HMG Box-containing Protein 1 Transcriptionally Suppresses Promoter Activation of Macrophage Migration Inhibitory Factor

Abstract:HBP1(HMG box-containing protein 1) is a transcriptional repressor and a member of the sequence-specific HMG box family of transcription factors.It has been shown that the up-regulation of HBP1 inhibits the proliferation and metastasis of cancer cells,and the HBP1 gene lies within the 7q31 chromosomal region that is frequently deleted or translocated in cancers.Therefore,HBP1 functions as an anti-oncogene.Although the molecular mechanisms underlying HBP1 suppressing the growth of cancer cells remain poorly understood,it is undoubtedly that identification of HBP1 target gene will provide new clues for interpreting its functions.In this study,we found that the promoter of macrophage migration inhibitory factor(MIF) gene contains a large high-affinity HBP1 DNA-binding element at positions-811 bp to-792 bp from the transcriptional start site.Deletion and mutation analyses showed that HBP1 inhibited MIF promoter activity through this element.Furthermore,chromatin immunoprecipitation(ChIP) assays indicated that HBP1 bound this element in vivo and repressed the transcription of endogenous MIF.Moreover,the anti-tumorigenic effects of HBP1 were partially reversed in cells treated with purified recombinant MIF(rMIF).These results indicated that MIF is a target gene of HBP1.HBP1 inhibition of cancer cells growth is achieved at least partially by suppressing MIF expression.
