Influence of Herba Erigeromtis Extract on Learning and Memory Ability and Cholinesterase Activity of Rats with Dementia

Abstract:Objective To investigate the influence of Herba Erigenomtis( HE) extract on learning and memory ability and cholinesterase activity in rats with dementia. Methods Wistar rats were randomly divided into 5 groups. The animal model with dementia was produced by bilateral intracterthroventricular injection with β-amyloid peptide ( Aβ) 25-35 and abdominal cavity injection with D-galactose. Learning and memory ability of rats was detected by Morris Water Maze test,and the activities of acetycholinesterase (AChE) and butyrylcholinesterase (BuChE) were measured by Ellman's colorimetric method. Results In the rats with dementia in the model group as compared with normal and sham operation groups,the escaping latent period and the time in passing the platform were obviously increased ( P 0. 05) ,and the times of crossing the platform significantly were declined ( P 0. 05) ; the activities of AChE and BuChE were increased in brain tissues and decreased in plasma ( P 0. 05) . After treatment by HE extract,the ability of learning and memory was improved in the rats with dementia and the activity of cholinesterase in brain tissues and plasma was shown a tendency to recover to normal level ( P 0. 05) . Conclusion HE extract may improve the learning and memory ability of the rats with dementia and show a tendency to recover the activity of cholinesterase to normal level.
