Expression of cyclooxygenase-2 in nonalcoholic fatty liver disease and its significance

Abstract:AIM: To explore the expression of cyclooxygenase2(COX-2) in nonalcoholic fatty liver disease(NAFLD) and its significance.METHODS: The rat NAFLD model was established by giving a fat-riched diet.The expression level of COX-2 in rat livers was assayed by immunohistochemistry,RT-PCR and Western-blot.RESULTS: Light microscopy indicated that the liver injury was more obvious in the model group than in the treatment group.The levels of serum aspartate aminotransferase(AST),alanine aminotransferase(ALT),thromboxane B2(TXB2) and interleukin-1(IL-1) in the fatty liver rats were increased as compared with those in the normal controls(P<0.05).Compared to model group,levels of IL-1 and TXB2 decreased significantly in treatment group(P<0.05).The 6-Keto-prostagland(6-k-PGF1α)in hepatic tissue was lower in model group than in control group,while higher in treatment group than in model group(P<0.05).COX-2 mRNA was positive in model group and higher than in control group(P<0.05).The level of COX-2 protein was obviously higher in model group than in control group(P<0.05),but decreased significantly after Celecoxib treatment(P<0.05).CONCLUSION: COX-2 promotes the development of fatty liver by mediating lipid peroxidation and lipopolysaccharide.Cyclooxygenase-2 inhibitor alleviates the liver injury in NAFLD by downregulating the expression of COX-2 and depressing the oxidative stress.
