L saline, 1.5 mg/kg per day emodin, 3 mg/kg per day ciclosporin A and 3 mg/kg ciclosporin A plus 1.5 mg/kg emodin per day respectively. General conditions of rats were observed after operation. Ten rats in each group were killed 7 days after transplantation. Hepatic samples and serum were collected to observe the intensity of acute rejection in liver tissue, expression of Fractalkine(Fkn), levels of albumin (ALB) and alanine aminotransferase (ALT) in serum. The rest rats in each group were fed and continued to give different drugs or vehicle buffer until they were died and the mean of survival time were recorded. RESULTS: Seventeen rats in the model control group, 18 in the emodin group, 18 in the ciclosporin A group and 18 in the ciclosporin A plus emodin group. ①Compared with the model control group, the mean survival time was significantly longer in other groups (P < 0.05), and the mean survival time was the longest in the ciclosporin A plus emodin group. ②Compared with the model control group, on the 7th day after transplantation, the severity of acute rejection was remarkably lower in the other groups (P < 0.05), but ALB level was obviously increased and ALT level was markedly decreased (P < 0.05). The expression of Fkn in liver tissue was significantly decreased (P < 0.05), especially in the ciclosporin A plus emodin group. CONCLUSION:Emodin can inhibit the severity of rat hepatic allograft acute rejection, and has synergistic effect with ciclosporin A.

Abstract:BACKGROUND: High performance immunodepressive agent for inhibiting acute rejection is necessary for organ transplantation. Many Chinese medicine ingredients have immunological regulation. Previous studies of this work group successfully established rat models of whole human blood supply liver transplantation,and primarily found that emodin had inhibitory effects on acute rejection following liver transplantation in rats. These inhibitory effects could be achieved by various pathways. OBJECTIVE: To observe the effects of emodin on hepatocyte apoptosis in rats during early acute rejection following allograft liver transplantation. DESIGN,TIME AND SETTING: The randomized controlled animal study was performed at the Laboratory of Department of Hepatobiliary Surgery,First Affiliated Hospital,Xi’an Jiaotong University from April to September 2005. MATERIALS: A total of 80 Sprague Dawley rats of both genders were used as donors. A total of 80 male Wistar rats served as receptors. Rat models of Sprague Dawley→Wistar whole human blood supply liver transplantation were established. METHODS: The 80 rat models were randomly and equally divided into 4 groups. From day 1 following transplantation,rat models in the model control group intraabdominally received saline,and rat models in the emodin group underwent 1.5 mg/kg per day emodin,and those in the ciclosporin A group were subjected to ciclosporin A 3 mg/kg per day,and those in the ciclosporin A + emodin group were administered with 3 mg/kg per day + emodin 1.5 mg/kg per day. MAIN OUTCOME MEASURES: Ten rats in each group were killed 7 days after transplantation. The intensity of acute rejection in liver tissue,the apoptotic index of hepatocytes and Bcl-2 protein expression were observed. The remaining rats in each group were fed and continued to give different drugs or vehicle buffer until they were died,and the survival time were recorded. RESULTS: Compared with the model control group,the survival time was significantly longer in the emodin group,ciclosporin A group and ciclosporin A + emodin group (P < 0.05). The survival time was the longest in the ciclosporin A + emodin group. At day 7 after transplantation,the severity of acute rejection and the hepatocyte apoptotic index were significantly decreased,but Bcl-2 protein level was significantly increased in the emodin group,ciclosporin A group and ciclosporin A + emodin group compared with the model control group (P < 0.05). The severity of acute rejection and the hepatocyte apoptotic index were significantly lower,but Bcl-2 protein levels were significantly higher in the ciclosporin A + emodin group than in emodin group,ciclosporin A group and model control group (P < 0.05). CONCLUSION: Emodin inhibits hepatocyte apoptosis,improves hepatic function,has synergistic effects with ciclosporin A,and relieves hepatic functional lesion induced by ciclosporin A.
Emodin Inhibits The Differentiation And Maturation Of Dendritic Cells And Increases The Production Of Regulatory T Cells

