Abstract:The 15-day intact adult male assay was used to evaluate effects of isoflurane on the testes and sexual hormones. Forty adult male Sprague-Dawley rats were divided into five groups exposed to air containing 0, 50, 300, 1800 or 10,800 ppm isoflurane. After the treatments, serum was collected for the hormones assay. The right testis was to be used for daily sperm production. The left testis was processed for histopathology and electron microscopy observation. Daily sperm productions were significantly decreased at doses of 300, 1800 and 10,800 ppm. Impaired seminiferous tubules were noted at doses of 300, 1800 and 10,800 ppm. Ultrastructural changes included nucleus agglutination of spermatocytes, big lipid drops and autophagosome in cytoplasm. The serum follicle-stimulating hormone and testosterone concentrations reduced significantly at doses of 1800 and 10,800 ppm. Isoflurane induced impairments of seminiferous tubules and spermatogenesis. The testicular damages caused by isoflurane can be related to the imbalances in the sexual hormones.
Reproductive performance in SD rats and offspring growth and development

Abstract:[Objective]To statistically analyze the reproductive toxicity data collected from laboratory of Zhejiang Academy of Medical Sciences,to study the characteristics of SD rat reproductive performance,to analyze the growth of newborn mice prior to weaning,and compare differences between male and female offspring in the growth and development. [Methods]Statistical analysis was performed on the historical data that were from reproduction toxicity test comparison group of two generation in 8 batch. Observation included pregnancy situation,litter situation,litter weight,litter size,weight of pups that prior to weaning. The Excel software was used to analyze the related data statistically. [Results] The study showed SD rat pregnancy period was 22-25 days and mating behavior distributed in the first estrous cycle. Mating success rate was 94. 27%( 87. 50%-100. 00%),pregnancy rate was 92. 19%( 87. 50%-100. 00%),live birth rate was 98. 81%( 95. 24%-100. 00%),birth survival rate was 94. 67%( 87. 78%-99. 14%),feeding survival rate was 95. 85%( 89. 74-98. 82%). Born 0d,4d,7d,14 d,21 d average weight of pups were( 6. 54 ± 0. 64) g,( 10. 66 ± 1. 72) g,( 16. 74 ± 2. 42) g,( 32. 93 ± 5. 28) g,( 51. 64 ± 6. 59) g,the average weight of female offspring were( 6. 40 ± 0. 58) g,( 10. 48 ± 1. 62) g,( 16. 47 ± 2. 32) g,( 32. 41 ± 4. 02) g,( 50. 72 ± 6. 49) g,the average weight of male pups were( 6. 68 ± 0. 70) g,( 10. 86 ± 1. 84) g,( 17. 05 ± 2. 55) g,( 33. 05 ± 4. 37) g,( 52. 59 ± 7. 05) g,the difference between them was statistically significant( P 0. 05). [Conclusion]The selected SD rats in this study have high reproductive index,and they are suitable for reproductive toxicity test. The study finds that the body weight of lactating female and male offspring are significantly different. The statistical analysis of male and female offspring body weight and other relevant parameters separately can better reflect the reproductive toxicity study in rats.

Abstract:Objectives: The purpose of this study was to explore the effects of inhalational anaesthetics on the male reproductive system in neonatal rats.Methods: Postnatal day 6 (P6) male rat pups were assigned to 2 groups of 40 rats each. The control group did not receive anaesthesia, and the test group was exposed to air containing 3% sevoflurane in a chamber. After sevoflurane exposure, the effects on the male reproductive system in neonatal rats exposed to inhalational anaesthetics were tested. The rats were maintained to P60. The fertility, sperm and spermatid count, sperm motility, organ weights and histological changes in rats were determined.Results: Compared with those of the control group, the sperm count and motility of the sevoflurane group were significantly decreased (p < 0.05), and there was also a decrease in the number of foetal rats from the sevoflurane group (p < 0.05). Furthermore, the arrangement of the seminiferous tubule was regular in the control group, whereas the arrangement of the seminiferous tubule was distorted and the spermatocytes were detached and irregularly lined in the sevoflurane group. TUNEL analysis showed that the number of apoptotic cells in the testes of the sevoflurane group was significantly increased (p < 0.05). SOD and MDA analyses showed that SOD was decreased, while MDA expression was increased.Conclusions: These data indicated that young rats suffered as a result of inhalation of the anaesthetic sevoflurane and had reduced reproductive system function during adolescence in males. These studies are useful as a foundation for clinical studies.

