Abstract:The present study explored the role of dopaminergic transmission in spinal cord in a model of carrageenan-induced inflammatory pain by examining the effects of selective agonists and antagonists of dopamine receptors. The results were as follows: (1) trans-(-)-4aR-4,4a,5,6,7,8,8a,9-octahydro-5-propyl-1H-pyrazolo[3,4-g] quinoline hydrochloride (LY171555), a dopamine D(2) receptor agonist, produced anti-hyperalgesia (150 and 300 nmol) or hypoalgesia (300 nmol) in the inflamed hindpaws and non-inflamed hindpaws, respectively; spiperone hydrochloride (8-[4-(4-fluorophenyl)-4-oxobutyl]-1-phenyl-1,3,8-triazaspiro[4,5]decan-4-one hydrochloride), a dopamine D(2) receptor antagonist, decreased the pain threshold of non-inflamed hindpaws (300 nmol). (2) (+/-)-SKF38393 hydrochloride ((+/-)-1-phenyl-2,3,4,5-tetrahydro-(1H)-3-benzazepine-7,8-diol hydrochloride), a dopamine D(1) receptor agonist, had no effect on either hindpaw, even at a higher dose (300 nmol); R(+)-7-Chloro-8-hydroxy-3-methyl-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine hydrochloride (R(+)-SCH23390 hydrochloride), a dopamine D(1) receptor antagonist, induced anti-hyperalgesia in the inflamed hindpaws (300 nmol). The present results suggest that the dopaminergic system in the spinal cord is involved in the central modulation of inflammatory hyperalgesia, and that the different effects are probably induced by different receptors.
[Effects of Intrathecal Transient Receptor Potential Vanilloid 1 Blocker on Cervical Distension Induced Visceral Pain Response].

Abstract:Objective: With the aid of an acute visceral pain model of uterine cervical distension (UCD), the present study aimed to observe the effects of intrathecal administration of transient receptor potential vanilloid 1 (TRPV1) antagonist SB-366791 on UCD induced-visceral nociception as well as its involved molecular mechanisms. Methods: A total of 30 Sprague-Dawley-derived adult virgin female rats were used. UCD model was established under isoflurane inhalation anesthesia. Briefly, a lower abdominal incision at midline was made to expose the uteral cervix, two metal rods were inserted through both sides of the cervix separately, one rod was fixed and the other one was connected to a pulley system with application of manual weighted traction (0, 25, 50, 75 or 100 g) for simulating 1 h of cervical distension. In addition, 12 Sprague-Dawley-derived adult virgin female rats were subjected to intrathecal catheter implantation, and UCD was established 7 days later. The rats were divided randomly into two groups; one group was administrated with intrathecal SB-366791 while the other was administrated with the same volume of saline as control. The 75 g distension force was then applied for an hour and the electromyographic (EMG) of musculus rectus abdominis, heart rate as well as respiratory frequency were measured continuously during the surgery. The spinal cord (T(12)-L(2)) was collected 30 minutes after UCD for the detection of changes of c-FOS and TRPV1 expression. Results: UCD increased EMG activity (P<0.05) and c-FOS expression (P<0.05) in the deep dorsal horn region and central canal of the spinal cord (T(12)-L(2)) in a stimuli-dependent manner, the expression of TRPV1 in the T(12)-L(2) spinal cord also increased in response to UCD stimulation (P<0.05). Compared with the saline group, intrathecal SB-366791 significantly decreased EMG activity (P<0.05) as well as spinal c-FOS (P<0.05) expression induced by UCD. Conclusions: UCD in rats increases EMG activity of musculus rectus abdominis as well as spinal c-FOS and TRPV1 expression. Intrathecal administration of TRPV1 antagonist SB-366791 significantly decreases the visceral nociception induced by UCD.

