Abstract:We report an unprecedented assembling performance of amide-containing, homo-structured poly(N-alkylacrylamide)s, through which they can assemble into discrete spherical particles with size ranging from tens of nanometres to several micrometres. For a specific polymer, a facile control over topology and scale of assembled structures can be accomplished via solvent conditions. Assembling using appropriate solvent gives rise to capsule-like assemblies. The molecular structure of polymers, such as the side chain chemistry and topology display striking impact on the size and morphology of resulting assemblies. These polymers might function as novel model systems for investigating complex functions performed by amide moieties in proteins, while this assembling offers a simple, facile but powerful and cost-effective manufacturing route for nano- or microstructured materials that may find great potential in diverse applications varying from cosmetics, drug delivery, nanomedicine, to biotechnology.

Abstract:Today various nano-moieties including nanoparticles, nanocubes, nanosticks, nanoflowers and nanoprimes have been synthesised. It is an interesting challenge to build nanostructures using those nano-moieties as blocks owing to their distinct properties and numerous applications. Herein, we demonstrate a smart way, self-assembling, to arrange nanoparticles and nanocubes into a line, sphere, network, bowl-bottom etc. The obtained anisotropic nanostructures have been employed to enhance the sensor sensitivity and to identify the hot-spot position for surface-enhanced Raman scattering (SERS).

Abstract:The enzyme-triggered self-assembly of peptides has flourished in controlling the self-assembly kinetics and producing nanostructures that are typically inaccessible by conventional self-assembly pathways. However, the diffusion and nanoscale chemical gradient of self-assembling peptides generated by the enzyme also significantly affect the outcome of self-assembly, which has not been reported yet. In this work, we demonstrated for the first time a spatiotemporal control of enzyme-triggered peptide self-assembly. By simply adjusting the temperature, we could change both the catalytic activity of the enzyme of phosphatase and their aggregation states. The strategy kinetically controls the production rate of self-assembling peptides and spatially controls their distribution in the system, leading to the formation of nanoparticles at 37 °C and nanofibers at 4 °C. The nanofibers showed ∼10 times higher cellular uptake by 3T3 cells than the nanoparticles, thanks to their higher stability and more ordered structures. Using such spatiotemporal control, we could prepare optimized nanoprobes with low background fluorescence, rapid and high cellular uptake, and high sensitivity. We postulate that this strategy would be very useful in general for preparing self-assembled nanomaterials with controllable morphology and function.

Abstract:Peptide self-assemblies show intriguing and tunable physicochemical properties, and thus have been attracting increasing interest over the last two decades. However, the micro/nano-scale dimensions of the self-assemblies severely restrict their extensive applications. Inspired by nature, to genuinely realize the practical utilization of the bio-organic super-architectures, it is beneficial to further organize the peptide self-assemblies to integrate the properties of the individual supermolecules and fabricate higher-level organizations for smart functional devices. Therefore, cumulative studies have been reported on peptide microfabrication giving rise to diverse properties. This review summarizes the recent development of the microfabrication of peptide self-assemblies, discussing each methodology along with the diverse properties and practical applications of the engineered peptide large-scale, highly-ordered organizations. Finally, the current limitations of the state-of-the-art microfabrication strategies are critically assessed and alternative solutions are suggested.

Abstract:Supramolecular self-assembly in biological systems holds promise to convert and amplify disease-specific signals to physical or mechanical signals that can direct cell fate. However, it remains challenging to design physiologically stable self-assembling systems that demonstrate tunable and predictable behavior. Here, the use of zwitterionic tetrapeptide modalities to direct nanoparticle assembly under physiological conditions is reported. The self-assembly of gold nanoparticles can be activated by enzymatic unveiling of surface-bound zwitterionic tetrapeptides through matrix metalloprotease-9 (MMP-9), which is overexpressed by cancer cells. This robust nanoparticle assembly is achieved by multivalent, self-complementary interactions of the zwitterionic tetrapeptides. In cancer cells that overexpress MMP-9, the nanoparticle assembly process occurs near the cell membrane and causes size-induced selection of cellular uptake mechanism, resulting in diminished cell growth. The enzyme responsiveness, and therefore, indirectly, the uptake route of the system can be programmed by customizing the peptide sequence: a simple inversion of the two amino acids at the cleavage site completely inactivates the enzyme responsiveness, self-assembly, and consequently changes the endocytic pathway. This robust self-complementary, zwitterionic peptide design demonstrates the use of enzyme-activated electrostatic side-chain patterns as powerful and customizable peptide modalities to program nanoparticle self-assembly and alter cellular response in biological context.