Abstract:The aim of this study was to characterize the effects of emodin on dendritic cells (DCs) and CD4(+)CD25(+) regulatory T cells (Tregs). Myeloid DCs were prepared from peripheral blood mononuclear cells of healthy human donors and treated with emodin at different concentrations. The phenotype and T cell stimulatory capacity of these DCs were analyzed. The expression ratios of CD80 and CD83 in DCs in the presence of emodin (100 mu g/ml) were significantly decreased compared with that in DCs without emodin treatment (P<0.05). IL-12p70 production of DCs decreased significantly with emodin treatment (P<0.05). Furthermore, an approximately 2-fold decrease was observed in the ability of DCs pre-treated with emodin to induce T-lymphocyte proliferation. In addition, we found that emodin treatment increased the number of Tregs, which expressed lower levels of human leukocyte antigen (HLA-DR), glucocorticoid-induced tumor necrosis factor receptor (GITR), and cytotoxic T-lymphocyte associated antigen-4 (CTLA-4) as compared to cells without emodin treatment. Our results suggest that emodin inhibits the differentiation and maturation of DCs and induces Tregs, which may be helpful for the modulation of the immune rejection after liver transplantation.
Inflammasome Activation Involved in Early Inflammation Reaction after Liver Transplantation.

Abstract:Liver transplantation has emerged as a vital therapy for end-stage liver diseases. Acute -phase inflammation play an important role in liver graft injury.Recent studies have revealed that inflammasome are responsible for initiating inflammation in early stage of acute organ rejection in liver transplantation, however the underlying mechanism remains unclear. Here we explored to block inflammasome activation to see whether it can alleviate early inflammation reaction during rejection of allgrafts in a rat model and gain further insights into the mechanism of inhibiting inflammation in allografts. By using Ac-YVAD-CMK, a highly selective caspase-1 inhibitor, to inhibit inflammation reaction involved in allograft rejection in a rat model. Our results showed that the rejection activity index (RAI) of Ac-YVAD-CMK-treated allografts is significantly diminished in similar magnitude to that of isografts. Compared with isografts, the expression of apoptosis-associated speck-like protein containing a caspase recruitment domain (ASC) and IL-1β in allograft group increased significantly with the development of rejection, exhibiting apparent correlation. Expression of IFN-γ mRNA in untreated allografts was maximal on day 3 while in Ac-YVAD-CMK-treated allografts and isografts, IFN-γ mRNA levels remained low over the duration of the time course. ELISA results revealed serum elevation of IL-1β by day 7 after othotopic liver transplantation (OLT) in comparison with isografts. There were no statistically significant differences between isografts and Ac-YVAD-CMK-treated allografts. For the first time, our data reveal that inhibition of the inflammasome activation pathway attenuates inflammation reaction of hepatic transplant rejection.

Abstract:OBJECTIVE:This study aimed to investigate the relationship between the severity of ischemia/reperfusion (I/R) injury and the acute rejection (AR) of allogeneic liver transplants in rats.METHODS:The experimental rats were divided in different groups: normal control group (sham group, group I); syngeneic liver transplant control group (similar gene group, group II); and allogeneic liver transplant groups (groups III to VI). The rats were humanely killed at 1, 3, 5, and 7 days after transplantation or sham operation to determine the severity of I/R injury, rejection classification, and hepatocyte apoptosis. Messenger RNA (mRNA) and protein expression levels of Fas, perforin, and granzyme B were assessed in the liver tissues using real-time polymerase chain reaction and immunohistochemistry, respectively.RESULTS:The rejection scores of the transplanted liver tissues gradually increased until these scores were proportional to the severity of I/R injury in groups III, IV, and V. The maximum scores were reached at 7 days after transplantation as the duration of transplantation was extended. The mRNA and protein expression levels of Fas, perforin, and granzyme B were significantly increased at 1, 3, 3, 5, and 7 days after liver reperfusion in groups III, IV, and V compared with those in groups I, II, and VI (P < .05).CONCLUSION:The occurrence of AR after allogeneic liver transplantation in rats was positively correlated with the severity of I/R injury. Given that I/R injury caused serious damage to the transplanted liver, the occurrence of AR consequently decreased.
Effects of Emodin on Cell Apoptosis of Intestinal Mucosa and Serum Leptin in Rats with Severe Acute Pancreatitis