Abstract:The level of serotonin (5-HT) and its major metabolite 5-hydroxyindoleacetic acid (5-HIAA) in dorsal horn of spinal cord and periaqueductal gray (PAG) were measured using in vivo microdialysis coupled with high performance liquid chromatography and electrochemical detection. Intraplantar injection of carrageenan was used to evoke release. Extracellular concentrations of 5-HT and 5-HIAA in spinal dorsal horn and PAG significantly increased following carrageenan inflammation. The peak occurred at 3 h and then gradually returned to baseline at 5-7 h. The carrageenan-induced release of 5-HT and 5-HIAA in. spinal cord rather than in PAG was decreased by intracerebroventricular (i.c.v.) injection: of naloxone, and both in spinal dorsal horn and PAG, the release was increased by i.c.v. injection of bicuculine. These results suggest that activity of 5-HT in spinal cord and PAG increases with carrageenan inflammation; the carrageenan-induced release of 5-HT and 5-HIAA in spinal cord may be tonically modulated by supraspinal opioid and GABA systems, whereas the release in PAG may only be tonically modulated by endogenous GABA system in supraspinal level. The role of 5-hydroxytryptamine1A and 5-hydroxytryptamine1B receptors in modulating spinal nociceptive transmission in normal and carrageenan-injected rats.

Abstract:Single unit extracellular recordings from the dorsal horn neurons were obtained with glass micropipettes in pentobarbital-anesthetized rats. A total of 115 wide dynamic range (WDR) neurons were studied in 94 rats. In normal rats, the size of nociceptive receptive fields (RFs) of WDR neurons was approximately 123.3±8.21 mm2 (n=88). Following carrageenan-induced inflammation, the RFs were markedly enlarged (332.4±30.1 mm2, n=27, P<0.001). The frequency of background activity of the WDR neurons in carrageenan-injected rats (11.3±2.1 imp/s, n=27) was greater than that in normal rats (7.1±0.8 imp/s, n=88, P<0.05). In 82% of WDR neurons in normal rats, there was a separation between the A- and C-responses. In contrast, in 67% of the neurons in carrageenan-injected rats, the response to suprathreshold electrical stimuli was a long train with no separation between the A- and C-responses. In carrageenan-injected rats, the magnitude and duration of the nociceptive responses were significantly increased compared to those in normal rats, and the average C-response threshold (7.7±1.1 mA, n=27) was lower than that in normal rats (10.4±0.7 mA, n=88, P<0.05). Intrathecal injection of the 5-hydroxytryptamine1A (5-HT1A) receptor agonist 8-hydroxy-DPAT hydroxybromide (8-OH-DPAT) (0.305, 1.525, 3.05, and 15.25 mM) dose-dependently increased Aδ- and C-responses and post-discharge in most of the WDR neurons. Following carrageenan-induced inflammation, the 8-OH-DPAT-induced facilitatory effect on Aδ- and C-responses and post-discharge was significantly enhanced (P<0.05). Intrathecal injection of the 5-hydroxytryptamine1B (5-HT1B) receptor agonist CGS12066A (0.222, 1.11, 2.22, and 11.1 mM) dose-dependently enhanced the C-response and post-discharge without influencing the Aδ-response. In carrageenan-injected rats, CGS12066A not only enhanced the facilitatory effect on the C-response and post-discharge, but also facilitated the Aδ-response. Intrathecal injection of the 5-HT1A receptor antagonist NAN-190 (0.2 mM) alone did not influence Aδ- and C-responses and post-discharge of WDR neurons in normal rats. When 0.2 mM NAN-190 was co-administered with 3.05 mM 8-OH-DPAT, the facilitatory effect of 8-OH-DPAT on Aδ- and C-responses and post-discharge was completely antagonized, whereas CGS12066A-induced facilitation on the C-response and post-discharge was not influenced by co-administration of 0.2 mM NAN-190 and CGS12066A. These data suggest that 5-HT1A and 5-HT1B receptor subtypes mediate the facilitatory effect of 5-HT on nociceptive processing in the spinal cord of rats. The excitability of dorsal horn WDR neurons and the sensitivity of the neurons to intrathecal 5-HT1A and 5-HT1B receptor agonists might increase following carrageenan-induced inflammation.
The Spinal Antinoceptive Effects of the Capsaicin Administrated Subarachnoidly in the Two Phases of the Inflammation Pain

