Abstract:This study was designed to investigate the effect of endothelin-1(ET-1)on the proliferation of broiler pulmonary artery smooth muscle cells(PASMC)and the corresponding receptor involved in this process.The proliferative effect of ET-1 on cultured PASMC was detected with methyl thiazolyl tetrazolium(MTT)assay,and the underlying receptor pathway was evaluated by block of endothelin A receptor(ETAR)sites using BQ123 and ETBR sites using BQ788,respectively.It was found that the ET-1 significantly enhanced the proliferation of broiler PAMSC(P0.05);however,this effect was blocked by BQ123.In contrast,BQ788 had no effect on PASMC proliferation stimulated by ET-1(P0.05).The results demonstrated that ET-1 enhanced the proliferation of broiler PASMC,which was mediated by ETAR rather than ETBR.