Abstract:OBJECTIVE:To explore the mechanisms of emodin in protecting intestinal mucosal barrier in rat with severe acute pancreatitis (SAP).METHODS:Sixty SD rats were randomly divided into three groups: sham-operation group, untreated group, and emodin group. SAP in rats of the untreated group and the emodin group was induced by retrograde pumping of 3.0% sodium cholate to the common bile duct. Specimens were obtained 24 hours after the severe acute pancreatitis was induced. Serum level of leptin, serum activity of amylase and plasma content of endotoxin were measured. Ileum mucosa from ileocecal junction was observed by light microscopy and electron microscopy to measure pathological and ultrastructural changes. Apoptosis of ileum mucosal cells was detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling method, and expression of Bax in ileum mucosal cells was measured by immunohistochemical method.RESULTS:Compared with the sham-operation group, there was significant increase in the levels of leptin, endotoxin, the activity of amylase, apoptosis index and Bax expression in the untreated group (P<0.01). Compared with the untreated group, the level of endotoxin, apoptotic index and Bax expression level in the emodin group were significantly reduced (P<0.01) and the leptin level was increased (P<0.05). More severe pathological changes appeared in the untreated group than in the sham-operation group under the light and electron microscopes; meanwhile less severe damage was observed in the emodin group as compared with the untreated group.CONCLUSION:Emodin can inhibit the apoptosis of intestinal mucosa cells and up-regulate the serum leptin content to protect the intestina1 barrier function and prevent the translocation of bacteria and endotoxin.
Effect of Intestinal Microbiota Alteration on Hepatic Damage in Rats with Acute Rejection After Liver Transplantation

Abstract:The previous studies all focus on the effect of probiotics and antibiotics on infection after liver transplantation. Here, we focus on the effect of gut microbiota alteration caused by probiotics and antibiotics on hepatic damage after allograft liver transplantation. Brown-Norway rats received saline, probiotics, or antibiotics via daily gavage for 3 weeks. Orthotopic liver transplantation (OLT) was carried out after 1 week of gavage. Alteration of the intestinal microbiota, liver function and histopathology, serum and liver cytokines, and T cells in peripheral blood and Peyer’s patch were evaluated. Distinct segregation of fecal bacterial diversity was observed in the probiotic group and antibiotic group when compared with the allograft group. As for diversity of intestinal mucosal microbiota and pathology of intestine at 2 weeks after OLT, antibiotics and probiotics had a significant effect on ileum and colon. The population of Lactobacillus and Bifidobacterium in the probiotic group was significantly greater than the antibiotic group and the allograft group. The liver injury was significantly reduced in the antibiotic group and the probiotic group compared with the allograft group. The CD4/CD8 and Treg cells in Peyer’s patch were decreased in the antibiotic group. The intestinal Treg cell and serum and liver TGF-β were increased markedly while CD4/CD8 ratio was significantly decreased in the probiotic group. It suggested that probiotics mediate their beneficial effects through increase of Treg cells and TGF-β and deduction of CD4/CD8 in rats with acute rejection (AR) after OLT.

