Small interference RNA inhibits the endogenous expression of beta-site APP cleaving enzyme-1 protein in neuro-2a cells

Abstract:AIM: To investigate the inhibitory effect of small interference RNA (siRNA) on the expression of beta-site APP cleaving enzyme-1 protein (BACE1) in neuro-2a cells. METHODS: The experiment was conducted in Zhongshan School of Medicine, Sun Yat-sen University and College of Agriculture and Biology, Shanghai Jiao Tong University between December 2004 and June 2006. ① Neuro-2a cells were simultaneously or respectively transfected with enhanced green fluorescent protein (EGFP), pEGFP-C1 Vector and siEGFP by Lipofectamine 2000, and the expression ratios of EGFP in neuro-2a cells were calculated under inverse fluorescent microscope. ② Then, siBACE1-1, siBACE1-2 and siBACE1-3, which were synthesized by transcription in vitro, were used to transfect neuro-2a cells, respectively. After 24, 48 and 72 hours, the inhibition ratio of BACE1 gene and time-effect of siBACE1 interference were determined by real time RT-PCR. RESULTS: ① 43% neuro-2a cells expressed highly EGFP after transfected with exogenous EGFP gene. siEGFP transfection could effectively inhibit the exogenous EGFP gene expressions of neuro-2a cells. ② Three sites of siBACE1 had distinct suppression efficiency for the inhibition of BACE1 gene. The levels of BACE1 mRNA decreased by 60% by siBACE1-3, and 13% by siBACE1-1, respectively. There was no apparent decrease by siBACE1-2. ③ The inhibition ratio of BACE1 was correlated with interfering time. There were no statistic difference in BACE1 mRNA expression between the siBACE1 after transfected for 24 and 48 hours and the control group (P > 0.05), however, most of BACE1 mRNA was decreased in neuro-2a cells after transfected by siBACE1-3 and siBACE1-1 for 72 hours. CONCLUSION: Expression of endogenous BACE1 gene in neuro-2a cells is effectively inhibited by siBACE1s, which are synthesized by transcription in vitro. The inhibition ratio is related to interfering sites of BACE1 and interfering time.
