Abstract:The genomic DNA of Rhododendron was extracted by the revised CTAB method.The concentrations of Mg2+,dNTPs and primers which affected the SRAP-PCR reactions were optimized,and the SRAP molecular marker system in Rhododendron was established.The optimum system of 20 μl was as follows: template DNA 50 ng,10×PCR buffer(without Mg2+),Mg2+ 2.50 mmol / L,dNTPs 0.20 mmol / L,primers 0.40 μmol / L,Taq DNA polymerase 1.0 U.Ten genotypes of Rhododendron were amplified and detected under the optimum system.SRAP fingerprinting amplified by 16 pairs of primers revealed that 261 unambiguous bands were obtained,of which 251 sites were polymorphic,and the polymorphism frequency was 96.17%.The results indicated that the system was steady and reliable,which would be helpful to study molecular biology of Rhododendron.

Abstract:An orthogonal experiment L 16 ( 43 ) including three factors ( dNTP, primer and Taq DNA Polymerase ) and four levels was designed to optimize SRAP -PCR reaction system of Kenaf.Be-sides, we set up 8 concentration gradients of DNA template to test its influence on amplifying results.At last, we optimized the SRAP-PCR reaction system for Kenaf.It contained 2μL 10 ×Taq reaction buff-er, 20ng DNA template, 220μmol/L dNTP concentration, 0.35μmol/L primer concentration, 0.5U Taq DNA Polymerase , 20μL of the cumulative volume.In addition, 100 primer combinations were se-lected from 256 SRAP primer combinations due to their distinct Gershgorim bands and high polymor -phism.The optimal SRAP reaction system and 100 primer combinations would contribute to the basis for evaluating genetic diversity , identifing varieties , analyzing genetic relationship , building linkage map and QTL map for Hibiscus cannabinus L.

Random Amplified Polymorphic DNA (RAPD) is an inexpensive protocol that can provide information about the genetic diversity among Capsicum accessions within a germplasm bank, but it is highly sensitive to reaction conditions and manipulation. This work presents an optimized protocol for DNA isolation from dry Capsicum seeds, and a standardization of five conditions for PCR using RAPD primers. The RAPD protocol was optimized for the following parameters: 50 ng of DNA template per mix, 2.5 mM MgCl2, 200 μM dNTP's, 25 ng of primer per mix, and 45 thermal cycles, resulting in reproducible and clear amplified fragments. Good quality products were presented in the extractions and amplifications carried out; therefore, the optimized protocols are sufficient to be used in future work with RAPD.

Abstract:Randomly amplified polymorphic DNA (RAPD) markers quickly provide linkage information, especially in conifers where haploid megagametophytes can be used for genotyping. Traditionally use of slab gel electrophresis results in qualitative data that can be manually manipulated to gain semiquantitative information about the polymorphic RAPD fragments. We have proposed the use of an integrated microfluidic chip-based system as a new tool in the analysis of polymorphic RAPD fragments. The chip-based method was found to be very sensitive,requiring much less sample and only quarter the time compared to the agarose gel method. The automated data analysis sizes and quantitates the DNA fragments, thus yielding a more thorough,reproducible, sensitive, and rapid analysis.

Kitamura were also established.The optimal SRAP-PCR reaction system is as follows: total volume 20 μL,containing 3.125 mmol·L-1 Mg2+,187.5 μmol·L-1 dNTPs,10.0 μmol·L-1 primer,50 ng template DNA,0.5 U Taq DNA polymerase and 1×PCR buffer.The effects of five factors on amplification result of SRAP-PCR are different,in which,the effect of dNTPs concentration is the greatest,but that of Taq DNA polymerase amount is the least.The optimal SRAP-PCR reaction system is identified by means of genomic DNA from 'Aoyunhanxiao','Yuhualuoying' and their F1 progenies,and the amplification pattern with rich polymorphism and clear band is obtained.It is concluded that this SRAP-PCR reaction system used for genomic DNA from Dendranthema×grandiflorum is steady and reliable.