Abstract:AIM:To study the influences of emodin and reconstruction of double blood supplies on liver regeneration of reduced size graft liver in rat model. METHODS:A total of 45 SD-SD rat reduced size liver transplantation models were randomly divided into three groups (A-C). The conventional reduced size liver transplantation was performed on rats in group A, while the hepatic artery blood supply was restored in groups B and C. The emodin (1.5 mg/kg/d) was given by intraperitoneal route in group C only. The recipients were killed on the seventh day after the operation. The proliferative cell nuclear antigen (PCNA), TBil and ALT of serum were detected, and the pathological changes of liver cell were observed. RESULTS:The numbers of the rats that survived in A, B, and C group on the seventh day after operation were 14, 13, 13, respectively. The levels of TBil (31.5+/-5.2 micromol/L, 23.2+/-3.1 micromol/L vs 38.6+/-6.8 micromol/L), and ALT (5 351+/-1 050 nKat, 1300+/-900 nKat vs 5779+/-1202 nKat) in serum in groups B and C were lower than those in group A (P<0.05), while the expression of PCNA in groups B or C was higher than that in group A (22.0+/-3.5%, 28.2+/-4.2% vs 18.6+/-3.2%, P<0.05). The deeper staining nuclei, double nuclei, multi-nuclei and much glycogen were observed in liver cells of groups B and C, especially in group C, while fewer were found in liver cells of group A. CONCLUSION:The reconstruction of arterial blood supply is very important for rat liver regeneration after reduced size liver transplantation. Emodin has the effect of promoting liver regeneration and improving liver function in rats after reduced size transplantation. The possible mechanism is improving proliferation of liver cell and protecting liver cells from injury.
Observation of Immune Status in Acute Rejection Model of DA-Lew Rat Liver Transplantation

Abstract:Objective To study the development and incidence of acute rejection in acute rejection model of DA-to-Lew rat liver transplantation.Methods The rats were divided into isogeneic(Lewis-Lewis) group(n=24) and allogeneic(DA-Lewis) group(n=24).On day 3,5,7 and day 10 after surgery,three recipient rats of each group were randomly sacrificed respectively.Pathological changes of the liver and the levels of aspartate aminotransferase(AST),total bilirubin and cytokines were observed.Results There were significant changes in the general performance of the rats,the function and pathological changes of the liver and cytokines level in all the time points of observation between allogeneic group and isogeneic group.Severe acute rejection was observed on day 7 after liver transplantation.Conclusion The study suggests severe acute rejection develops on day 7 after surgery in acute rejection model of DA-to-Lew rat liver transplantation and the severity of acute rejection can be evaluated at this time point.

Abstract:Allograft rejection continues to be a significant cause of morbidity and graft failure for liver transplant recipients. Existing immunosuppressive regimens have many drawbacks, thus safe and effective long-term immunosuppressive regimens are still required. Luteolin (LUT), a natural component found in many plants, has a variety of biological and pharmacological effects and shows good anti-inflammatory activity in inflammatory and autoimmune diseases. Nevertheless, it remains unclear how it affects acute organ rejection after allogeneic transplantation. In this study, a rat liver transplantation model was constructed to investigate the effect of LUT on acute rejection of organ allografts. We found that LUT significantly protected the structure and function of liver grafts, prolonged recipient rat survival, ameliorated T cell infiltration, and downregulated proinflammatory cytokines. Moreover, LUT inhibited the proliferation of CD4+ T cells and Th cell differentiation but increased the proportion of Tregs, which is the key to its immunosuppressive effect. In vitro, LUT also significantly inhibited CD4+ T cell proliferation and Th1 differentiation. There may be important implications for improving immunosuppressive regimens for organ transplantation as a result of this discovery.

