Abstract:ObjectiveTo isolate nestin positive cells from human fetal pancreas and to detect their capability of proliferation and differe ntiation into pancreatic endocrine cells in vitro.MethodsThe islet like cell clusters (ICCs) were isolated from human fetal pancreas with c ollagenase digestion. The free floating ICCs were hand picked up and cultured in a new dish. After the ICCs developed into the monolayer epithelium like cell s, they were passaged and induced to differentiate. Reverse transcription polyme rase chain reaction (RT PCR), immunofluorescence stain and radioimmunoassay (RI A) were used to detect the expression of their molecular markers and to identify their capability of differentiating into pancreatic endocrine cells in vitro. Results (1) The monolayer epithelium like cells had high prol iferative potential and could be passaged more than 16 times in vitro. (2) RT P CR and immunofluorescence stain analysis showed that these cells expressed nesti n and ABCG2, two of the markers of stem cells. (3) RT PCR analysis showed that the mRNA expressions of insulin, glucagon and pancreatic duodenal homeobox gene 1 (PDX 1) were detected,whereas the expressions of nestin and neurogenin 3 ( Ngn3) disappeared in the nestin positive cells treated with several cytokines a nd without serum. Furthermore, the intra cellular insulin content was detected by RIA after the induction. ConclusionNestin positive cells isolated from the human fetal pancreas possess the characteristics of pancreati c progenitor cells, and have high proliferative potential and the capability of differentiation into insulin producing cells in virto. This holds some promise of a new cell source for islet transplantation.

Abstract:AIM:To isolate nestin-positive progenitor cells from human fetal pancreas and to detect their surface markers and their capability of proliferation and differentiation into pancreatic islet endocrine cells in vitro.METHODS:Islet-like cell clusters (ICCs) were isolated from human fetal pancreas by using collagenase digestion. Furthermore, the intra-cellular insulin content was detected by RIA after the induction culture.CONCLUSION:Nestin-positive cells isolated from human fetal pancreas possess the characteristics of pancreatic progenitor cells since they have highly proliferative potential and the capability of differentiation into insulin-producing cells in vitro. Interestingly, the nestin-positive pancreatic progenitor cells share many phenotypic markers with mesenchymal stem cells derived from bone marrow.

Abstract:Objectives: To identify whether Nestin-positve cells or Nestin-negative cells in pancreas enrich potential pancreatic stem/progenitor cells.Methods: We generated transgenic mice carrying enhanced green fluorescent protein (EGFP) under the control of the nestin second-intronic enhancer and subsequently divided their embryonic pancreatic cells into different subpopulations according to the expression of EGFP and CD24 and characterized these subpopulations by in vitro culture.Results: The EGFP expression correlated well with that of endogenous Nestin. Only the Nestin(neg)CD24(low/-) subpopulation was able to proliferate and generate immature islet-like cell clusters in long-term culture. Immature islet-like cell clusters could be induced to differentiate into insulin-, glucagon-, and somatostatin-positive cells.Conclusions: Pancreatic endocrine stem/progenitor cells are enriched in the Nestin(neg) CD24(low/-) population of embryonic pancreas.
Reversible immortalization of Nestin-positive precursor cells from pancreas and differentiation into insulin-secreting cells.

Abstract:Pancreatic stem cells or progenitor cells posses the ability of directed differentiation into pancreatic β cells. However, these cells usually have limited proliferative capacity and finite lifespan in vitro. In the present study, Nestin-positive progenitor cells (NPPCs) from mouse pancreas that expressed the pancreatic stem cells or progenitor cell marker Nestin were isolated to obtain a sufficient number of differentiated pancreatic β cells. Tet-on system for SV40 large T-antigen expression in NPPCs was used to achieve reversible immortalization. The reversible immortal Nestin-positive progenitor cells (RINPPCs) can undergo at least 80 population doublings without senescence in vitro while maintaining their biological and genetic characteristics. RINPPCs can be efficiently induced to differentiate into insulin-producing cells that contain a combination of glucagon-like peptide-1 (GLP-1) and sodium butyrate. The results of the present study can be used to explore transplantation therapy of type I diabetes mellitus.

