Effect of Kudouzi on contractile activity of smooth muscle in guinea pigs

Abstract:AIM: To investigate effect and pathway of kudouzi decoction on smooth muscle of isolated splanchna in guinea pig. METHODS: The experiment was completed at the Physiological Laboratory of Basic College of Ningxia Medical College in December 2004. ① Totally 24 adult healthy guinea pigs of either gender with 3 colors of fur were selected in this study. ② Kudouzi provided by Yinchuan Hospital of Traditional Chinese Medicine was made into 1 kg/L decoction. Experimental mass concentrations were 1 × 104, 3×10-4, 1×10-3, 3×10-3, 1×10-2 and 2×10-2 kg/L. ③ Experimental method: Cholecyst, stomach, small intestine and bladder were obtained from guinea pigs. Three muscular strips of smooth muscles were established in each cholecyst with 72 in total, and 12 muscular strips were in stomach, small intestine and bladder, respectively. Muscular sockets of 12 muscular strips in cholecyst, stomach, small intestine and bladder were added with 1 × 104, 3×10-4, 1×10-3, 3×10-3, 1×10-2 and 2×10-2 kg/L kudouzi decoction, respectively. The interval was 2 minutes and the volume was 50 μL. ④ According to random lot method, 60 muscular strips were divided into 5 groups: atropine + kudouzi group, phenolamine + kudouzi group, verapamil + kudouzi group, aleryl + kudouzi group and indomethacin + kudouzi group, with 12 in each group. Guinea pigs in the above groups were treated with 1 × 10-6 mol/L atropine antagonist (blocking agent of cholinergic M receptor), 1×10-6 mol/L phenolamine (blocking agent of adnephrin α receptor), 1×10-7 mol/L verapamil (Ca2+ antagonist), 1×10-6 mol/L aleryl (blocking agent of histamine H1 receptor) and 1×10-6 mol/L indomethacin (blocking agent of prostaglandin receptor), and then kudouzi at above concentrations was added 2 minutes later. ⑤ Signals were output with JH-2 muscle tension converter; tension of samples, average amplitude of systolic wave and changes of systolic frequency were recorded with BL-410 biological function experimental system. ⑥ Differences of data were compared with mean t test. RESULTS: ① Tension of muscular strips in cholecyst was increased, systolic frequency was fastened, and average amplitude of systolic wave was shortened after adding with 1 × 104, 3×10-4, 1×10-3, 3×10-3, 1×10-2 and 2×10-2 kg/L kudouzi decoction. ② When mass concentration of kudouzi decoction was 2×10-2 kg/L, tension and systolic frequency of muscular strips of isolated cholecyst were less in phenolamine + kudouzi group than those in kudouzi group (t=2.340 1, 2.140 4, P < 0.05), but average amplitude of systolic wave was higher than that in kudouzi group (t=2.825 6, P < 0.05). Tension of muscular strips was weaker in verapamil + kudouzi group than that in kudouzi group (t=2.346 4, P < 0.05). ③ When mass concentration of kudouzi decoction was 1×10-2 kg/L, tension of muscular strips of isolated cholecyst was less in phenolamine + kudouzi group than that in kudouzi group (t=2.430, P < 0.05), but average amplitude of systolic wave was higher than that in kudouzi group (t=2.227 9, P < 0.05). Tension of muscular strips was weaker in verapamil + kudouzi group than that in kudouzi group (t=2.657 6, P < 0.05). ④ There was no effect of kudouzi decoction at various mass concentrations on tension of muscular strips, average amplitude of systolic wave and systolic frequency in stomach, small intestine and bladder of guinea pigs (P > 0.05). CONCLUSION: ① Kudouzi decoction has dosage-dependence on increasing tension of muscular strips, fastening systolic frequency and decreasing average amplitude of systolic wave. The effect can be blocked by phenolamine and verapamil partially, and the mechanism is possibly related with either adnephrin α receptor or Ca2+ channel of cell membrane of smooth muscle in cholecyst. ② There is no effect of kudouzi decoction on contraction of smooth muscle in stomach, small intestine and bladder of guinea pigs.