Abstract:Aim of the study: To investigate the immunosuppressive effects of HPLC qualitied ethyl acetate extract (EAE) from Urtica dentate Hand oil skin allograft rejection ill a murine model.Materials and methods: Allo-skin transplantation model was established by placing skin allograft of C57BL/6 mice in the wound bed which was oil the back of Balb/c Mice. We used FACS to Study the effects of EAE oil dendritic cells (DCs) maturation and CD4(+)CD25(+)T regulatory cells (Tregs) differentiation. We also Studied spleen lymphocyte proliferation and T-bet gene expression in DCs. Concentration of Th1/Th2 cytokines was monitored as markers of Th1/Th2 responses by ELISA.Results: A significant prolongation of skin allografts Survival was observed as a dose-depenclent manner the animals treated with EAE. By FACS, we found that treatment with EAE (200 mg kg(-1)) resulted ill all immature statement of DCs and stimulated the differentiation of CD4(+)CD25(+)Tregs. Additionally, the expression of T-bet gene and the proliferation of spleen lymphocytes were efficiently abated in EAE treated mice. comparing to the model control, EAE-treated recipients showed a significant down-regulation (P<0.01) of Th1 cytokines (IL-2, IFN-gamma) and all Obviously increase (P<0.01) of Th2 cytokine (IL-10) ill the serum, which presented in a close-related way.Conclusions: The anti-allograft rejection effect of EAE by enhancing CD4(+)CD25(+)Tregs differentiation and sustaining DCs immaturation Makes EAE to be a possible choice for treating autoimmune diseases in a way of inducing a stable immunological tolerance state. Abstract:Objective To investigate effect of emodin on hepatic fibrosis in rats.Methods The rat hepatic fibrosis model was induced by the subcutaneous injection of 40% CCl4 (twice a week for 6 weeks) dissolved in olive oil. The emodin-treated rats were treated with low-dose, mediate-dose and high-dose emodin (20, 40 and 80 mg/kg body weight, once a day for 42 days) dissolved in 0.5% sodium carboxymethylcellulose ( CMC), except receiving CCl4. Control group received only olive oil and 0.5% CMC. Liver functions were determined by standard procedure. Serum hyaluronic acid and laminin were determined by radioimmunoassay. Liver hydroxyprolines were determined. Histopathological changes were examined by optical microscopy.Results Compared with model group, the emodin-treated rats showed (1) liver functions were improved, alanine transaminase (ALT) and alkaline phosphatase (AKP) were obviously reduced, and total protein (TP) and albumin (ALB) were significantly increased; (2) serum hyaluronic acid and laminin were markedly reduced; (3) liver hydroxyproline was significantly decreased; (4) the degrees of fibrosis were reduced. The changes of parameters mentioned above were significant (P<0.05 or P<0.01).Conclusion Emodin has effect on hepatic fibrosis in rats. The hepatoprotective of emodin may be one of mechanisms for liver fibrosis.
Costimulatory Pathway Blockade Prevents Acute Rejection in Rat Liver Transplantation

Abstract:Objective To investigate the effect of CD40Ig in acute rejection of rat liver transplantation.Methods Thirty-two cases of orthotopic liver transplantation were performed in Lewis to BN rats by using “the two-cuff technique”,and all rats were divided to 3 group.Group A (n=10) was control.Group B (n=11) and group C (n=11):Lipofectamine 2000-pcDNA3.1 or Lipofectamine 2000-pcDNA3.1 CD40Ig complex injected into Lewis portal vein ex vivo before the cold-storage stage of the liver.On the 5th day after transplantation,in each group 3 rats were killed to study the pathological changes and apoptosis,and to detect the CD40Ig expression by immunohistochemistry.The lymphocytes were obtained from spleen.The mixed lymphocyte reaction (MLR) were performed to determine the tolerance status and sheep anti-human IgG-FITC labled T cells were counted by flow cytometry.Postoperative survival time of the rats in each group was recorded.The pathological changes of the dead rats were observed.Results The mean survival time of group A and B were (11.00±4.28) and (12.75±5.57) days respectively.There have been serious acute rejection in allograft liver in group A and B and apoptosis index was 33.67±5.69 and 39.00±5.29 respectively.In group C,the mean survival time in group C was (41.25± 13.70) days (P0.01),the histochemistry revealed that CD40Ig was positive,the allograft liver had light acute rejection and apoptosis index was 0.27±0.21 (P0.01).The allograft livers of the dead rats have shown light acute rejection in part and chronic rejection mostly.MLR assay demonstrated that the Lewis mice in group C were specifically tolerant to the BN mice.The IgG-FITC labled T cells accounted for 11.57 % in all T cells of group C.Conclusion CD40Ig transfected liver could inhibit T cell costimulatory pathway,prevent acute rejection,and prolong survival.However,it could not impact the chronic rejection.