Abstract:OBJECTIVE:To study the effects of inhaled anesthetics on human sperm motility in vitro.METHODS:Sperm samples were obtained from 20 healthy men by masturbation and prepared by the swim-up technique. The effects of isoflurane and sevoflurane at the clinical concentration (1.4%-5.6%) and high concentration (5.6%-84%) on human sperm motility in vitro were observed at 25 degrees C by the computer-assisted sperm analysis (CASA).RESULTS:The sperm vitality and motility were significantly increased on 0.5-4 h exposure to isoflurane at the clinical concentration and decreased gradually at high concentration (42%-84%). The effect of isoflurane on human sperm motility and vitality at the clinical concentration was reversible when the anesthetic withdrawn. Sevoflurane had no effects on human sperm motility and vitality at either the clinical or high concentration.CONCLUSION:Isoflurane has a reversible increasing effect at the clinical concentration and a significant decreasing effect at the high concentration on the motility and vitality of human sperm, while sevoflurane does not affect human sperm motility and vitality at either concentration.

Abstract:Fluorinated hydrocarbons as inhalation anaesthetics were widely used in surgery.The toxicity studies of inhalation anaesthetics were localized mainly on hepatotoxicity,nephrotoxicity and cardiac vascular inhibition.By contrast,harmful reproductive effects of inhalation anaesthetics were not sufficiently documented.The recent studies show that fluorinated hydrocarbons are capable to induce cytotoxic and genotoxicity,but the mechanism of such effects are unknown.

Abstract:Reproductive issues are becoming an increasing global problem. There is increasing interest in the relationship between microbiota and reproductive health. Stable microbiota communities exist in the gut, reproductive tract, uterus, testes, and semen. Various effects (e.g., epigenetic modifications, nervous system, metabolism) of dysbiosis in the microbiota can impair gamete quality; interfere with zygote formation, embryo implantation, and embryo development; and increase disease susceptibility, thus adversely impacting reproductive capacity and pregnancy. The maintenance of a healthy microbiota can protect the host from pathogens, increase reproductive potential, and reduce the rates of adverse pregnancy outcomes. In conclusion, this review discusses microbiota in the male and female reproductive systems of multiple animal species. It explores the effects and mechanisms of microbiota on reproduction, factors that influence microbiota composition, and applications of microbiota in reproductive disorder treatment and detection. The findings support novel approaches for managing reproductive diseases through microbiota improvement and monitoring. In addition, it will stimulate further systematic explorations of microbiota-mediated effects on reproduction.

This review mainly introduces the effects of ambient air pollution on human reproductive health such as low birth weight, preterm birth, birth defects, and so on, and simply explore the possible risk factors and the possible functional mechanisms. Based on that, it can be used to provide theory basis for policy-decision makers to draw macroscopic regulative measures in order to reduce the incidence of adverse pregnancy outcomes.
Effect of Acrylonitrile on Male Reproductive Health Among Occupationally Exposed Workers

Abstract:Objective To explore the effects of acrylonitrile(ACN) on reproductive function in male workers. Methods 275 male workers exposed to ACN and 229 male control workers were investigated by retrospective questionnaires. Results The incidence of abnormal pregnancy outcome in wives of male workers exposed to ACN was significantly higher than that in control group. Relative risk(RR) of prolonged childbearing, barrenness and spontaneous abortion in the wives of workers exposed to ACN were 3.65(95%CI:1.53 ~ 8.69), 2.22(95%CI:1.18 ~ 5.58) and 1.51(95%CI:1.14 ~ 3.07) respectively. The incidences of barrenness and spontaneous abortion in the wives of male workers working in lower-concentration workshop were significantly lower than whom in other three higher concentration workshops(P0.05). Though incidence of stillbirth among 4 workshops had no significant difference, but all the stillbirth occurred in the higher concentration workshops. The incidence of spontaneous abortion in the group of both the couples exposed to ACN was significantly higher than that in the group of only male workers exposed(RR=2.33, P0.05). The rate of self-report about sexual functional disorder in male worker exposed to ACN was significantly higher than that in male non-exposed workers(RR=2.43, 95%CI:1.11 ~ 5.32). Conclusion ACN is potential harmful to reproduction, and may induce adverse reproductive outcome. It also indicated that ACN may cause sexual functional disorder in male exposed workers.

Abstract:Numerous studies have identified specific areas in which the effects of maternal smoking during pregnancy may occur including fetal growth retardation, neonatal deaths, pregnancy complications leading to premature delivery and possible effects on lactation and long-term effects on surviving children. Despite the number of papers which have been published on this subject there is still no clear understanding of the extent to which smoking in general, and various components of cigarette smoke in particular, exert a detrimental effect on the human reproductive potential. Although epidemiological evidence is scanty there are indications that cigarette smoking decreases fertility in women, increases the frequency of menstrual abnormalities and decreases the age of spontaneous menopause. Cigarette smoking in males has also been implicated as a cause of decreased sperm numbers and an increased frequency of abnormal sperm morphology as well as a decrease in sexual performance. Experimental evidence from humans and rodents suggests that nicotine can alter the hypothalamic-pituitary axis through its stimulation of growth hormone, cortisol, vasopressin and oxytocin release which in turn inhibit luteinizing hormone and prolactin release.

