Laboratory Study on the Protective Effect and the Mechanism of DWN on Gastric Mucosal Damage

Abstract:Objective: To study the protective effect and the mechanism of Chinese herbal medicine Dan-Wei-Ning(DWN) on alcohol-induced gastric mucosal damage. Methods: Forty SD rats were randomly divided into five groups: control group, model group and three DWN-treated groups. The rats in three treated groups were given 1 ml, 2 ml and 4 ml of DWN respectively by intragastric administration, while the rats in control group and model group were given 1 ml of normal saline. Two hours later all rats except those in the control group were given 1 ml of absolute alcohol intragastrically. After another two hours, the rats were sacrificed and their stomachs were taken out to observe the mucous injury and determine the ulcer index and inhibition rate. Pathological changes were observed under microscope. Maleic dialdehyde (MDA), superoxide dismutase (SOD), and prostaglandin E2 (PGE2) were determined in the gastric homogenate. Results: DWN could lessen gastric mucous injury, inhibit ulcerous formation and improve pathological changes of damaged gastric mucosa induced by alcohol. DWN potently inhibited both decrease of SOD and PGE2 and increase of MDA in damaged gastric mucosa induced by ethanol in a dose-dependent manner. Conclusion: DWN has a potent protective effect on ethanol-induced gastric mucosal injury by the reduction in lipid peroxidation, augmentation in the antioxidant activity and increase in PGE2.