Abstract:Objective To study the effect of combined immune therapy with anti-CD40L mAb and low-dose cyclosporine(CsA) on the liver graft survival in rats,and Th1/Th2 cytokine profile.Methods Recipients were divided into the following groups.Group A: SD→SD rats,Group B: SD→Wistar rats without any treatment,Group C: SD→Wistar rats with CsA monotherapy from d1 to d5,Group D: SD→Wistar rats with CsA from d1 to d5 and anti-CD40L mAb in d0 and d2.The recipient survival and liver graft pathology observed.On post-transplant d7 ELISA technique was used to detect the level of cytokines in peripheral blood.Results The survival period of recipients in Group A and D was significantly longer than that in Group B and C.Mild to severe acute rejection reactions were observed in liver specimens from Group B and C while the degree of tissue injury in specimens from Group D was decreased signiflcantly.The serum levels of IL-2 and IFN-γ in Group B were significantly higher than those in other groups.On the contrary,the serum level of IL-4 and IL-10 in Group D was elevated much more than that in Group B(P0.05).Conclusions The combined immune therapy can prolong graft survival and effectively suppress acute rejection in ROLT.High expression of Th2 cytokines is beneficial to long-term survival of recipients and graft.
Effect of 1,25-Dihydroxyvitamin D3 on Preventing Allograft from Acute Rejection Following Rat Orthotoic Liver Transplantation

Abstract:Background We investigated the role of 1, 25-dihydroxyvitamin D3 (1, 25-(OH)2D3) in preventing allograft from acute rejection following orthotopic liver transplantation. Methods A rat orthotopic liver transplantation model was used in this study. SD-Wistar rats served as a high responder strain combination. Recipients were subjected to administration of 1, 25-(OH)2 D3 at dosages ranging from 0.25 μg·kg-1*d-1 to 2.5 μg·kg-1*d-1. Survival after transplantation as well as pathological rejection grades and IFN-γ mRNA, IL-10 mRNA transcription intragraft on day 7, and day 30 post-transplantation were observed. Results After recipients were treated with 1, 25(OH)2 D3 at dosages of 0.5 μg*kg-1*d-1 or 1.0 μ g*kg-1*d-1, survivals of recipients were prolonged. Ninety-five percent confidence intervals of survival were 46-87 days and 69-102 days (both P=0.0005 vs control group), respectively. On day seven post-transplantation, relative levels of IFN-γ mRNA transcription were 0.59±0.12 and 0.49±0.16, which was higher than the control group (P=0.005, P=0.003, respectively). Relative levels of IL-10 mRNA transcription were 0.83±0.09 and 0.76±0.09, which was lower than the control group (P=0.002, P=0.003, respectively). At a dosage of 0.5 μg·kg-1*d-1, the median of pathological rejection grade on day seven and on day thirty post-transplantation were 1.5 and 2.0 in comparison with the CsA-treated group (P=0.178, P=0.171, respectively). At a dosage of 0.5 μg·kg-1*d-1, the median of pathological rejection grade on day seven and day thirty post-transplantation were 1.5 and 1.5 in comparison with CsA-treated group (P=0.350, P=0.693, respectively).Conclusion After each recipient was treated with 1,25-(OH)2 D3 at a dosage of (0.5-1.0) μg·kg-1*d-1, transcription of cytokine intragraft was accommodated effectively and deviated to Th2 type, resulting in alleviation of acute rejection. 1, 25-(OH)2 D3 can prolong survival of recipient after orthotopic liver transplantation.