Abstract:Objective To evaluate the effects of the capsaicin administrated subarachnoidly in the two phase of the inflammation pain.Methods The rats having no functional disability were divided randomly into two groups which had been implanted with chronic lumbar intrathecal catheters, the vehical and capsaicin(0.25%) groups.Then the mechanism threshold of the pain and the thermal latency at the time of the forth and the seventh day were investigated.After the intraplantar of the carrageen,the threshold of the two groups were investigated also at the time of the third and fifteenth hour.After the record of the threshold,the skin of the paw was taken and dyed with HE.Results There was no difference between the threshold of the preintrathecal and the prointrathecal.After administrated the medicine,mechanism threshold and the thermal latency of the capsaicin group were increased distinguishedly.Compared that at the forth day with the seventh day,the value at the seventh day increased greatly.After the inflammation the threshold of the right hind paw was lowed compared with the left hind paw,that at the late phase of the inflammation were higher than that at the early phase.The paw skin in the vehical group was more inflamed than taht in the capsaicin group.Conclusion The capsaicin can elevated the mechanism threshold and the thermal latency significantly for several days.
Excitatory amino acid receptor antagonists and electroacupuncture synergetically inhibit carrageenan-induced behavioral hyperalgesia and spinal fos expression in rats

Abstract:The interaction between electroacupuncture and an N-methyl-D-aspartic acid (NMDA) receptor antagonist, (DL-2-amino-5-phosphonopentanoic acid; AP5), or an (+/-)-alpha-Amino-3-hydroxy-5-methylisoxazole-4-propionic acid/kainite (AMPA/KA) receptor antagonist, (6,7-dinitroquinoxaline-2,3 (1H,4H); DNQX) administered intrathecally on carrageenan-induced thermal hyperalgesia and spinal c-Fos expression was investigated. The latency of paw withdrawal (PWL) from a thermal stimulus was used as a measure of hyperalgesia in awake rats. Intrathecal (i.t.) injection of 1 and 10 nmol AP5, but not DNQX, markedly increased the PWL of the carrageenan-injected paw. At a dose of 100 nmol, either AP5 or DNQX significantly increased the PWL of carrageenan-injected paw, with AP5 being more potent. The PWLs of the non-injected and normal saline (NS)-injected paws were not detectably affected by the administration of NMDA or AMPA/KA receptor antagonists at the doses tested. Unilateral electroacupuncture stimulation of the 'Zu-San-Li' (St 36) and 'Kun-Lun' (UB 60) acupuncture points (60 and 2 Hz alternately, 1-2-3 mA) contralateral to the carrageenan-injected paw significantly elevated the PWLs of carrageenan- and NS-injected paws. Although neither i.t. injection of 0.1 nmol AP5 nor 1 nmol DNQX alone had an effect on the PWL of the carrageenan- and NS-injected paws, both significantly potentiated electroacupuncture-induced analgesia in carrageenan-injected rats, especially 0.1 nmol AP5. Fos expression evoked by intraplantar (i.pl.) injection of carrageenan was examined in the spinal cord with immunohistochemical methods. Three hours after i.pl. injection of carrageenan, the number of Fos-like immunoreactive (Fos-LI) neurons was significantly increased in all the layers of the ipsilateral spinal cord at L(4-5), with the highest density in laminae I-II and V-VI. Intrathecally pre-administered AP5 (10 nmol) or DNQX (100 nmol) significantly reduced the total number of carrageenan-induced Fos-LI neurons. The reduction was most apparent in laminae I-II and IV-V. Similarly, following bilateral electroacupuncture stimulation of the 'Zu-San-Li' and 'Kun-Lun' acupuncture points, the numbers of carrageenan-induced Fos-LI neurons in laminae I-II and V-VI were also markedly reduced. When a combination of electroacupuncture with 10 nmol AP5 or 100 nmol DNQX was used, the level of Fos expression in the spinal cord induced by carrageenan was significantly lower than electroacupuncture or i.t. injection of AP5 or DNQX alone. Effect of intraperitoneal injection of Lidocaine on expression of NMDAR1 in spinal dorsal horn of a rat model of incisional pain

