Abstract:The fragmentation patterns of N-diisopropyloxyphosphoryl-l-α-Ala (DIPP-l-α-Ala), N-diisopropyloxyphosphoryl-d-α-Ala (DIPP-d-α-Ala), N-diisopropyloxyphosphoryl-β-Ala (DIPP-β-Ala) and N-diisopropyloxyphosphoryl-γ-amino butyric acid (DIPP-γ-Aba) were investigated by electrospray ionization tandem mass spectrometry (ESI-MS/MS). DIPP-d-α-Ala showed the same fragmentation pathways as DIPP-l-α-Ala. In the fragmentation of protonated DIPP-β-Ala, the characteristic fragment ion [M+H−2C3H6−H2O−CH2CO]+ appeared and could be used to distinguish β-Ala from l-α-Ala and d-α-Ala through tandem mass spectra, even though they possess the same molecular weight. In the fragmentation of protonated DIPP-γ-Aba, the break of PN bond occurred and an interesting protonated lactam ion with five-membered ring was generated. Furthermore, in the MS3 spectrum of [M+Na−2C3H6]+ ion of DIPP-γ-Aba, a strong intensity of unique fragment ion, namely lactam-sodium adduct with five-membered ring, was observed, which could be considered as a mark for γ-amino acids. The stepwise fragmentations of their [M+Na]+ ions and [M−H]− ions showed that they all underwent a PN to PO bond migration through a five-membered or six-membered or even seven-membered ring transition state, respectively, which supported the great affinity of hydroxyl for phosphoryl group.

Abstract:Phosphonate peptides were synthesized and investigated by positive ion electrospray ionization mass spectrometry in conjunction with tandem mass spectrometry. It was found that, for the sodiated adduct, there were two rearrangement reactions. In path A, there was C-C covalent bond cleavage with consecutive migration of a hydrogen atom to lose the amino alcohol molecule. In path 8, two consecutive covalent cleavages of the C-N and P-O bonds occurred.

Abstract:The negative-ions of N-phosphoryl amino acids were studied by electrospray ionization tandem mass spectrometry (ESI-MS/MS). The negative-ion ESI-MS/MS of N-phosphoryl amino acids showed characteristic fragmentation patterns different from those observed in the corresponding positive-ion ESI-MS/MS and negative-ion fast-atom bombardment mass spectra. For negative-ion EST-MS/MS, a unique fragmentation from the N-terminal of N-phosphoryl amino acids or peptides containing a free beta-OH or CO2H group was observed to yield the characteristic fragment ion (RO)(2)P(O)O. The ease of the rearrangement depended on the position of the hydroxyl group in amino acids or peptides, and the N --> O rearrangement mechanism was proposed to involve the participation of the hydroxyl group. From previous solution-phase experiments and theoretical calculations, it was found that the beta-OH group was more active than gamma-OH, and the corresponding difference in negative-ion ESI-MS/MS was consistent with those previous findings. Abstract:Peptides were phosphonylated at their N-termini by reacting with ethoxyphenylphosphinate in the presence of triethylamine and tetrachloromethane under mild conditions. The phosphonylated peptides were analyzed by tandem electrospray ionization mass spectrometry. N-Terminal phosphonylation selectively increased the intensities of b(n)-type ions relative to other ion types. The resulting simplified mass spectra clearly show the sequential loss of amino acid residues from the C-termini of peptides, providing a convenient and rapid method for peptide sequencing.

Abstract:Lys-containing peptides were phosphonylated at their N-termini by reacting with ethoxyphenylphosphinate (EPP-H) in the presence of tetrachloromethane and triethylamine, and picomolar amount of the modified phosphonamidate peptides were determined by electrospray ionization mass spectrometry, together with tandem mass spectral technique, the intensive bn-type ions relative to other type ions were observed, which clearly provided sequences of the original peptides.