Abstract:In this paper the optimum RAPD reaction system of Spiraea was selected by experiment.It is the basis for preparation of genetic diversity and relatives of Spiraea germplasm resource study.An improved CTAB method was used to extract winter buds and tender leaf of ten Species of Spiraea DNA and factors which affect the RAPD amplification were optimized and selected.A single-factor optimization method used showed that a total volume of 20μl RAPD system of Spiraea consisted of 2μl 10×Taq buffer,1.0U Taq DNA polymerase,0.15mmol/L dNTP,2ng/μl DNA,0.2μmol/L primer and 2.0mmol/L Mg2+.The suitable PCR procedure is one cycle denaturing at 94℃ for 4min,40 cycles which involves denaturing at 94℃ for 45s,annealing at 37℃ for 1min and extending at 72℃for 1min,one cycle extending at 72℃ for 5min,and then kept at 4℃.The experiment shows that improved CTAB method could extracted winter buds of Spiraea DNA which could meet the requirement of RAPD reaction.The selected RAPD system amplified the polymorphism and its bands are clear.

Abstract:Using flue-cured tobacco cv.NC2326,G28 and their hybrid F-1 as materials,the effects of major(elements) in PCR reaction system on SSR analysis were studied.The results showed that under the conditions of(1.5U) TaqDNA polymerase, terminal MgCl-2 concentration of 1.5mmol·L~(-1),200μmol·L~(-1) dNTPs,0.25μmol·L~(-1) primers and 20ng templates of DNA in total volume of 20μL,the result was better in PCR reaction.

Abstract:Based on in vitro culture system of high capacity and fast regeneration of pepper ( Capsicum annuum L.)cotyledon. Pepper cultivars were transformed with double Cecropin B and D genes constructed in plasmid pCDB Ⅱ which was transferred into Agrobaterium tumefaciens . With the regenerated plants resistance to Kanamycin, the rate of transgenic pepper were surveyed and analysed on four pepper cultivars and three different differentiation groups of the same explant. The results showed that there was remarkable difference among the different pepper cultivars and there was no remarkable difference among the different differentiation groups on the detection of PCR level . No remarkable difference was found between the different pepper cultivars and between the different differentiation groups on the detection of Southern blotting level.

Abstract:Peppers are a major vegetable crop worldwide. With the completion of additional genome assemblies, a multitude of single-nucleotide polymorphisms (SNPs) can be utilized for population structure and genetic diversity analysis. In this study, we used target SNP-sequencing as a new high-throughput sequencing technology, screening out 425 perfect SNPs for analyzing the genetic diversity and population structure among 496 pepper lines from five pepper species in China and abroad. The perfect SNP panel exhibited commendable discriminative ability, as indicated by the average values of polymorphism information content, observed heterozygosity, minor allele frequency, and genetic diversity, which were 0.346, 0.011, 0.371, and 0.449, respectively. Based on phylogenetic, population structure, and principal component analyses, 484 C. annuum lines were divided into four subpopulations according to the shape of fruit: blocky fruit, wide-horn fruit, narrow-horn fruit, and linear fruit. These subpopulations displayed clear clustering with minimal or no overlap. Moreover, F statistic (Fst) analysis revealed considerable distinctions among these subpopulations. Additionally, we established a set of 47 core SNPs that could effectively differentiate among all pepper lines. This core SNP set could precisely classify the C. annuum lines into four distinct fruit-shape groups. The blocky and narrow-horn fruit subpopulations displayed the lowest and highest genetic diversity, respectively. This study highlights the importance of fruit shape as a crucial trait in pepper breeding. Moreover, this work indicates the immense potential of optimized target SNP technology in the addition of foreground markers of important traits to improve molecular breeding efficiency, and demonstrates its broad application prospects in the genetic analysis and variety identification of peppers.

Abstract:In order to obtain the optimization of ISSR-PCR reaction system of Phyllosticta citricarpa (McAlp-ine) van der Aa,the concentrations of five factors (Mg 2 +,dNTP,primers,Taq DNA polymerase,template) were re-spectively optimized by single factor test.The result showed that the optimized reaction system consisted of 1.5 mmol/L Mg 2 +,0.15 mmol/L dNTP,0.3 μmol/L primers,25 ng template DNA,0.5 U Taq DNA polymerase.The optimized ISSR-PCR reaction system was stable and credible by the amplification of 21 strains of P.citricarpa.The study would provide the basis for the genetic analysis of P.citricarpa.