Abstract:Objective To investigate the effect of intraperitoneal injection of Lidocaine on pain behavior and expression of N-Methyl-D-aspartate(NMDA) receptor-1(NR1) in spinal dorsal horn of a rat model of incisional pain.Method Total 30 adult male SD rats(weight 200.0~250.0 g) were randomly divided into five groups,6 rats in each group:group A(normal control group);group B(operation group):only a plantar incision was made;group C:Lidocaine 5.0 mg/kg was given intraperitoneally after preparation of a successful model of incisional pain;group D:Lidocaine 10 mg/kg was given intraperitoneally after preparation of a successful model of incisional pain;group E:Lidocaine 20 mg/kg was given intraperitoneally after preparation of a successful model of incisional pain.Except group A,an incision of 1 cm long was made in the planter region of the rats in other 4 groups under pentobarbital anesthesia according to the methods of Brennan.Pain behaviors was assessed using cumulative pain score 5 min after intraperitoneal injection of Lidocaine.One hour later,rats were deeply anesthetized and the spinal cord was immediately removed and sliced and examined for NR1 expression using immunohistochemistry.Results Compared with group A,the cumulative pain score and the expression of NR1 in spinal dorsal horn were increased significantly after plantar incision in group B,C,D and E(P<0.01).Compared with group B,the cumulative pain scores were decreased significantly in group D and E(P<0.05),and the expression of NR1 in spinal dorsal horn were also significantly decreased in group D and E(P<0.05).Conclusion In the rat model of incisional pain,Lidocaine 10 mg/kg intraperitoneal injection can alleviate pain behavior and the increased concentration of Lidocaine will not have much better effect,and the results also suggest that the pain alleviation effect of intraperitoneal injection of Lidocaine is related to its suppression of the expression of NR1.
Distinct Effects of D-Serine on Spinal Nociceptive Responses in Normal and Carrageenan-Injected Rats

Abstract:Single unit extracellular recordings from dorsal horn neurons were performed with glass micropipettes in pentobarbital-anesthetized rats. A total of 60 wide dynamic range (WDR) neurons were obtained from 34 rats. In normal rats (20/34), spinally administered D-serine (10 nmol), a putative endogenous agonist of glycine site of NMDA receptors, significantly enhanced the C- but not Abeta-, and Adelta-fiber responses of WDR neurons in the spinal dorsal horn. When 1 nmol of the glycine site antagonist 7-chlorokynurenic acid (7-CK) was co-administered with 10 nmol D-serine, the facilitation of D-serine on C-fiber response was completely blocked. 7-CK (1 nmol) alone failed to influence Abeta-, Adelta-, and C-fiber responses of WDR neurons. In contrast, in carrageenan-injected rats (14/34), 10 nmol D-serine had no effect on C-fiber response, while 1 nmol 7-CK per se markedly depressed C-fiber response of WDR neurons. These findings suggest that under physiological conditions, glycine sites in the spinal cord were available but became saturated following peripheral inflammation. Thus, increased endogenous d-serine or glycine may be involved in nociceptive transmission by modulating NMDA receptor activities. The glycine site of NMDA receptors may become a target for the prevention of inflammatory pain.

Antinflammatory pain effect of ifenprodil administrated subarachnoidly

Abstract:Objective To evaluate the effects of N-Molhyl-d-Aspartate(NMDA) receptors subunit NR2B antagonist(ifenprodil) administrated subarachnoidly at two phases of inflammatory pain.Methods The rats implanted with chroinc lumbar intrathecal catheters were divided randomly into control(N),model(M),ketamine(K) and ifenprodil(I) groups.The rats in group N(received) normal saline in the right hind paw,but the other groups received 2% carrageenan 100μl.Three hours after injection of carrageenan the drugs were injected in group K and I,and normal saline was injected into the subarachnoid space in group N and M.The mechanical threshold of pain and the thermal latency were observed at 3 and 15 hours after inflammation occureence.Results From the model groups,the mechanical threshold and thermal latency at two phases were reduced significantly in group M more than those in group N,K and I(P<0.05).Ketamine could attenuate pain at the late phase(P<0.05),but the ifenprodil did it at two phases(P<0.05).Conclusion Both ketamine and ifenprodil could(elevate) pain threshold of the inflammation with different mechanism and latency.Intrathecal ifenprodil has better analgesic effect than ketamine.
Involvement of spinal neurokinin-1 receptors in the maintenance but not induction of carrageenan-induced thermal hyperalgesia in the rat.