Abstract:Protein phosphorylation is an important post-translational modification (PTM), and involves in many cell activities and disease processes. Nowadays, mass spectrometry (MS) is a powerful tool in peptide-based proteomics. But phosphopeptides, especially multiply phosphorylated peptides, are difficult to be detected by MS. One opinion is that the presence of unphosphorylated peptides lead to the ionization suppression of phosphopeptides. But contrary opinion exists. Moreover, the suppression effect caused by different types of phosphopeptides hasn't been studied yet. In this study, a set of synthetic standard phosphopeptides (singly, doubly and triply phosphorylated peptides) with the same amino acid sequence, and the unphosphorylated peptides were injected into mass spectrometer with or without mixing. Through the ratios of signal intensities of the phosphopeptides before and after mixing, we confirmed that the signals of phosphopeptides were suppressed by those of the unphosphorylated peptides. In addition, the signals of the doubly phosphorylated peptides were slightly suppressed by those of the singly phosphorylated peptides with the same amino acid sequence. The signals of the triply phosphorylated peptides were notably suppressed by the singly and doubly phosphorylated peptides with the same amino acid sequence. Therefore, the singly and multiply phosphorylated peptides should be fractionated and enriched before mass spectrometry analysis.

Abstract:N-phosphorylation labeling was utilized to analyze the low molecular weight compounds by using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). A wide range of natural amino acids and short peptides was successfully analyzed by MALDI-TOF MS without matrix background interferences. The N-phosphorylation labeling reaction was carried out easily within 30 min in a one-pot reaction under mild reaction conditions. The phosphoryl derivatization reaction is a global labeling approach with high selectivity and high specificity with targeting only on the N-terminal and ε-amino group of Lys. The incorporation of a neutral phosphoryl group with high gas-phase affinity of protons not only improves the ionization efficiency of target molecules and simultaneously decreases the ion suppression effects in MALDI-TOF MS analysis, but also greatly reduces or eliminates the matrix background interferences by suppressing the matrix signals and increasing the molecular weight of the targeted compounds. By applying the N-phosphorylation labeling approach, many amino acids could be detected in serum samples by using MALDI-TOF MS.

Abstract:The conjugates of phosphoamino acids with 3'-azido-3'-deoxythymidine were synthesized and their structures were determined by various spectral methods. In positive and negative ion electrospray mass spectrometry (ESI-MS), the fragmentation pathways were investigated in conjunction with tandem mass spectrometry (MS/MS). The results showed that there were very different characteristic fragment ions in the positive ion MS/MS spectra and the negative ion MS/MS spectra.

Abstract:We have developed a labeling method for sequencing of mixed peptides through modification of peptides and multistage electrospray ionization-mass spectrometry (ESI-MS). Mixed peptides were selectively phosphonylated on amino group through reaction of 2-bromoethyl phenylphosphinate (BEPP-H) or methyl/methyl-d3 (1:1) phenylphosphinate (MPP-X) with peptides in the presence of tetrachloromethane and triethylamine under mild conditions to provide the labeled phosphonamidate peptides, and their primary ESI mass spectra showed doublet and triplet peaks. The triplet and doublet peaks were corresponding to the protonated molecules of the modified peptides having and having no one lysine residue, respectively, and the singlet peaks were from the non-peptides. Further, tandem mass spectra (MS/MS) of the readily identified protonated molecules unambiguously gave sequence of each member in mixed peptides.

Abstract:Amino acid phosphoramidates of adenosine were synthesized and determined by positive and negative ion electrospray ionization mass spectrometry (ESI-MS) in conjunction with tandem mass spectrometry (MS/MS). The fragmentation pathways were investigated. In the positive ion mass spectra abundant characteristic fragment ions appeared, and many complementary ions were found. In the negative ion mass spectra only a few fragment ions were observed, and most of them contained phosphoryl groups. The results show that ESI-MS is a useful tool for structural determination of amino acid phosphoramidates of nucleosides. Copyright (C) 2000 John Wiley & Sons, Ltd.

Abstract:A novel rearrangement reaction with a carbonyl oxygen migration was observed in the electrospray ionization tandem mass spectra of N-diisopropyloxyphosphoryl dipeptides and their analogues. A possible mechanism was proposed and supported by the MS/MS study, derivatization of different functional groups and deuterium labeling experiments. It was found that metal ions could catalyze the rearrangement through a five-membered ring intermediate. A strong affinity between the phosphoryl group and oxygen atom in the gas phase was proposed to result in this kind of rearrangement reaction, which might provide some basic information on the nature of phosphorylation in biochemistry. The replacement of N-terminal alpha-alanine by beta-alanine stopped the migration, which provides a simple method for differentiating the alpha- and beta-alanine residues at the N-terminus of peptides.