Abstract:The islets were isolated from ICR mice by using the intraductal collagenase technique,then nestin+ cells were separated and expanded.Those cells were induced into medium containing low glucose concentration(2.5 mmol/L) and low serum(2%).The results indicated that mouse islets contain nestin+ cells.After isolation,they can differentiate into islet-like structure at low glucose concentration(2.5 mmol/L) and low serum(2%) conditions in long-term culture,demonstrating that nestin+ mouse islet cells may have the biological properties of pancreas precursor cells.

Abstract:Objective To explore the isolation, culture of nestin positive cells of the pancreas in newborn rats. Methods The whole pancreas of neonatal rats were digested with collagenase, followed by incubation under pH 7. 6 serum RPMI 1640 for 24 - 36 h and then under pH 7. 4 serum free RPMI 1640 (bFGF.EGF 20 ng/ml.1% N2) for 18-24 d. The expression of insulin.glucagon and nestin were detected by immunofluorescence technique. Nestin.CK19 were detected by RT-PCR. Resides Cells attached to the surface of plates after 24 h incubation under pH 7. 6 condition, and islet-like masses were obtained after 18 -24 d incubation. A monolayer of cells grew out after 24 h of passage of islet-like masses. Nestin positive cells was detected after 24 - 36 h incubation, with no expression of insulin and glucagon. Positive cells of insulin and glucagon were detected in islet-like masses after 24 h passage. Nestin positive cells were detected by RT-PCR in islet-like masses after 24 h passage, but no CK19. Conclusion Insulin and glucagon were expressed in islet-like masses after passage. Nestin positive cells in the pancreas of neonatal rats possessed character of islet stem cells.

Abstract:Objective To establish the method of isolation,culture, and differentiation of Nestin positive cells of non-islet cells in adult rats in vitro. Methods The whole pancreas of three adult rats were digested in vitro with the intraductal collagenase distention method, then both islets and non-islets were collected and cultured with 10% fetal bovine serum (pH 7.6) and serum-free RPMI 1640 (pH 7.4).The formation process of new islet-like cell masses were examined by light microscopy,and the glucose-stimulated insulin release (SI) from 40 just isolated islets and 40 new produced islet-like cell masses were tested.The expression of Nestin was tested by immunocytochemistry. Results Functioning islets in good morphology could be obtained from pancreas of adult rats by the intraductal collagenase distention method;there were adherent cells after 36 h culture,but no islets; the Nestin positive cells proliferated fast and formed new islet-like cell masses after 18-22 d. Nestin expression could be confirmed among adherent cells,but no insulin and glucagon, and Nestin positive cells can be seen in passage cells of new produced islet-like cell masses. Insulin secretion levels in 40 new isolated islets were (63.6±4.0), (202.2±14.8)mIU·L~ -1 in 3.3 and 16.7 mmol·L~ -1 glucose, respectively;and (3.5±0.2), (3.3± 0.2)mIU·L~ -1, respectively, in 40 new produced islet-like cell masses (P0.001).Conclusion Functioning islets can be obtained from the pancreas of adult rats. Nestin positive cells of non-islet cells can be obtained from pancreas of adult rats in pH 7.6 condition through culture in vitro,and can form islet-like cell masses;Nestin positive cells of non-islet cells in pancreas of adult rats possess the characters of islet stem cells.

Abstract:Pancreatic β and α cells play essential roles in maintaining glucose homeostasis. However, the mechanisms by which these distinct cell populations are generated, expand, and mature during pancreas development remain unclear. In this study, we addressed this critical question by performing a single-cell transcriptomic analysis of mouse β and α cells sorted from fetal to adult stages. We discovered that β and α cells use different regulatory strategies for their maturation and that cell proliferation peaks at different developmental times. However, the quiescent and proliferative cells in both the β lineage and α lineage are synchronous in their maturation states. The heterogeneity of juvenile β cells reflects distinct cell-cycling phases, origins, and maturation states, whereas adult β cells are relatively homogeneous at the transcriptomic level. These analyses provide not only a high-resolution roadmap for islet lineage development but also insights into the mechanisms of cellular heterogeneity, cell number expansion, and maturation of both β and α cells.