Abstract:The study was undertaken to assess the antihyperalgesic effect of L-732,138, (N-acetyl-l-tryptophan-3,5-bistrifluoromethyl benzyl ester), a non-peptide neurokinin-1 (NK1) receptor antagonist in rats when given intrathecally. The peripheral inflammation associated with behavioral hyperalgesia to a thermal stimulus was induced by intraplantar (i.pl.) injection of carrageenan. The thermal hyperalgesia was measured by paw withdrawal latency. Intrathecal (i.t.) injection of L-732,138 (100nmol) at 3h after carrageenan markedly attenuated the paw withdrawal latency of the inflamed paw, but not that of the non-inflamed paw. L-732,138 (100nmol, i.t.) given 10min prior to carrageenan injection had no effect on the carrageenan-induced decrease in paw withdrawal latency to noxious thermal stimulus. The results demonstrate that NK1 receptor is involved in the maintenance but not the induction and development of thermal hyperalgesia evoked by carrageenan.

Abstract:The present study investigated the effects of interleukin-1β (IL-1β) injected intrathecally (i.t.) on carrageenan-induced thermal hyperalgesia and spinal c-Fos expression. The paw withdrawal latency (PWL) by the thermal stimulus was taken as an index of the thermal hyperalgesia of rats. I.t. injection of 10 ng IL-1β significantly increased the PWL of the carrageenan-injected paw. Expression of c-Fos induced by intraplantar (i.pl.) injection of carrageenan was examined in the spinal cord with immunohistochemical methods. Three hours after i.pl. injection of carrageenan, the number of c-Fos-like immunoreactive (c-Fos-LI) neurons was significantly increased in laminae I–II, III–IV and V–VI of the ipsilateral spinal cord at L4–5 with the higher density in laminae I–II and V–VI. I.t. pre-injected IL-1β significantly decreased the number of carrageenan-induced c-Fos-LI neurons in laminae I–II in the ipsilateral spinal cord and also inhibited the hyperalgesia induced by i.pl. carrageenan. These results suggested that i.t. injection of IL-1β suppressed the central nociceptive input into laminae I–II and produced an antinociceptive effect.
Activated Spinal Astrocytes Contribute to the Later Phase of Carrageenan-Induced Prostatitis Pain.

Abstract:Background Prostatodynia is the main symptom of chronic prostatitis and the main reason that patients go to the hospital for treatment. Although a variety of factors, including inflammatory immune response, nervous system sensitization, and psychological factors, have been shown to play important roles in the induction and development of chronic pain in prostatitis, the underlying cause of chronic prostatodynia maintenance in prostatitis patients remains unclear. Methods A mouse model of chronic prostatitis induced by carrageenan injection was used. The von Frey test was used to measure pain behavior. The microglial and astrocyte activations were immunohistochemically demonstrated with antibodies against Iba1 and GFAP. The expression of cytokine or signaling pathway was detected by enzyme-linked immunosorbent assay (ELISA) and Western blotting. Results In this study, we provide several lines of evidence to demonstrate that activated spinal astrocytes contribute to the later phase (5 weeks after carrageenan injection) of carrageenan-induced prostatitis pain. First, activation of spinal astrocytes but not microglia was found in the spinal cord dorsal horn at 5 weeks. Second, intrathecal injection of the astroglial toxin L-2-Aminoadipate acid (L-AA) but not microglial inhibitor minocycline reduced mechanical allodynia at 5 weeks. Third, chronic prostatitis induced a profound and persistent upregulation of connexin-43 hemichannels in spinal astrocytes, and spinal injection of the connexin-43 inhibitor carbenoxolone (CBX) effectively reduced pain symptoms. Fourth, increased expression and release of chemokine C-X-C motif ligand 1 (CXCL1) in the spinal dorsal horn and intrathecal injection of a CXCL1 neutralizing antibody or the CXCR2 (a major receptor of CXCL1) antagonist SB225002 significantly reduced mechanical allodynia at 5 weeks. Conclusions In this study, we found that a novel mechanism of activated spinal astrocytes plays a crucial role in maintaining chronic prostatitis-induced persistent pain via connexin-43-regulated CXCL1 production and secretion.
Peripheral NMDA and non-NMDA receptors contribute to nociception: an electrophysiological study

