Abstract:The palladium-catalyzed tandem reactions of 2,7-alkadiynylic carbonates with 2,3-allenyl sulfamides for the syntheses of fused tricycles such as 1,3,6,7,8,9-hexahydrofuro[3,4-h] isoquinoline, 2,3,6,7,8,9-hexahydro-1H-pyrrolo[3,4-h]isoquinoline, and 2,3,4,7,8,9-hexahydro-1H-cyclopenta[h]isoquinoline derivatives have been developed. A mechanism has been proposed based on the isolation of a by-product.

Abstract:In this paper, we report a novel sequential reaction sequence consisting of a series of processes including Sonogashira coupling, propargyl-allenyl isomerization, [3+2]cycloaddition, denitrogenation, and highly selective biradical coupling reaction, providing an efficient synthesis of fused polycyclic pyrrole derivatives. The reaction features one C-C bond and two C-N bonds formed to construct the pyrrole unit, together with two rings efficiently assembled in one stroke.

Abstract:An efficient synthesis of novel fused heterocycles has been established via microwave-assisted palladium-catalyzed intramolecular direct arylation. The acyclic aryl bromides are readily available from microwave-assisted one-pot annulation of N-(2-bromobenzyl)-2-aminophenois and ethyl 2-bromoalkanoates. The intramolecular direct arylation is then performed in the presence of palladium(II) acetate and dppf (10 mol% each) in toluene using potassium carbonate (2 equiv) as the base under microwave heating (150 degrees C, 1 h) to afford the products in 43-99% yields. Steric effect is observed for Ar-Ar bond formation, giving a substantial amount of the debromination byproduct.

Abstract:A variety of dihydroisobenzofuran derivatives has been prepared in good yields via an interesting sequential reaction consisting of palladium‐catalyzed coupling, propargyl‐allenyl rearrangement, [4+2] cycloaddition and aromatization. A double‐coupling, rearrangement, [4+2] cycloaddition and aromatization process is also described for the synthesis of more complex and diverse structures.

Abstract:A palladium-catalyzed domino process for the quick assembly of tricyclic-fused heterocycles starting from aryl iodides and functionalized isocyanides containing a disubstituted terminal alkene has been developed. The process is triggered by intermolecular isocyanide insertion, followed by Heck-type carbopalladation of the intramolecular alkene moiety and subsequent C(sp2)-H activation. Moreover, an asymmetric version of this reaction could also be realized in good yield with moderate enantioselectivity after preliminary exploration of chiral ligands.

Abstract:A DABCO‐promoted bicyclization/rearrangement reaction has been developed for the synthesis of tetrasubstituted furans from 1,4‐enediones at room temperature. This transformation involves aza‐Michael addition, intramolecular bicyclization, and rearrangement processes. In addition, the 4‐ureidofuran‐3‐carboxylate ester products can further be converted to furo[3,4‐d]pyrimidine‐2,4‐diones through intramolecular substitution cyclization under heating conditions.

Abstract:A cooperative Rh(II)/Pd(0) dual-catalysis strategy that promotes a cyclization/allylic alkylation cascade of stable α-diazo-δ-keto-esters has been developed. Highly substituted 3(2<i>H</i>)-furanones with a C2-quaternary center can be obtained efficiently under mild conditions via one-pot synthesis. Remarkably, this binary catalytic system shows high chemo-, regio-, and stereoselectivity and excellent tolerance to various functionalities.

Abstract:Described herein is the development of a palladium(II)-catalyzed two- or three-component reaction of 2-(1-alkynyl)-2-alken-1-ones with nucleophiles and allylic chlorides. Various types of nucleophiles such as O-, N-, C-based nucleophiles and olefin-tethered O-, N-, C-based nucleophiles were investigated. The scope, mechanism and application of this Pd(II) -catalyzed domino reaction were studied. In these transformations, the palladium catalyst exhibits a dual role, serving simultaneously as a Lewis acid and a transition metal. Two possible reaction pathwavs (cross-coupling reaction vs. Heck reaction) from the same intermediate furanylpalladium species were observed. The reaction pathway is dependent on the property of the nucleophile and the length of the tethered chain as well. When olefin-tethered O- based nucleophiles were used., only the cross-coupling reaction pathway was observed, in contrast, both reaction pathways were observed when olefintethered C-based nucleophiles were employed. The product ratio is dependent on the length of the tethered chain. Furthermore, ring-closing metathesis (RCM) of corresponding furans with C=C bonds provides an easy method for the preparation of functionalized oxygen-heterocycles - 3,4-fused bicyclic furans. It is also noteworthy that allylic chloride can be as an oxidant besides its well known function as an alkylating reagent.

Abstract:The first enantioselective formal [5+4] cycloaddition is realized under palladium catalysis to deliver benzofuran-fused nine-membered rings. These medium-sized heterocycles and derivatives undergo unique rearrangements induced by transannular bond formation, resulting in the production of two classes of densely substituted polycyclic heterocycles in excellent efficiency and stereoselectivity.