Abstract:Background . Postoperative intra-abdominal adhesions are a major complication after abdominal surgery. Although various methods have been used to prevent and treat adhesions, the effects have not been satisfactory. Emodin, a naturally occurring anthraquinone derivative and an active ingredient in traditional Chinese herbs, exhibits a variety of pharmacological effects. In our study, we demonstrated the effect of emodin treatment on preventing postoperative adhesion formation. Materials and Methods . A total of 48 rats were divided into six groups. Abdominal adhesions were created by abrasion of the cecum and its opposite abdominal wall. In the experimental groups, the rats were administered daily oral doses of emodin. On the seventh day after operation, the rats were euthanized, and blood and pathological specimens were collected. Abdominal adhesion formation was evaluated by necropsy, pathology, immunohistochemistry, Western blot, and enzyme-linked immunosorbent assay analyses. Results . Abdominal adhesions were markedly reduced by emodin treatment. Compared with the control group, collagen deposition was reduced and the peritoneal mesothelial completeness rate was higher in the emodin-treated groups. Emodin had anti-inflammatory effects, reduced oxidative stress, and promoted the movement of the intestinal tract ( P < 0.05 ). Conclusion . Emodin significantly reduced intra-abdominal adhesion formation in a rat model.

Abstract:Abstract Objective The aim of this study is to investigate the protective effects of emodin (EMD) on the lung injuries in the rat models of liver fibrosis. Methods Liver fibrosis was established in rats and the effect of intervention using EMD treatment was determined. Liver and lung weight coefficients were measured and lung content of TNF-α (tumor necrosis factor α), MDA (malondialdehyde), NO (nitric oxide), and ONOO- (peroxynitrite) were determined. Finally, histopathological changes were evaluated. Results Compared with the normal control group, the lung weight coefficient was significantly increased in the fibrosis model group. Moreover, pulmonary edema and inflammatory responses were observed. Levels of TNF-α, MDA, NO, and ONOO- in the lung homogenate were significantly increased in the fibrosis model group. After EMD treatment, the lung weight coefficients were significantly reduced. Moreover, pathological changes in the lung tissue were dramatically alleviated. Levels of TNF-α, MDA, NO, and ONOO- were significantly decreased. Conclusion EMD exhibits protective effects against lung injuries in a rat model of liver fibrosis.

Abstract:BACKGROUND AND AIM:Rejection remains a problem in the transplantation field. The aim of this study was to establish acute and chronic rejection models in rats and to investigate the roles of non-interleukin (IL)-2 T cell growth factors such as IL-15, IL-7 and IL-13 during rejection. METHODS:A liver transplant model was established using Dark Agouti and Brown Norway rats. The rats were divided into group A, left without treatment; group B, received cyclosporinee (1 mg/kg/day); and group C, cyclosporinee (4 mg/kg/day). Histopathological, reverse transcriptase-polymerase chain reaction and western blot were performed in liver specimens obtained from different time-points after transplantation in the three groups. RESULTS:In group A, the livers showed irreversible acute cellular rejection with cell infiltration. In group B, chronic liver rejection was found, with graft infiltration, ductular damage or proliferation, obliterative arteriopathy and liver fibrosis. No apparent histological alterations were observed in group C. IL-15, IL-7 and IL-13 messenger RNA and their protein were all highly expressed in the liver specimens of groups A and B. Upregulated expression was found in IL-15 since the first day after transplantation and in IL-7 and IL-13 since day 6. The extent of IL-15 upregulation was more than that of IL-7 and IL-13. CONCLUSIONS:Liver transplantation in Dark Agouti to Brown Norway rats with low-dose immunosuppression can induce chronic rejection. In the process of acute and chronic allograft rejections, non-IL-2 T cell growth factors such as IL-15, IL-7 and IL-13 play roles. Strategies should pay more attention to regulating these cytokines after liver transplantation.
