Cytotoxicity Bioassay for Detection of Ciguatoxin

Abstract:The neuroblastoma cells were cultured in vitro.CTX at various doses,and ouabain and veratridine were added into the cultured cells.CTX can stimulate the sodium channel and induce cell death synergically with ouabain and veratridine.The culture was terminated at different time of coincubation and MTT,a viable cell dye,was used to measure quantitatively the viable cells in the form of optical density values at 570 nm.The linear relationship between P-CTX-1 dosage and optical density values at 570 nm was assayed.The results showed that the incubation time affected the linear relationship between P-CTX-1 dosage and cell viability.The regression equations at 10 h,15 h and 20 h were respectively y=-0.082 6x+0.923 0(R2=0.978 9),y=-0.044 3x+0.601 5(R2=0.962 1) and y=-0.020 4x+0.323 7(R2=0.928 1),where x means CTX dosage,while y means cell optical density values,and R means the regression coefficient.In conclusion,10 h is the best incubation time to detect ciguatoxin toxicity.The morphologic observation confirmed this result.