Abstract:Peptides were phosphonylated at their N-termini through reaction of ethoxyphenylphosphinate (EPP) with peptides in the presence of triethylamine and tetrachloromethane under mild conditions. The phosphonamidate peptides were analyzed by tandem electrospray ionization mass spectrometry after addition of trace amounts of sodium chloride to the resulting solution above, and ESI-MS/MS of the sodiated molecules gave predominantly [b n + Na + OH] + and [b n + Na − HCO] + ions that clearly showed the sequential loss of amino acid residues from C-terminus of peptides, and the original peptides were readily assigned. The rapid, high sensitive and convenient method can widely be used for determination of peptide sequence.

Abstract:Amino acid methyl ester phosphates were synthesized and determined by using positive-ion mode electrospray ionization mass spectrometry (ESIMS) in combination with multistage tandem mass spectrometry. The fragmentation pathways were investigated, and it was observed that most fragment ions contained the phosphoryl group. It was interesting to observe that the fragmentation pathways of the protonated molecule show some differences when compared with those of the sodium ion adduct. The methoxy group of amino acid methyl ester can migrate from the carbonyl group to the phosphoryl group in the sodium ion adduct.

Abstract:The study of the dissociation of the protonated molecular species [M+H](+) and selected fragment ions allowed proposals for the main fragmentation pathways of the title compounds. The main fragments are formed by expelling a molecule of thymine, thymidine (d4T) or isopropyl. The most striking feature of the tandem mass (MS/MS) spectra is the cleavage of C-CO bonds between N-methylpyrrole and carbonyl groups in the presence of the amidine. Abstract:In this article, it was demonstrated that separation and determination of 20 amino acids were accomplished by micellar electrokinetic chromatography (MEKC) coupling with novel multiphoton excited fluorescence (MPEF) detection method. Different from MPEF achieved by expensive fs laser, continuous wave (CW) diode laser of ultra-low cost was uniquely employed in our MPEF system. Amino acids were fluorescently labeled with fluorescein isothiocyanate (FITC), and were subjected to sodium dodecyl sulfate (SDS)-based MEKC separation and CW-based MPEF detection. The result was compared with that by single photon excited fluorescence (SPEF), which indicated that MPEF had the advantages of better mass detectability and higher separation selectivity over SPEF. Quantitative analysis was performed and revealed linear dynamic range of over 2 orders of magnitude, with mass detection limit down to ymole level. To evaluate the reliability, this method was successfully applied for analyzing a commercial nutrition supplement liquid.

Abstract:The positive and negative ESI-MS/MS spectra of N-ethoxy(phenyl) phosphoryl amino acids (EPP-AA) were investigated by electrospray ionization (ESI) ion trap mass spectrometry. The fragmentation pathways of [ M + Na ](+) and [ M - H ](-) ions are proposed and rationalized. The observation may have some potential applications in the interpretation of the MS/MS spectra of novel N-phosphoryl compounds. The complexity of MS/MS spectra of EPP-AA [ M + Na ](+) ions is decreased compared with that. of N-dialkyloxyphosphoryl amino acid. Therefore, the new phosphonamidate method may be considered one of the superior methods that can be used in sequencing peptides and proteins extensively.

Abstract:Free or resin-bound peptides were phosphorylated at their N-termini by reacting with dimethyl phosphite in the presence of tetrachloromethane and triethylamine, and some of them were labeled using partial deuterium-labeled dimethyl phosphites (molar ratio of m + 6 and m = 1:1 or m + 6, m + 3 and m = 1:2:1) as the phosphorylating agents. The monophosphorylation of the lysine-containing peptides selectively occurred on the amino group of the N-terminus, not the side-chain of lysine residue. The resin-bound phosphoramidate peptides were cleaved by TFA before ESI-MS. The modified peptides were determined by electrospray ionization mass spectrometry, the protonated molecules of the unlabeled phosphoramidate peptides showed the singlet peaks, and those of the labeled phosphoramidate peptides displayed the doublet and/or triplet peaks. Tandem mass spectra (MS/MS) of the chosen protonated molecules gave sequential loss of the amino acid residues from the C-termini of the peptides, providing a convenient and rapid method for peptide sequencing.