Abstract:Template DNA used for ISSR-PCR was extracted from Dioscorea zingiberensis C.H.Wright leaf tissue.Through single factor experiment,the influencing factors on ISSR-PCR amplification,i.e.DNA quality and concentration,Mg2+ concentration,Taq polymerase dosage,dNTPs concentration and annealing temperature,were optimized.The ISSR system with optimal amplification parameters was established as following:15 μL reaction volume containing 20 ng/L DNA template,1×Taq polymerase buffer,1.0 U Taq polymerase,2.5 mmol/L MgCl2,0.2 μmol/L primer and 0.3 mmol/L dNTPs.With this system,ten of the 46 ISSR primers produced clear bands with high polymorphisms.Also,the optimal annealing temperatures for the primers were identified.This study laid a technological foundation for application of ISSR marker in analysis of D.zingiberensis germplasm resource.
RAPD analysis and comparison of main agronomic characters of Zuili (Prunus salicina Lindl. ) from different provenances

Abstract:The PCR amplification of total genomic DNA from eighteen provenances of Zuili (Prunus salicina Lindl. ) and three outgroups were researched by RAPD marker method, and on this basis, the genetic relationship of eighteen provenances was analyzed by cluster analysis. In addition, the comparative analysis of main agronomic characters of eighteen provenances was also conducted. RAPD analysis result shows that seventy-one bands are amplified with fourteen random primers selected from seventy random primers, in which there are twenty-nine polymorphic bands with a percentage of 40.8%. The cluster analysis result shows that eighteen provenances and three outgroups are divided into seven groups when Euclidean square distance is 0.060, in which each of three outgroups is an independent group, eighteen provenances of Zuili are divided into four groups. Each of two provenances including YXM and HW3 is an independent group, five provenances including HW2, WS, HP1, FB1 and FB2 are clustered in one group, other eleven provenances are clustered in one group. The peel colour of eighteen provenances of Zuili all is red, and the percentages of deep red and dark red each is 50%. According to maturing period of fruit, eighteen provenances of Zuili are approximately divided into early and late maturing type, maturing period is in mid and late June or early and mid July, respectively. By comparison, the early maturing type has some features including higher single fruit weight, lower soluble solid content and earlier flowering stage by 3-4 d. According to comprehensive analysis of RAPD marker and agronomic characters, it is suggested that these provenances of Zuili in Jiaxing area exert mutation and differentiation during cultivation process. YXM provenance has a close relationship with most of eighteen provenances, and also has some excellent features, so YXM is probably an excellent provenance and worth doing further research.

Abstract:METHODS: A retrospective review of patient charts and exercise attendance records was conducted to access the average weekly frequency of structured exercise and modulators of attendance over a 1-year period (July, 2014 through June, 2015) in a subset of long-term CR program participants. Exercise sessions were offered 5 days/wk, excluding weekends and holidays. Patients that discharged from the program temporarily (e.g., extended vacation) or who were non-compliant due to medical reasons, symptoms, or extended hospitalizations, were excluded. RESULTS: 122 cardiac patients (mean ± SD age = 73.2 ± 8.7 yrs, 84% men) comprised the study population. Most (77%) were not working and/or retired. Their mean ± SD body mass index (BMI), ejection fraction, and exercise training workloads were 28.0 ± 4.0 kg/m 2 , 55.5 ± 12.9%, and 5.4 ± 2.1 METs, respectively. This patient cohort (n = 122) attended < 2 sessions/wk (n = 41; 34%), ≥ 2 to 3 sessions/wk (n = 59; 48%), and ≥ 3 sessions/wk (n = 22; 18%). Age, BMI, ejection fraction, depression score, distance from the Center, and related co-morbid conditions, including diabetes and chronic obstructive pulmonary disease, were unrelated to attendance. In contrast, educational status was significantly related to attendance (p <0.03), with the highest percentage of advanced degrees being in the highest attending group (≥ 3 sessions/wk). The most compliant cohort also demonstrated higher training workloads (p <0.04) than their less compliant counterparts. CONCLUSION: Less than 1 in 5 long-term CR participants consistently attend ≥ 3 exercise training sessions per week. CR participants should be strongly encouraged to complement their medically-supervised training sessions with home training, increased lifestyle physical activity, or both.

Abstract:Random amplified polymorphic DNA(RAPD)was used to study the differencial genes between the Lentinula edodes(Qingyuan) strain 135 with poor resistance and strain 9015(a new strain by natural variation).Several differencial genes were obtained,and then cloned and sequenced.The results in GenBank revealed that the partially possible Open Reading Frames(ORF) of these fragments had higher homology with some hydrolases and pheromone precursor receptor genes of several kinds of organisms.And we presume these genes fragments could be as the specific molecular markers for specifying Lentinula edodes 135 and 9015.