Abstract:Objective:To study the expression of nestin and its mRNA in pancre as of different phases, including the embryonic, newborn and adult rats. Methods: The mRNA of nestin was detected by RT-PCR,the number of nestin-positive cells and the levels of nestin protein were accessed by the methods of immunohistoche mistry, and the results were analyzed by the statistical methods. Results:Nestin was expressed in E18.5,newborn and adult phases of rats. And nestin positive ce lls were mainly located in the islets and sparsely in the periacinar. Conclusion : The level of mRNA and protein of nestin in E18.5 was higher than that in new b orn and adult phase. The positive cells in islets in E18.5 was higher than adult phase. The nestin positive islet-derived cell in E18.5 may be a better resourc e for the research of stem cell than adult rat. [

Abstract:Human embryonic stem cells (hESCs) are pluripotent and capable of undergoing multilineage differentiation into highly specialized cells including pancreatic islet cells. Thus, they represent a novel alternative source for targeted therapies and regenerative medicine for diabetes. Significant progress has been made in differentiating hESCs toward pancreatic lineages. One approach is based on the similarities of pancreatic β cell and neuroepithelial development. Nestin-positive cells are selected as pancreatic β cell precursors and further differentiated to secrete insulin. The other approach is based on our knowledge of developmental biology in which the differentiation protocol sequentially reproduces the individual steps that are known in normal β cell ontogenesis during fetal pancreatic development. In the present study, the hESC cell line PKU1.1 was induced to differentiate into insulin-producing cells (IPCs) using both protocols. The differentiation process was dynamically investigated and the similarities and differences between both strategies were explored. Our results show that IPCs can be successfully induced with both differentiation strategies. The resulting IPCs from both protocols shared many similar features with pancreatic islet cells, but not mature, functional β cells. However, these differently-derived IPC cell types displayed specific morphologies and different expression levels of pancreatic islet development-related markers. These data not only broaden our outlook on hESC differentiation into IPCs, but also extend the full potential of these processes for regenerative medicine in diabetes.

Abstract:Progenitor cells in the adult pancreas are potential sources of endocrine beta cells for treating type 1 diabetes. Previously, we identified tri-potent progenitor cells in the adult (2-4 month-old) murine pancreas that were capable of self-renewal and differentiation into duct, acinar, and endocrine cells in vitro. These progenitor cells were named pancreatic colony-forming units (PCFUs). However, because PCFUs are a minor population in the pancreas (similar to 1%) they are difficult to study. To enrich PCFUs, strategies using cell-surface marker analyses and fluorescence-activated cell sorting were developed. We found that CD133(highC)D71(low) cells, but not other cell populations, enriched PCFUs by up to 30 fold compared to the unsorted cells. CD133(highC)D71(low) cells generated primary, secondary, and subsequent colonieswhen serially re-plated in Matrigel-containing cultures, suggesting self-renewal abilities. In the presence of a laminin hydrogel, CD133(highC)D71(low) cells gave rise to colonies that contained duct, acinar, and Insulin(+) Glucagon(+) double-hormonal endocrine cells. Colonies from the laminin hydrogel culture were implanted into diabetic mice, and five-weeks later duct, acinar, and Insulin(+) Glucagon(-) cells were detected in the grafts, demonstrating tri-lineage differentiation potential of CD133(highC)D71(low) cells. Abstract:Pancreatic endocrine cells, which are clustered in islets, regulate blood glucose stability and energy metabolism. The distinct cell types in islets, including insulin-secreting β cells, are differentiated from common endocrine progenitors during the embryonic stage. Immature endocrine cells expand via cell proliferation and mature during a long postnatal developmental period. However, the mechanisms underlying these processes are not clearly defined. Single-cell RNA-sequencing is a promising approach for the characterization of distinct cell populations and tracing cell lineage differentiation pathways. Here, we describe a method for the single-cell RNA-sequencing of isolated pancreatic β cells from embryonic, neonatal and postnatal pancreases.

