Mutagensis in combination with In vitro selection of black pepper(Piper nigrum Linn.) somaclones resistant to foot rot

Abstract:Based on the in vitro shoot-tip multiplication technique, application of in vitro selection in combination with EMS (ethyl methanesulphonate) mutagensis in selection of black pepper somaclones resistant to foot rot (or quick wilt) disease caused by the fungus Phytophthora capsici [Phytophthora palmivora var. piperis or Phytophthora palmivora (Butl.) Butler MF4]was carried out with a large-leaf variety of Piper nigrum Linn., Lampong Type, widely cultivated in Hainan but highly susceptible to P. capsici, used as explant resources. The results showed that the frequency of plants resistant to P. capsici was increased with the EMS treatment of shoot tips before inoculation. The frequencies of plants resistant to P. capsici were between 4.3 % and 26.67 % with different EMS concentrations and different durations of EMS treatment of the shoot tips but without in vitro selection. In contrast, there was no plant that had disease resistance found in the control plant population. The EMS mutagensis in combination with in vitro selection with the culture filtrate of P. capsici further enhanced the efficiency and effectiveness of selection procedure compared with either EMS pretreament or in vitro selection, resulting in selection of 33.33 %~100 % disease-resistant somaclones. Of these treatment groups, combination of 1.0 % EMS treatment for 60 min with in vitro selection with 25 % culture filtrate of P. capsici was found optimum.
