Extraction, purification and characterization of Spirulina phycocyanin

Abstract:Phycocyanin is an important natural food colorant extracted from blue-green algae. As a kind of blue-green algae, Spirulina contains abundant nutrients, including proteins (especially phycocyanin), amino acids, polysaccharides, carotenoids, various vitamins and mineral elements. Spirulina platensis and Spirulina maxima are two best species of Spirulina genus. In this study, an effective method was used to extract and purify Spirulina phycocyanin. Phycocyanin was efficiently extracted from Spirulina platensis powder with water as solvent using water bursting method, resulting in high extraction yield of 48.35 mg/g. Subsequently, crude phycocyanin was purified using activated charcoal. The optimal conditions for purification were 8 % activated charcoal dosage, mixing time of 4 min and mixing speed of 200 rpm. Under optimal conditions, phycocyanin purity was 1.77. Phycocyanin was found to be stable when temperature was lower than 40 °C. In addition, the stability decreased rapidly when temperature was >60 °C or phycocyanin was exposed to light. It was also found that phycocyanin had higher antioxidant activity. Radical scavenging activities of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2, 2-casino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) were 60.10 % and 91.49 %, respectively, and the highest ferric reducing antioxidant power was 0.21. In conclusion, phycocyanin with high extraction yield and a considerable purity was efficiently extracted from Spirulina , which had higher antioxidant activity. This study laid a good foundation for the wide application of Spirulina phycocyanin in food and cosmetics industry.