Abstract:The present study investigated the effects of peripheral administration of N-methy-D-aspartate (NMDA) and non-NMDA receptor antagonists on C-fiber evoked responses of the spinal dorsal horn neurons in the spinalized rats. When DL-2-amino-5-phosphonovaleric acid (AP5) (10 mM, 1 mM, 0.1 mM, 20 μl) or 6,7-dinitroquinoxaline-2, 3-dione (DNQX) (1 mM, 0.1 mM, 0.01 mM, 20 μl) was subcutaneously injected into the receptive field on the hindplantar region, C-fiber evoked responses of the dorsal horn neurons were profoundly inhibited in a dose-dependent manner. Three hours after subcutaneous injection of carrageenan into the ipsilateral hindpaw, NMDA and non-NMDA antagonist-induced inhibition of C-fiber evoked responses was more potent than that in the normal rat (Student’s t-test, p < 0.05). In the carragenan-treated rats, DNQX-induced inhibition was stronger than AP-5-induced one (Student’s t-test, p < 0.05). The results suggest that peripheral NMDA and non-NMDA receptors are involved in mediating excitation of nociceptors.

We also studied the co-localization of 5-HT1A receptor mRNA and gamino butyric acid (GABA) or enkephalin (ENK) immunoreactivities using a combined fluorescent ISH and immunofluorescent histochemical double-staining technique. The finding of this study demonstrated that 5-HT1A receptor mRNA was widely distributed in the spinal dorsal horn with the highest density in laminae III–VI. Following carrageenan-induced inflammation, the 5-HT1A receptor mRNA expression in all layers of ipsilateral dorsal horn was significantly enhanced, and the peak occurred after 8 h. Furthermore, the number of 5-HT1A receptor mRNA and GABA or ENK immunoreactive double-labeled cells was also markedly increased 8 h after carrageenan injection. These findings suggested that following peripheral inflammation, the synthesis of 5-HT1A receptor was increased in the lumbar spinal dorsal horn neurons, especially in spinal GABA and ENK neurons. q 2002 International Association for the Study of Pain. Peripheral Inflammation Alters Desensitization Of Substance P-Evoked Current In Rat Dorsal Root Ganglion Neurons

Abstract:The neuropeptide substance P is synthesized in a proportion of neurons of the peripheral and central nervous system, and the receptor for substance P, tachykinin NK1 receptor, has been identified in numerous areas of the central nervous system including the spinal cord. The present investigation was to confirm the existence of tachykinin NK1 receptor on rat dorsal root ganglion (DRG) neurons and characterize the adaptation of inward current evoked by substance P during carrageenan-induced peripheral inflammation. Using whole-cell voltage recording technique, our results demonstrated that 1 mu M substance P elicited significant inward current in a small population of small-diameter DRG neurons of control rats (7%, n = 218) and in a bigger proportion of DRG neurons of carrageenan-inflamed rats (15%, n = 203). Desensitization of substance P-evoked current was exhibited in almost all of substance P-responsive DRG neurons in control rats (94%, n = 16), while it diminished in a bigger proportion of DRG neurons (35%, n = 31) in inflamed rats. Furthermore, desensitization of substance P-evoked current was recovered by the activation of protein kinase C (PKC) by application of phorbol 12,13-dibutyrate, a selective agonist of PKC, in the DRG neurons of the inflamed rats. All these results indicated that tachykinin NK1 receptor in the primary afferents might contribute to the development and maintenance of inflammatory pain. (C) 2011 Elsevier B.V. All rights reserved.

