Abstract:Toosendansins E-I (1-5), five new limonoids together with nine known limonids(6-), were isolated and identified from the fruits of Melia toosendan. Their skeletons were belonged to meliacarpins (1 and 2), nimbin (3), and vilasinins (4 and 5). All the isolates were identified using 1D & 2D NMR spectroscopic experiments, and the absolute configurations of 1 and 2 with 1,3-dicinnamoyl moieties were achieved by CD method. Compounds 1 and 4 showed moderate inhibitory activity against osteosarcoma cell line U-2 OS, and compound 4, ohchinolal (12), meliatoxin B1 (13) and 1,12-diacetyltrichichilin B (14) showed obvious reversal activity of multidrug resistance in MCF-7 cell line at 6.25 mu M.

Abstract:In the present study, two new limonoids, 1α, 7α-dihydroxyl-3α-acetoxyl-12α-ethoxylnimbolinin (1) and 1α-tigloyloxy-3α-acetoxyl-7α-hydroxyl-12β-ethoxylnimbolinin (2), together with other four known limonoids (3-6), were isolated from the fruits of Melia toosendan. Their structures were elucidated by means of extensive spectroscopic analyses (NMR and ESI-MS) and comparisons with the data reported in the literature. The isolated compounds were evaluated for their antibacterial activities. Compound 4 exhibited significant antibacterial activity against an oral pathogen, Porphyromonas gingivalis ATCC 33277, with an MIC value of 15.2 μg·mL(-1). Compound 2 was also active against P. gingivalis ATCC 33277, with an MIC value of 31.25 μg·mL(-1). In conlcusion, our resutls indicate that these compounds may provide a basis for future development of novel antibiotics.

Abstract:Four nimbolinin-type limonoids, 12α/β-1-O-tigloyl-1-O-deacetyl-nimbolinin B (1), 1-deacetylnimbolinin B (2), nimbolinin B (3) and nimbolinin A (4), were isolated from the fruits of Melia toosendan. 1 was a new compound and existed as a mixture of a pair of tautomers, 12α- (1a) and 12β- (1b). The structures of both tautomers were fully determined by extensive spectroscopic methods including UV, IR, NMR and ESI-MS. Tautomeric behaviors and their relative molar ratios in compounds 1–4 were further investigated using optical rotation, TLC, 1H NMR and HPLC. Equilibrium equation of nimbolinin was proposed accordingly, with 12α- and 12β-isomers interchanging via a 12-hemiacetal intermediate.

Abstract:Two new furan fragment isomerized limonoids, meliazedalides A and B (compounds 1 and 2), were isolated from the fruits of Melia azedarach Linn.. Their chemical structures were elucidated on the basis of HR-ESI-MS and 1D and 2D NMR data, which belonged to nimbolinin- and trichilin-class, respectively. Compound 2 exhibited weak inhibitory effect on NO production in lipopolysaccharide (LPS)-activated RAW 264.7 macrophages with IC50 being 37.41 μmol·L−1.

Abstract:Four new limonoids (1–4), together with five known limonoids (5–9), were isolated from the fruits of Melia toosendan. Their structures were elucidated based on extensive spectroscopic analyses (1D- and 2D-NMR, HRESIMS, IR, [α]D). The isolated compounds were evaluated for their neurite outgrowth-promoting activities. Abstract:In this study five new limonoids, toosendone [24,25,26,27-tetra-nor-6alpha-acetoxy-21,22-epoxy-7alpha-tigloyl-1alpha,3alpha,28-trihydroxyapotirucalla-(apoeupha)-14,20,22-trien-12-one, 1] and 12-ethoxynimbolinins A-D (2-5), together with five known limonoids, 1-acetyltrichilinin (6), 1-cinnamoyltrichilinin (7), trichilinin B (8), 1,7-di-O-acetyl-14,15-deoxyhavanensin (9) and 12-O-methylnimbolinin B (10),were isolated from the fruits of Melia toosendan. Their structures and relative configurations were established based on spectroscopic analysis. Compound 4 exhibited significant antibacterial activity against the oral pathogen, Porphyromonas gingivalis ATCC 33 277, with an MIC value of 15.6 microg/mL. Compounds 7 and 8 were also active against P. gingivalis ATCC 33 277, with MIC values of 31.3 and 31.5 microg/mL respectively.

Abstract:Limonoids are a kind of bioactive triterpenoids and mainly exist in the Meliaceae and Rutaceae plants. In this research, two new nimbolinin-type limonoids (1 and 2), one new trichilin-type limonoid (3), and one new havanensin-type limonoid (4), along with 11 known nimbolinin-type limonoids (5–15) were obtained from the fruits of Melia toosendan. Their structures were elucidated by a combination of high-resolution ESI-MS, 1D and 2D NMR spectra. Nimbolinin-type limonoid 11, 13, and 8 showed obvious effects on autophagy, as they significantly induced the accumulation of autophagosome and enhanced the lysosomal activity. A preliminary structure-activity relationship study suggested that substitution at C-12 and C-1 both have an evident influence on the activity. To the best of our knowledge, this is the first report on the autophagic activity of nimbolinin-type limonoids.

