Effects of Human Tissue Kallikrein Gene on Activities of AMP-activated Protein Kinase in Diabetic Rats

Abstract:Objective To investigate the effects of human tissue kallikrein (HK) gene on activities of AMP-activated protein kinase in streptozotocin (STZ)-induced diabetic rats. Methods Forty-eight male Wistar rats were divided into control and experimental groups randomly. Wistar rats in experimental group were injected with low dose STZ (25 mg/kg) and fed with diets enriched in both glucose and fat to establish type 2 diabetic models, then subdivided into 2 subgroups randomly (each having 8 diabetic rats). Recombinant adeno-associated viruses expressing HK gene (HK subgroup) or Lacz gene (Lacz subgroup) were injected into diabetic rats through tail veins. All rats were killed after 12 weeks. The expression of HK protein in rats and the phosphorylation of AMPK were detected by Western blot. Results The expression of HK protein was detected in liver, kidney and heart of diabetic rats in the HK subgroup. The expression of HK protein in HK subgroup was increased significantly as compared with that in Lacz group, and there was no significant difference in the expression of AMPK between the groups in liver, kidney, heart, muscle and adipose. The phosphorylation level of AMPK in HK subgroup was significantly higher than in Lacz subgroups, but there was no significant difference between Lacz subgroup and control group. Conclusion Recombinant adeno-associated viruses-mediated HK gene delivery can promote the expression of AMPK in major organs of type 2 diabetic rats.