Abstract:Pancreatic β cells are of great interest for the treatment of type 1 diabetes. A number of strategies already exist for the generation of β cells, but a general approach for reprogramming nonendodermal cells into β cells could provide an attractive alternative in a variety of contexts. Here, we describe a stepwise method in which pluripotency reprogramming factors were transiently expressed in fibroblasts in conjunction with a unique combination of soluble molecules to generate definitive endoderm-like cells that did not pass through a pluripotent state. These endoderm-like cells were then directed toward pancreatic lineages using further combinations of small molecules in vitro. The resulting pancreatic progenitor-like cells could mature into cells of all three pancreatic lineages in vivo, including functional, insulin-secreting β-like cells that help to ameliorate hyperglycemia. Our findings may therefore provide a useful approach for generating large numbers of functional β cells for disease modeling and, ultimately, cell-based therapy.

Abstract:Embryonic stem (ES) cell technology may serve as a platform for the discovery of drugs to treat diseases such as diabetes. However, because of difficulties in establishing reliable ES cell differentiation methods and in creating cost-effective plating conditions for the high-throughput format, screening for molecules that regulate pancreatic beta cells and their immediate progenitors has been limited. A relatively simple and inexpensive differentiation protocol that allows efficient generation of insulin-expressing cells from murine ES cells was previously established in our laboratories. In this report, this system is characterized in greater detail to map developmental cell stages for future screening experiments. Our results show that sequential activation of multiple gene markers for undifferentiated ES cells, epiblast, definitive endoderm, foregut, and pancreatic lineages was found to follow the sequence of events that mimics pancreatic ontogeny. Cells that expressed enhanced green fluorescent protein, driven by pancreatic and duodenal homeobox 1 or insulin 1 promoter, correctly expressed known beta cell lineage markers. Overexpression of Sox17, an endoderm fate-determining transcription factor, at a very early stage of differentiation (days 2-3) enhanced pancreatic gene expression. Overexpression of neurogenin3, an endocrine progenitor cell marker, induced glucagon expression at stages when pancreatic and duodenal homeobox 1 message was present (days 10-16). Forced expression (between days 16 and 25) of MafA, a pancreatic maturation factor, resulted in enhanced expression of insulin genes, glucose transporter 2 and glucokinase, and glucose-responsive insulin secretion. Day 20 cells implanted in vivo resulted in pancreatic-like cells. Together, our differentiation assay recapitulates the proceedings and behaviors of pancreatic development and will be valuable for future screening of beta cell effectors.
Combination of GLP-1 and Sodium Butyrate Promote Differentiation of Pancreatic Progenitor Cells into Insulin-Producing Cells

Abstract:We identified that nestin-EGFP-positive cells isolated by fluorescent-activated cell sorting (FACS) were downstream progenitor cells from colony-initiating pancreatic precursor cells. Under differentiation condition, these nestin-EGFP-positive cells could generate islet and neural cells. To investigate the conditions that allowed nestin-EGFP-positive progenitor cells (NPPCs) to efficaciously differentiate into insulin-producing cells in vitro, protocols were designed with glucagon-like peptide-1 (GLP-1) and the histone deacetylase inhibitor, sodium butyrate. We demonstrated that the combination of GLP-1 and sodium butyrate resulted in the increasing of levels of transcripts encoding pancreatic developmental factors and insulin. As a consequence, the amount of insulin-producing cells and insulin secretion were enhanced. These results indicated that NPPCs which were cultured in the presence of GLP-1 and sodium butyrate could specially differentiate into insulin-producing cells.
In vitro multilineage differentiation and self-renewal of single pancreatic colony-forming cells from adult C57BL/6 mice.