Abstract:Peptides, the fundamental building units of biological systems, are chiral in molecular scale as well as in spatial conformation. Shells are exquisite examples of well-defined chiral structures produced by natural biomineralization. However, the fundamental mechanism of chirality expressed in biological organisms remains unclear. Here, we present a system that mimics natural biomineralization and produces enantiopure chiral inorganic materials with controllable helicity. By tuning the hydrophilicity of the amphiphilic peptides, the chiral morphologies and mesostructures can be changed. With decreasing hydrophilicity of the amphiphilic peptides, we observed that the nanostructures changed from twisted nanofibers with a hexagonal mesostructure to twisted nanoribbons with a lamellar mesostructure, and the extent of the helicity decreased. Defining the mechanism of chiral inorganic materials formed from peptides by noncovalent interactions can improve strategies toward the bottom-up synthesis of nanomaterials as well as in the field of bioengineering.

Abstract:Peptides are excellent building units to construct supramolecular biomaterials for diverse biomedical applications. A vast amount of research effort over the past two decades has focused on the rational design of peptidic building units and the formulation protocols required to create supramolecular assemblies that can chemically and structurally emulate the functions of naturally occurring nanostructures. A rapidly growing interest in the field is the spatiotemporal regulation of the formation and immolation pathways of peptide assemblies, an essential characteristic to control if these constructs are to fulfill their roles as biologically active and compatible materials. In this review, we discuss the necessity for control over the assembly and morphological transformation of peptides and peptide conjugates, the critical role of these properties for their ultimate biological application, as well as the methods by which this can be programmed into the molecular structure. A particular focus will be on recent advances in the use of enzyme and redox processes for exerting spatiotemporal control over a variety of peptide-based nanostructures.

Abstract:This article demonstrates the ability of viral fusion peptides to form spiral‐like nanostructures using uniaxial compression in a Langmuir–Blodgett trough. By controlling compression forces, nanofibrils with specific curvatures and inter‐fiber distances are achieved, transitioning from fluid to gel states. These peptide spirals also serve as templates for gold replicas, offering new possibilities in engineering functional nanomaterials. Self‐assembling peptides (SAPs) are versatile building blocks in the creation of hierarchical nanostructures. Although SAPs promise precise control over assembled morphology and dimensions, experimental validation is still crucial. In this sense, recent efforts have focused on nanospiral structures, which mimic natural architectures and hold potential for biomedical and nanotechnological applications. Here, it is demonstrated that SARS‐CoV‐2 fusion peptides are able to form specific spiral‐like structures using interfacial assembly as a fabrication methodology, along with fine‐tuning of the spiral ensembles through an imposed interfacial flow due to the uniaxial compression in a Langmuir–Blodgett trough. Molecular characterization by atomic force microscopy, neutron reflectometry, interfacial shear rheology, and infrared spectroscopy, highlighted how variations in fusion peptide sequences influence assembly, highlighting the importance of molecular design. These spiral structures are subsequently modified to serve as templates for metallic replicas, expanding the potential of peptide‐guided self‐assembly in fabricating tailored nanostructured surfaces with sub‐10 nm dimensions over cm2 areas.

Abstract:Short peptides that are made of natural amino acids were never seriously considered as useful materials as recent as 16 years ago. However, the discovery of a class of self-assembling peptides that spontaneously undergo self-organization into well-ordered structures resulted in a conceptual change. Since then diverse classes of short peptides have been invented with broad applications including 3D tissue cell culture, reparative and regenerative medicine, tissue engineering, slow drug release, stabilization of membrane proteins to develop nanobiotechnology and molecular devices. Furthermore one of the self-assembling peptides has shown promise not only to slow down prion infection, but also to extend ∼50% animal life. Molecular design using short peptides as new materials may play increasingly important role in nanoscience, nanotechnology, nanobiotechnology and nanomedicine.

Abstract:The self-assembly of nanoparticles is a challenging process for organizing precise structures with complicated and ingenious structures. In the past decades, a simple, high-efficiency, and reproducible self-assembly method from nanoscale to microscale has been pursued because of the promising and extensive application prospects in bioanalysis, catalysis, photonics, and energy storage. However, microscale self-assembly still faces big challenges including improving the stability and homogeneity as well as pursuing new assembly methods and templates for the uniform self-assembly. To address these obstacles, here, a novel silver-coated nanopore is developed which serves as a template for electrochemically generating microcyclic structures of gold nanoparticles at micrometers with highly homogenous size and remarkable reproducibility. Nanopore-induced microcyclic structures are further applied to visualize the diffusion profile of ionic flux. Based on this novel strategy, a nanopore could potentially facilitate the delivery of assembled structures for many practical applications including drug delivery, cellular detection, catalysis, and plasmonic sensing.