Abstract:Toosendanone A (1), a new euphane (tirucallane)-type triterpene bearing a five-membered ring in the side chain and the first cyclopentanyl protolimonoid, was isolated from the bark of Melia toosendan, along with two new tirucallanes, toosendanic acids A (2) and B (3). The structure and absolute configuration of compound 1 was elucidated by spectroscopic data interpretation and X-ray diffraction analysis. Compounds 1-3 were evaluated for cytotoxicity against a small panel of cancer cell lines.

Abstract:Nine new limonoids, meliazedarines A–I (1–9), seven known analogues (10–16), and five known triterpenoids (17–21) were isolated from the fruits of Melia azedarach. Abstract:INTRODUCTION Melia toosendan Sieb. et Zucc. has been used as a Chinese folk medicine for roundworm treatment since ancient times. Many diverse limonoids have been isolated from Meliaceae plants, but it remains difficult to isolate and identify other limonoids because of their small natural concentrations. OBJECTIVE This study was performed to overcome the difficulties associated with fast and accurate identification of limonoids and establish a reliable and sensitive method for the analysis of minor limonoids in M. toosendan fruits. METHODS An efficient strategy for enrichment, detection, and identification of minor limonoids from M. toosendan fruits using solid-phase extraction with high-performance liquid chromatography-quadrupole time-of-flight tandem mass spectrometry (SPE-HPLC-Q-TOF-MS/MS) was developed herein. RESULTS Characteristic fragmentations and fragmentation ions containing trichilin-, nimbin-, and vilasinin-class limonoid skeletons were initially studied, and characteristic diagnostic ions involved retro Diels-Alder (RDA) reactions or homolytic cleavages, which were used to identify minor limonoids. In total, 13 limonoids, including four new ones, were identified. CONCLUSION This is the first report on the analysis of M. toosendan fruits to identify limonoids. This novel analysis method may stimulate further research regarding the identification of limonoids in other plant species.

Abstract:Four new limonoids, tociliatonoid B-E (1–4) were obtained from the EtOAc extract of the twigs and leaves of Toona ciliata var. yunnanensis. Their structures were elucidated through comprehensive spectroscopic and X-ray crystallographic analysis. Abstract:Four previously unreported tirucallane-type triterpenoids ( 1–4 ), together with four known analogues ( 5–8 ), were isolated from the fruits of Melia toosendan Sieb. et Zucc. Their planar structures were comprehensively elucidated by detailed analyses of HRESIMS, 1D and 2D NMR spectra data. The relative configurations of 1–4 were determined by NOESY experiments. The comparison of experimental and calculated electronic circular dichroism (ECD) spectra led to the establishment of the absolute configurations of new compounds. All isolated triterpenoids were evaluated for their α -glucosidase inhibitory activities in vitro . Compounds 4 and 5 showed moderate α -glucosidase inhibitory activities with IC 50 values of 120.3 ± 5.8 and 104.9 ± 7.1 μM, respectively.

Abstract:The phytochemical study of the fruits of Melia azedarach (Meliaceae) led to the isolation and characterisation of two novel natural limonoids1-deoxy- 3, 20-dicinnamoyl-11-methoxy-meliacarpinin ( 1 ) and 12 β - O- methyl nimbolinin A ( 2 ), along with twelve known limonoids. Its structure was identified by 1D- and 2D-NMR, HR-ESI-MS and comparison with published data. The anti-inflammatory effect of the compounds was measured in vitro in RAW 264.7 cells by evaluating the production of NO stimulated by LPS. Compounds 1, 8 and 14 indicated significant anti-inflammatory effect with inhibition rate of 11.76, 8.45 and 6.59 μM, respectively. Limonoid 1 significantly inhibited the production of NO, TNF- α and IL-1 β in RAW 264.7 cells. Therefore, limonoid derivative may be a promising source of bioactive metabolite for inflammatory diseases.

Abstract:OBJECTIVE To investigate the chemical constituents of Melia toosendan.METHODS The chemical constituents were isolated by various column chromatographic methods and structurally elucidated by spectral evidences.RESULTS Sixteen compounds were obtained and identified as epipinoresinol(1),clemaphenol A(2),medioresinol(3),(±)balanophonin(4),evofolin- B(5),quercetin(6),isoquercitrin(7),rutin(8),ferulic acid(9),p-hydroxybenzaldehyde(10),coniferylaldehyde(11),syringic acid (12),isovanillic acid(13),p-hydroxybenzoic acid(14),5-hydroxymethyl-fuffural(15)and cirsiumaldehyde(16).CONCLUSION Compound 6 was firstly isolated from this plant while other compounds were isolated from this genus for the first time.