Abstract:During the anglysis of the genetic diversity of Juglans mandshurica Maxim,to gain clear and repeatable profiles,the genomic DNA from Juglans mandshurica Maxim was extracted using the modified CTAB method.In order to establish an optimum ISSR-PCR amplification system on Juglans mandshurica Maxim,the authors carried out a single factor test and Orthogonal design test on the bases of four levels of five factors(Taq DNA polymerase,Mg~(2+),dNTP,DNA template,primer) respectively.Through comparing and analyzing,a more suitable ISSR-PCR reaction system was established;namely 20μX reaction system containing 1.0 U Taq DNA polymerase,2.0 mmol/L Mg~(2+),0.2 mmol/L dNTP,50～250 ng DNA template and 0.4μmol/L primer.13 primers with rich polymorphism and stable amplification for ISSR were screened.Besides,annealing temperature and cycles were also optimized.The establishment of the optimized system lays an important foundation for the future use of ISSR technology in researching the genetic diversity, relationship and protection of species of Juglans mandshurica Maxim.

Abstract:Pepper is an important condiment, and its aroma affects its commercial value. In this study, transcriptome sequencing and combined headspace solid-phase microextraction and gas chromatography–mass spectrometry (HS-SPME-GC-MS) were used to analyze the differentially expressed genes and volatile organic compounds in spicy and non-spicy pepper fruits. Compared with non-spicy fruits, there were 27 up-regulated volatile organic compounds (VOCs) and 3353 up-regulated genes (Up-DEGs) in spicy fruits. The results of KEGG enrichment analysis of the Up-DEGs combined with differential VOCs analysis showed that fatty acid biosynthesis and terpenoid biosynthesis may be the main metabolic pathways for aroma differences between non-spicy and spicy pepper fruits. The expression levels of the fatty acid biosynthesis-related genes FAD, LOX1, LOX5, HPL, and ADH and the key terpene synthesis gene TPS in spicy pepper fruits were significantly higher than those in non-spicy pepper fruits. The differential expression of these genes may be the reason for the different aroma. The results can provide reference for the development and utilization of high-aroma pepper germplasm resources and the breeding of new varieties.

Abstract:Introduction: Pepper ( Capsicum spp.) is a vegetable that is cultivated globally and has undergone extensive domestication, leading to a significant diversification in its agronomic traits. With the advancement of genomics in pepper and the reduction in sequencing costs, the high-throughput detection of single nucleotide polymorphisms (SNPs) and small insertions-deletions (indels) has become increasingly critical for analyzing pepper germplasms and improving breeding programs. As a result, there is a pressing need for a cost-effective, high-throughput, and versatile technique suitable for both foreground and background selection in pepper breeding. Methods: In the present study, Python-based web scraping scripts were utilized to systematically extract data from published literatures and relevant sequence databases focusing on pepper genomes. Subsequent to data extraction, SNPs and indels were meticulously identified and filtered. This process culminated in the delineation of core polymorphic sites, which were instrumental in the development of specific probes. Following this, comprehensive phenotypic and genotypic analyses were conducted on a diverse collection of 420 pepper germplasms. Concurrently, a genome-wide association study (GWAS) was conducted to elucidate the genetic determinants of helical fruit shape in peppers. Results: In this study, a 45K pepper Genotyping-By-Target-Sequencing (GBTS) liquid-phase gene chip was developed on the GenoBaits platform. This chip is composed of 45,389 probes, of which 42,535 are derived from core polymorphic sites (CPS) in the background genetic landscape, while 2,854 are associated with foreground agronomic traits, spanning across 43 traits. The CPS probes are spaced at an average interval of 68 Kb. We have assessed the performance of this chip on 420 pepper germplasms, with successful capture of target DNA fragments by 45,387 probes. Furthermore, the probe capture ratio surpassed 70% in 410 of the 420 germplasms tested. Using this chip, we have efficiently genotyped 273 germplasms for spiciness levels and elucidated the genetic relationships among 410 pepper germplasms. Our results allowed for precise clustering of sister lines and C. chinense germplasms. In addition, through a GWAS for helical fruit shape, we identified three quantitative trait loci (QTLs): heli2.1, heli11.1, and heli11.2. Within the heli11.1 QTL, a gene encoding the tubulin alpha chain was identified, suggesting its potential role in the helical growth pattern of pepper fruits. Discussion: In summary, the 45K pepper GBTS liquid-phase gene chip offers robust detection of polymorphic sites and is a promising tool for advancing research into pepper germplasm and the breeding of new pepper varieties.