Abstract:Biomolecular self-assembly provides a facile way to synthesize functional nanomaterials. Due to the unique structure and functions of biomolecules, the created biological nanomaterials via biomolecular self-assembly have a wide range of applications, from materials science to biomedical engineering, tissue engineering, nanotechnology, and analytical science. In this review, we present recent advances in the synthesis of biological nanomaterials by controlling the biomolecular self-assembly from adjusting internal interactions and external stimulations. The self-assembly mechanisms of biomolecules (DNA, protein, peptide, virus, enzyme, metabolites, lipid, cholesterol, and others) related to various internal interactions, including hydrogen bonds, electrostatic interactions, hydrophobic interactions, π–π stacking, DNA base pairing, and ligand–receptor binding, are discussed by analyzing some recent studies. In addition, some strategies for promoting biomolecular self-assembly via external stimulations, such as adjusting the solution conditions (pH, temperature, ionic strength), adding organics, nanoparticles, or enzymes, and applying external light stimulation to the self-assembly systems, are demonstrated. We hope that this overview will be helpful for readers to understand the self-assembly mechanisms and strategies of biomolecules and to design and develop new biological nanostructures or nanomaterials for desired applications.

Abstract:In contemporary biopolymer synthesis, conservative synthesis of monomers to polymers has been achieved by using organocatalyst. However, the high-efficiency conversion in biopolymer synthesis is rare, due to the difficulties in precisely monitoring and controlling the reaction immediates1,2. Here, we report a novel molecular machine for controllable single-biomolecule synthesis with biological protein nanopore (NP-Synth.), which combines the advantages of manipulating reaction sites at atomically level and real-time recording of the reaction trajectories. Driven by the applied voltage, the two reactant molecules from the reducing phase successively enters into the nanopore confinement. The catalytical site inside the nanopore facilitates the rapid conjugation of these two molecules. The nanopore confinement here directs the orientation of the reactive group of the reactant for directly driving the synthesis. Following the real-time current traces, the reaction process is optimized by adjusting the length of the reactant and the applied potential. This work shows that the NP-Synth. can produce, by combining well-defined protein nanopore, ordered biopolymers that were previously inaccessible to organic chemistry.

Abstract:Peptide-functionalized polymeric nanoparticles were designed and self-assembled into continuous nanoparticle fibers and three-dimensional scaffolds via ionic complementary peptide interaction. Different nanoparticle compositions can be designed to be appropriate for each desired drug, so that the release of each drug is individually controlled and the simultaneous sustainable release of multiple drugs is achieved in a single scaffold. A self-assembled scaffold membrane was incubated with NIH3T3 fibroblast cells in a culture dish that demonstrated non-toxicity and non-inhibition on cell proliferation. This type of nanoparticle scaffold combines the advantages of peptide self-assembly and the versatility of polymeric nanoparticle controlled release systems for tissue engineering.

Abstract:Peptide self-assembling materials have recently emerged as one of the most promising biomaterials. In this study, we synthesized three amphipathic cyclopeptides (CPs1-3) with systematically modified side chains to control the morphologies of self-assembly. Owing to the varying modifications of the side chains, various micro/nanostructures were achieved because of different driving forces. The obtained morphologies were characterized by transmission electron microscopy (TEM) and scanning electron microscope (SEM). Apart from hydrogen bond interactions for CPs, CP1 self-assembles into spindle-like aggregates with an average diameter of 70 nm and a length of 300 nm owing to the salt-bridge interactions in a neutral medium. CP2 can form micro/nanotubes with the diameters ranging from 750 nm to 2 mm via the hydrophilic and hydrophobic interactions between the cyclopeptide ring and the alkyl chain. While CP3 forms solid nanospheres with an average diameter of 516 nm through aromatic-aromatic pi-pi interactions. This approach can lead to the fabrication of new functional supramolecular assemblies and materials and broaden the insights for the mechanism of the self-assembly.

Abstract:The self-assembly of a small number of nanoparticles into superstructures that mimic the geometry of molecules provides an unprecedented route for creating materials with precisely defined structures and potentially programmable functionalities. Such nanoparticle clusters (NPCs), also known as colloidal molecules, have a wide range of applications due to the decisive ensemble effect. Here, a universal and straightforward strategy is developed to construct NPCs with tunable molecular-like geometries by confining the self-assembly of hydrophobic nanoparticles within micelles formed by amphiphilic copolymers. It is found that confinement assembly of both spherical and anisotropic nanoparticles can lead to NPCs, the molecular-like conformation of which is widely tunable by adjusting the ratio between copolymers and nanoparticles. Mechanistic studies reveal the formation of large-vesicle intermediates along the path toward forming NPCs. This work establishes a facile and general strategy of assembling finite nanoparticles with precisely tunable geometries without introducing any directional interactions, which can accelerate the exploration of clustered superstructures toward broad applications.
