Studies on Adverse Effects of Soy Isoflavones on Reproductive System of Female Rats at Development Stages

Abstract:The purpose of this study was to explore the effects of fetal,neonatal,infant and prepubertal exposure to soy isoflavones(SIF)on female reproductive system,and identify the most sensitive development period and the dose level of SIF to cause adverse effects on reproductive system.In the fetal exposure study,SIF was adiministered to pregnant rats by gavage during gestation day(GD)12~18.In the neonatal and infant exposure studies,SIF was adiministered to maternal rats by gavage during postnatal day(PND)5~10 and PND 15~20,respectively.And in the prepubertal exposure study,SIF was adiministered directly to the offspring by gavage during PND 25~30.The test levels of SIF were 0(control),10,50,100,150 and 200 mg/kg BW.Some indicators related to the development of female reproductive system were measured after SIF treatment.The following facts were found:(1)In the animals of fetal exposure,compared with those in the control group,the anogenital distance(AGD)on PND 1 in 100,150 and 200 mg/kg BW groups was increased,the vaginal opening time(VO)in 200 mg/kg BW group was advanced and the ratio of ovary to body weight in 150 and 200 mg/kg BW groups was significantly higher on PND 14.(2)On PND 11,in the animals of neonatal exposure,the ratios of ovary to body weight and uterine to body weight in 150 and 200 mg/kg BW groups were higher and estrus cycle was prolonged,significantly when compared with those of the animals in the control group.(3)On PND 21,in the animals of infant exposure and on PND 31 in the animals of prepubertal exposure,SIF had no significant effect on the indicators mentioned.And on PND 70,in animals of all the 4 modes of exposure(fetal,neonatal,infant and prepubertal),no influence on these indicators was observed.It is concluded that SIF in the exposure level of 150 mg/kg BW was sufficient to induce significant adverse effects to female reproductive development.And fetal stage and neonatal stage in life were,at least,more sensitive to SIF exposure.