Abstract:Infertility and subfertility problems are important health issues.Environmental risk factors have the possibility of affecting reproductive process,causing subfertility and abnormal pregnancy outcome.This paper is an overview of important environmental factors on infertility.Heavy metal such as lead and cadmium,organic pollutants such as polycyclic aromatic hydrocarbons and physical factors such as heat exposure are commonly known reproductive risk factors,but the influence of many other environmental pollution exposure on human fertility were not very conclusive.Scientific research methods and modern molecular biology techniques are needed in the study of environmental factors and human infertility.

Abstract:The high altitude area is characterized by low pressure and hypoxia, and rapidly entering the high altitude area will cause a series of damage to the body. Some studies have shown that hypoxia can cause damage to the reproductive system. In recent years, researchers have been paying attention to the effects of hypoxia on hormone level, ovarian reserve, embryonic development, testicular development, sperm motility level, and have begun to explore its injury mechanism, but its mechanism is not clear. In this paper, the mechanism of hypoxia on the reproductive system is reviewed, which is expected to provide a new idea for solving the problem of the low fertility rate of humans and animals at high altitudes.

Abstract:Exposure to air pollutants is known to be an important risk factor in reducing semen quality in men across the world. Poor semen quality results in decline in the global fertility rate and significant personal stress, dysfunctional sexual relationships, and psychosocial problems. Continuous monitoring and effective efforts to reduce air pollution in industries and the environment and making positive changes in daily lifestyle can prevent adverse effects on semen quality and reduce the high prevalence of men infertility. This review aims to summarize studies associating pollutant concentrations of polycyclic aromatic hydrocarbons (PAHs), formaldehyde (FA), and BTEX (benzene, toluene, ethyl-benzene, and xylene) on semen quality. In this systematic review, Scopus, PubMed and Web of Science databases were searched until November 13, 2022. The PECO statement was formulated to clarify the research question, and articles that did not satisfy the criteria outlined in this statement were excluded. Generally, 497 articles were obtained through searching databases, and after the investigations, 26 articles that met the entry criteria were extracted and finally considered in the systematic review. The results showed that occupational and environmental exposures to PAHs, formaldehyde, and BTEX were associated with increased metabolite concentration of toxic pollutants in body fluids. These toxin-associated pollutants directly or indirectly cause detrimental effects on sperm motility, vitality, DNA fragmentation, and morphology. There is evidence on the impact of PAHs, formaldehyde, and BTEX pollutants on the reduction of semen quality. Therefore, proving the relationship between air pollutants and testicular function in semen quality can play an effective role in macro policies and adopting stricter laws to reduce the emission of air pollutants and promote a healthy lifestyle to improve reproductive health in young men.

Abstract:Background As the population ages,elderly patients requiring surgery are increasing.The impact of anesthetic management on the outcome of elderly surgical patients deserves more attention.Objective The review focuses on the effects of inhalational versus intravenous anesthetics on the outcome of elderly surgical patients.Content For patients undergoing cardiac surgery,inhalational anesthesia may have more advantages than propofol intravenous anesthesia,but this effect requires further confirmation by multicenter,large sample size,randomized controlled trials.In terms of short-term postoperative brain function,current results are conflicting and no clear conclusion can be reached.With regard to long-term postoperative brain function,retrospective studies showed that general anesthesia may be associated with increased risk of dementia,but this need to be confirmed by prospective studies with long-term follow-up.For patients with malignant tumor,propofol intravenous anesthesia may be superior to inhalational anesthesia in the aspect of immune function after surgery,but the clinical significance of this effect is not totally clear.The effects of anesthetics on tumor invasiveness and long-term outcome of patients with malignant tumor deserve further study.Trend The effects of inhalational versus intravenous anesthetics on the outcome of elderly surgical patients deserve further studies.
Male reproductive system and simulated high-altitude environment: preliminary results in rats

Abstract:This study assessed the effects of a simulated high-altitude environment on the reproductive system of prepubertal male rats and the reversibility of these effects upon return to a normal environment. Three-week-old male Wistar rats were randomly allocated to 4 groups that were exposed to different conditions: a normal environment for 6 weeks and 12 weeks, respectively, hypobaric hypoxia for 6 weeks, and hypobaric hypoxia for 6 weeks followed by a normal environment for 6 weeks. Multiple pathophysiological parameters were evaluated at the histological, endocrine, and molecular levels. Hypobaric hypoxia exposure for 6 weeks during the prepubertal phase significantly altered physiological parameters, body functions, blood indices, and reproductive potential. Six weeks after returning to a normal environment, the damaged reproductive functions partially recovered due to compensatory mechanisms. However, several changes were not reversed after returning to a normal environment for 6 weeks, including disorders of body development and metabolism, increased red blood cells, increased fasting blood glucose, abnormal blood lipid metabolism, decreased testicular and epididymis weights, abnormal reproductive hormone levels, excessive apoptosis of reproductive cells, and decreased sperm concentration. In summary, a hypobaric hypoxic environment significantly impaired the reproductive function of prepubertal male rats, and a return to normal conditions during the postpubertal phase did not fully recover these impairments.