Abstract:In a previous study we established colony assays suitable for studying murine adult (2-4 months) pancreatic progenitor cells plated in semisolid media containing methylcellulose and extracellular matrix proteins. Using these assays, we found robust in vitro progenitor cell activities (multilineage differentiation and self-renewal) from pancreatic cells of adult mice in the CD-1 outbred background. However, it was not clear whether progenitor cell activities can be detected from inbred mice, a preferred mouse model for various genetic studies. It was also not clear whether a single cell is sufficient to self-renew. Here, we show that fluorescent activated cell sorting pancreatic CD133(+) but not CD133(-) cells from adult C57Bl/6 inbred mice are enriched for progenitor cells that self-renew and give rise to multilineage colonies in vitro. The number of cells in a colony is in proportion to its diameter. Around 60% of single handpicked 3-week-old colonies express trilineage markers, indicating most progenitors are tripotent for ductal, acinar, and endocrine lineage differentiation. Approximately 80% of primary (freshly sorted) colony-forming progenitor cells are capable of giving rise to secondary progenitors in vitro, indicating that a majority of the primary progenitors self-renew. A single cell is sufficient for self-renewal and a Wnt agonist, R-Spondin1, enhances the number of secondary progenitors from the primary progenitors. Together, our pancreatic colony assays allow quantitative analyses of progenitors at a single-cell level from inbred mice. These assays will be useful for elucidating in vitro mechanisms of pancreatic progenitor cell biology.

Abstract:Objective To study the induction of embryonic stem cell to insulin-producing cells and establish the basis for cell transplantation therapy in diabetes.Methods Embryonic stem cells(ESC) were amplified for 4 days in mouse embryonic fibroblast feed layer(MEF),then the amplified ESCs were detached from MEF and cultured in suspension cultures to form embryoid bodies(EB).The EB were cultivated in neural precursor cell selective media for about 4 to 5 days to differentiate to neural precursor cells.By adding nicotinamide,betacellulin,activin-A,hepatic growth factor and glucose in serum free insulin-producing cells(IPC) selective medium,the dithezone-positive IPC were harvested.Results The EB differentiated into endothelia-like cells in the first 2 days.The endothelia-like cells differentiated into neuron-like cells at later culture.86.5 % of the neuron-like cells were nestin positive.The nestin positive cells differentiated into insulin-producing cells when exposed in insulin-producing cell selective media for about 5 to 6 days.Conclusion It is demonstrated that the embryonic stem cell derived nestin positive cells can be induced into neural precursor and insulin-producing cells.

Abstract:Objective To investigate whether nestin-expressing cells are present in human fetal pancreas and the cells distribution. Methods Immuno- histochemical staining was used to determine the expression and distribution of nestin, insulin, glucagon and cytokeratin 19 (CK19) in the consecutive sections prepared from human fetal pancreas at 16 weeks of gestation. Results There were nestin-expressing cells in the 16-week human fetal pancreas. These cells resided in the islets of Langerhans, pancreatic ductal and acinic structures, and were the most abundant in both acini and small ducts. Some of the insulin and CK19 positively staining cells were positive for nestin expression. There were no nestin positive cells within the areas where glucagon-expressing cells resided. Conclusion The cells expressing nestin, a marker for pancreatic stem cells, may be a subpopulation of immature cells in the human fetal pancreas.

Abstract:OBJECTIVE:To verify the hypothesis that selected nestin positive cells derived from human fetal pancreas (according as medical ethnics) have surface markers similar to bone marrow mesenchymal stem cells (MSCs), and that these cells have multilineage potential.METHOD:The cell surface markers were determined by flow cytometry, and then the potential that these cells might be differentiated into adipocytes and osteoplasts were explored.RESULT:These cells have similar surface markers as MSCs of bone marrow origin. These cells was induced to differentiate into adipocytes and osteoplasts.CONCLUSION:Selected nestin positive cells derived from human fetal pancreas have certain characteristics of MSCs.