Abstract:The present study was to investigate the effect of intrathecal (i.t.) injection of interleukin-ID (IL-1beta) on nociception in normal and inflammatory rats. Peripheral inflammation was induced by intraplantar injection (i.pl.) of carrageenan into unilateral hind paw. The nociceptive threshold to noxious thermal stimulation was measured by the paw withdrawal latency (PWL). Intrathecal injection of IL-1beta (10 ng, 100 ng) significantly increased PWL in normal rats, the peak occurred at 5 min and the effect lasted for 30 min. Similarly, IL-1beta (10 ng, 100 ng, i.t.) significantly increased the PWL and lasted for more than 60 min in inflammatory rats. Both in normal and inflammatory rats, the IL-1beta-induced antinociceptive effect was completely abolished by IL-1ra (50 ng, i.t.), and apparently attenuated by naloxone (10 mug, i.t.) or mianserin (20 mug, i.t.). These results suggest that IL-1beta produces antinociceptive effect by binding IL-1 receptor at the spinal level, and is related to the activation of opioid and 5-HT systems. Activation of silent mechanoreceptive cat C and Aδ sensory neurons and their substance P expression following peripheral inflammation

Abstract:1. The effect of inflammation on the excitability and the level of substance P (XP) in cat mechanoreceptive C and A delta dorsal root ganglion (DRG) neurons were studied in vivo using intracellular recording and immunocytochemical techniques. 2. Following injections of carrageenan (Carg) into the cat hindpaw, the percentage of C neurons exhibiting spontaneous activity increased from 7.2 to 20.7% and the percentage of AS neurons increased from 6.9 to 18.6%. In contrast to most cells from normal, cats, which fired regularly below 10 Hz, many cells from Carg-treated cats fired at higher frequencies or in bursts. 3. Inflammation (Carg treatment) also depolarized membrane potentials, increased membrane input resistance, caused the disappearance of inward rectifying currents and lowered the mean current thresholds of tibial nerve-evoked responses in DRG neurons. 4. With inflammation, the percentage of C or A delta neurons responding to lon threshold mechanoreceptive stimuli increased (C neurons: normal, 13%; inflamed, 41%, A delta neurons: normal, 13 %; inflamed, 39 %), while the percentage of C or A delta neurons responding to high threshold mechanoreceptive stimuli remained unchanged. 5. Some receptive field (RF)-responsive cells were injected with Lucifer Yellow and their XP immunoreactivity was determined. Following Carg treatment, substantially higher percentages of RF-responsive cells were XP positive (C neurons: normal, 35.7%; inflamed, 60%; A delta neurons: normal, 18.2%; inflamed, 66.7%). 6. These combined increases in the excitability of DRG neurons and SP-containing RF responsive neurons could lead to sensitization of sensory neurons, thus contributing to the development of hyperalgesia.

Abstract:In the present study, we investigated the anti-nociceptive effect and the underlying mechanism of the analgesic-antitumor peptide (AGAP), a neurotoxin from the scorpion Buthus martensii Karsch. AGAP in doses of 0.2, 1 and 5 mu g was injected intraplantarly (i.pl.) before formalin injection 10 min at the same site. The suppression by intraplantar injection of AGAP on formalin-induced spontaneous nociceptive behaviors was investigated. The results show that AGAP could dose-dependently inhibit formalin-induced two-phase spontaneous flinching response. To investigate the mechanism of action of treatment with AGAP in inflammatory pain, the expressions of peripheral and spinal phosphorylated mitogen-activated protein kinases (phospho-MAPKs) including p-p38, p-ERK and p-JNK were examined. We found that formalin increased the expressions of peripheral and spinal MAPKs, which were prevented by pre-intraplantar injection of AGAP in inflammation pain model in mice. AGAP could also decrease the expression of spinal Fos induced by formalin. Furthermore, combinations the lower doses of the inhibitors of MAPKs (U0126, SP600125, or SB203580 0.1 mg) with the lower dose of AGAP (0.2 mu g), the results suggested that AGAP could potentiate the effects of the inhibitors of MAPKs on the inflammatory pain. The present results indicate that pre-intraplantar injection of AGAP prevents the inflammatory pain induced by formalin through a MAPKs-mediated mechanism in mice.
