Abstract:The understanding of physiology,pathology and pathogenesis of corneal diseases are furthered by study of limbal stem cells,stromal antigen presenting cells (APC) and endothelial cells,though many questions are still open to discussion.Contrast to previous assumption,corneas,especially the stromal layer,host a large number of dendritic cells,monocytes and macrophages that could initiate specific immune response to invading and captured pathogens.Conventional corneal keratocytes are capable of differentiating into functional APC.The existence of adequate number and various types of APC ensure effective initiation of immune response to foreign antigens in corneas.The corneal endothelial cells,however,play more important role in maintaining the homeostasis of corneas or when challenged with non-pathogenic factors,as in the case of penetrating keratoplasty.So far not much is known with the mechanism of chronological decrease of endothelial cells.Corneal dystrophy is another disorder with which not much is known about the pathogenesis though epidemiological surveys implied several disease-causing genes.The last prominent corneal disease discussed in this article is keratitis caused by virus,bacteria and fungus.The knowledge about corneal biology expands with the rapid progress of biomedical methodology.Whole genome scan and microarrays based on genomics,proteomics and bioinformatics are being applied in the ophthalmologic study.We look forward to significant contribution by Chinese scientists in this field.
