Effect of Prior Physical Exercise on Glial Cell Line-Derived Neurotrophic Factor During MPTP-Induced Parkinson's Disease in Mice

Abstract:Objective: The present work preliminarily study the effects of prior physical exercise on mitochondrial reactive oxygen species(ROS) generation,glial cell line-derived neurotrophic factor(GDNF) content changes in midbrain and striatum of Parkinson's(PD) mouse model.Methods: 60 male C57BL/6(6-8weeks) mice were randomly divided into sedentary or exercise group(12m/min,20 minutes/day,running on treadmill for 6 weeks).Each group were divided into two groups again,and were injected with moderate dose of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP) or saline.Finally mice were divided into the following groups:(1) saline injected(N);(2) pre-exercise and saline injected(EN);(3) MPTP injected(P);(4) pre-exercise and MPTP injected(EP).ROS generation rate in isolated midbrain brain and striatum mitochondria were detected.GDNF mRNA and GDNF content were measured by Real-time PCR and ELISA respectively.GDNF content in blood was measured simultaneously.Results: 1) compared with the N group,mitochondria ROS generation rate,GDNF mRNA expression and protein were significantly increased in midbrain and striatum of P group mice;2) compared with P group,GDNF mRNA expression and protein were significantly increased in midbrain and striatum mice in EP group whose ROS generation rate was declined;GDNF in blood was significantly increased.Conclusion: MPTP could increase mitochondrial ROS generation rate in the midbrain and striatum,which could increase GDNF expression and secretion.Pre-exercise training could promote the secretion of GDNF which reduced ROS generation and protected mouse brain against MPTP-induced brain injury.
