Abstract:AIM To evaluate bioequivalence of lansoprazole and its metabolites 5-hydroxy lansoprazole and lansoprazole sulphone in 22 healthy male vounteers after a single oral dose of 30 mg.METHODS LC:using Diamonsil C_(18) column(250 mm×2.1 mm,5 μm)with preliminary column Octadecy1 C_(18)(4.0 mm×3.05 mm);the mobile phase consiting of acetonitrile-2 mmol·L~(-1) ammonium acetate solution(60∶40,V/V);the flow rate was 0.2 mL·min~(-1);and the column temperature was 40℃.MS:using electrospray ionization (ESI),and selected ion mass spectral (m/z) 368.1/164.1 (lansoprazole),384.0/180.0 (5-hydroxylansoprazole),384/115.9 (lansoprazole sulphone),364.1/188.1(internal standard)to quantify.The plasma samples were treated with acetonitrile precipitation.RESULTS The pharmacokinetic parameters of lansoprazole (test) were:AUC:(12 989.16±13 356.96)μg·h·L~(-1),c_(max)(2 780±1 282)μg·L~(-1),t_(max):(1.73±0.74)h,T_(1/2):(2.74±2.01)h;5-hydroxy lansoprazole(test) were:AUC:(197.51±68.54)μg·h·L~(-1),c_(max):(73.45±39.09)μg·L~(-1),t_(max):(1.59±(0.76))h,T_(1/2):(3.26±1.97)h;lansoprazole sulphone (test) were:AUC:((1 496.74)±2 517.81)μg·h·L~(-1),c_(max):(160.75±146.12)μg·L~(-1),t_(max):(2.18±1.55)h,T_(1/2):(2.94±3.48)h.The pharmacokinetic parameters of lansoprazole (reference) were:AUC:(13 426.99±3 671.51)μg·h·L~(-1),c_(max):(2 945±(1 383))μg·L~(-1),t_(max):(1.90±0.63)h,T_(1/2):(2.53±2.06)h;5-hydroxy lansoprazole (reference) were:AUC:(211.77±(78.40))μg·h·L~(-1),c_(max):(77.70±43.78)μg·L~(-1),t_(max):(1.83±0.65),T_(1/2):(3.03±1.77)h;lansoprazole sulphone (reference) were:AUC:(1 510.80±2 525.48)μg·h·L~(-1),c_(max):(169.20±151.28)μg·L~(-1),t_(max):(2.27±1.37)h,T_(1/2):(2.64±2.96)h.There were no significant differences in all pharmacokinetic parameters between 2 capsules.(P>0.05).CONCLUSION The results of two one-side test show that 2 preparations are bioequivalent.
