Genetic polymorphism of DAB2IP, a novel GAP gene, has been associated with the risk of aggressive PCa. Also, loss of DAB2IP in PCa often leads to cell proliferation, cell survival, cell invasion and radioresistance. However, it is still unknown about its roles in AR signaling activation during PCa development. The objective of this study is to delineate the functional role and mechanism of action of DAB2IP on AR signaling in PCa. METHODS DAB2IP-stable transfectants PCa cells and siRNA-specific DAB2IP-knockdown normal human prostate epithelial cells were used. A series of biochemical, molecular biologic and tumor biologic assays such as pull down, immunoprecipitation, western blot, reporter gene assay, RT-PCR, MTT cell growth assay and immunostaining assay were employed. Knockout (DAB2IP−/−) animals and clinical specimens were used to validate in vitro results. RESULTS DAB2IP inhibits AR-mediated cell growth and gene transcription in PCa with two distinct mechanisms. First, DAB2IP can inhibit the non-genomic pathway of AR associated with c-Src. DAB2IP can interrupt the complex formation between c-Src and AR via its unique proline-rih (PR) domain, and then subsequently inactive the c-Src activity. Second, DAB2IP can also block the genomic pathway via inhibiting AR targeting gene expression. DAB2IP can interrupt AR nuclear translocation, in which DAB2IP recruits PP2A to dephosphorylate AR phosphorylation elicited by androgen stimulation. Recent studies reported the presence of AR variants in PCa cells appear to be constitutively active. DAB2IP appears to be a potent inhibitor to suppress the activity of AR variants. In addition, DAB2IP is capable of suppressing AR activity induced by non-androgen ligand (e.g., Wnt) in PCa cells. In vivo, DAB2IP-/- animals exhibit hyperplastic lesion in prostate epithelia and AR becomes hyperactive in these DAB2IP-/- cells. Consistently, loss of DAB2IP correlates with AR activation in clinical specimens.