Abstract:Objective To investigate the effect of isoflurane inhalation during gestation period on plasticity of hippo-campal synapses in offspring rats. Methods Ten healthy pregnant SD rats at 14 day gestation were randomly divided into 2 gorups ( n = 5 each): control group (group C) and isoflurane group (group I). The rats in group C were mechanically ventilated with O2 while in group I the rats inhaled 1.3% isoflurane in O2 for 2 h a day until labor. Four weeks after birth 4 offspring rats from each pregnant rats (2 male, 2 female) were tested for learning and memory abilities using Morris water maze. Then the offspring rats were sacrificed and hippocampi isolated. The synaptic structure of hippocampal CA1 area was examined by trans-electron microscopy. Results Morris water maze test showed that the escape latency was significantly shorter and the number of times of spanning flat roof greater in group C than in group I. The structure of hippocampus was intact in group C but incomplete in group I. Meanwhile the thickness of synaptic density was significantly decreased in group I. Conclusion Isoflurane anesthesia of pregnant rats may induce learning and memory disabilities in offspring rats by inhibiting the plasticity of synaptic structure in hippocampus.

Abstract:Propofol is the most widely used short-acting intravenous anesthetic in clinical practice. Existing studies have shown that propofol has many effects on the cardiovascular system in addition to its anesthetic effect. Propofol can antagonize a variety of tachyarrhythmias and reduce the risk of recurrence, regulate autonomic balance of the heart, modulate circulatory dynamics, thereby increasing blood perfusion to vital organs such as the kidney, intestine, and brain, and exert myocardial protection and cerebral protection during ischemia-reperfusion injury. In this paper, we review the potential mechanisms of these effects and provide and ideas for future research and novel drug development of propofol and its derivatives in cardiac electrophysiology and circulatory dynamics.

Abstract:Environmental hazards are an increasing concern due to the rapid pace of industrialization. Among these hazards, noise and carbon monoxide (CO) are common risk factors and have been shown to cause serious health problems. However, existing studies focused on the individual effects of noise and CO exposure and the combined effects of these two factors remain poorly understood. Our study aimed to examine the combined effects of noise and CO exposure on testicular function by constructing individual and combined exposure models. Our findings indicated that combined exposure to noise and CO was associated with a higher risk of testicular damage and male reproductive damage when compared to exposure alone. This was evidenced by poorer semen quality and more severe pathological damage to the testis. This combined exposure led to higher levels of oxidative stress and apoptosis in the testes, with bioinformatics analyses suggesting the signaling pathways involved in these responses. Specifically, activation of the P53 signaling pathway was found to contribute to the testicular damage caused by the combined exposure. Encouragingly, pterostilbene (PTE), a novel phytochemical, alleviated combined exposure‐induced testicular damage by reducing oxidative stress and germ cell apoptosis. Overall, we identified joint reproductive toxicity resulting from the exposure to noise and CO, and found that PTE is a promising potential treatment for injuries caused by these factors.

Abstract:In recent decades, there has been a consistent decline in semen quality across the globe, with environmental pollution emerging as the predominant factor. Persistent organic pollutants (POPs) have garnered considerable attention due to their potent biological toxicity and resistance to natural degradation. Within this class of pollutants, polycyclic aromatic hydrocarbons (PAHs) and halogenated aromatic hydrocarbons (HAHs) have been identified as detrimental agents that can disrupt cellular physiological functions by activating aryl hydrocarbon receptor (AhR). However, the precise role of AhR in the adverse effects of environmental pollutants on male mammalian fertility remains incompletely understood. This article provides a comprehensive review of the impact of various environmental pollutants, specifically PAHs such as benzo[a]pyrene, 3-methylcholanthrene, and 7,12-dimethylbenzo[a]anthracene, HAHs including 2,3,7,8-tetrachlorodibenzo-p-dioxins, polychlorinated biphenyls, polybrominated diphenyl ethers, and the pollutant complex PM2.5, as well as cigarette smoke condensates, on male mammalian reproductive function. Additionally, this review focuses on the role of the AhR in mediating these effects.
