Abstract:This project investigated the temperature effect on some chemical and biological characteristics of liquid swine manure under low level batch aeration conditions. An airflow rate of 0.129 L/min/L manure was used to aerate manure contained in reactors made of plexiglas tubes. Five temperatures, i.e. 5°C, 10°C, 15°C, 20°C, and 25°C, were examined. Data showed that manure pH would decrease with an increase in temperature. When temperature increased from 5°C to 25°C, the average oxidation-reduction potential decreased from +40 mV to m 60mV, resulting in a decrease in aerobic counts from 5.5 2 10 6 to 2.2 2 10 6 . This was accompanied by an increase in anaerobic bacterial counts (from 3.8 2 10 6 to 5.1 2 10 6 ). The increased anaerobic population produced more volatile fatty acids, leading to the decrease in pH. An inversely linear correlation was observed between pH and volatile fatty acids with a correlation coefficient of 0.8742. To prevent aerobic bacteria from decreasing and anaerobic bacteria from increasing, manure temperature should be kept under 15°C during aeration. To reduce odor generation potential (based on reduced VFA production), the oxidation-reduction potential should be maintained at +35 mV or higher.

Abstract:This study aimed to explore the effect of temperature on the persistence of fecal bacteria by multiple approaches in ambient anaerobic digestion systems treating swine manure. Both lab-scale (15 °C, 20 °C, and 25 °C) and field (26 °C on average) studies were conducted by high-throughput sequencing and culture-based methods. A community-wide Bayesian SourceTracker method was used to identify and estimate the fecal bacterial proportion in anaerobic effluent. High proportional contributions of fecal bacteria were observed in effluent at 15 °C (73%) and 20 °C (75%), while less was found at 25 °C (19%). This was further verified by a field study (23%) and an anaerobic reactor study at 37 °C (0.01%). To explore the potential reasons for differences in fecal bacterial proportions, bacterial taxa were divided into "lost" and "survivor" taxa in manure waste by LEfSe. The "survivor" taxa abundance was positively correlated with SourceTracker proportion (r = 0.913, P = 0.001), but negatively correlated with temperature (r = −0.826, P = 0.006). In addition, biomarkers in effluent were divided into "enriched" and "de novo" taxa. "Enriched" taxa, including acidogenic and acetogenic bacteria, were found at all temperatures, whereas taxa related to organic degradation were multiplied "de novo" at 25 °C. Variation partition analysis showed that temperature could explain 30% of variations in effluent bacterial community. Moreover, coliforms isolated from the manure and effluents at 15 °C and 20 °C were also phylogenetically related. This study provided comprehensive insight into the impact of temperature on the persistence of fecal bacteria in anaerobic effluent, with temperatures over 25 °C recommended to reduce fecal pollution.

Screening and application effect of microbial strains for duck manure fermentation

Abstract:Eight deodorizing strains and two fecal thermophilic strains were isolated from fresh duck feces samples. Among the 8 deodorizing strains, 3 strains with good deodorizing effect were selected and combined with 2 strains with high temperature, which were used to prepare compound microbial inoculum, and the composting effect was tested. In the composting experiment, natural composting was set as control, and there were two treatments with compound microbial inoculum. The duck manure was fermented and stacked. The results showed that the microbial inoculum treatment could obviously promote the temperature rise of duck manure fermentation, and the high temperature stage lasted for a long time. The C/N value of microbial inoculum treatment was 2.6 lower than that of natural stacking control, and the degree of maturity was higher. Compared with the control, the germination index reached the harmless standard five days earlier, the total nitrogen content increased by 4.08% compared with the natural composting, and the loss rate of nitrogen nutrient content was the lowest. Therefore, the strain composed of microbial inoculum 2 has certain application value in the treatment of duck manure waste.

Abstract:Anaerobic biological treatment at ambient temperature is a treatment technology for organic waste.Its advantages include low energy consumption,low microorganism growth rate and low sludge production.The anaerobic biological treatment were systemically and detailedly analyzed and summarized in this paper from temperature,technique and biotic-phase.Although there are some disadvantages in anaerobic biological treatment at ambient temperature due to the effect of temperature,for example,instability of treatment efficiency and low biogas production,it do not need the system of heating and warm keeping and can be improved in the technology and operation.This technology has attracted more and more attentions and the study and its application is increasing.

Abstract:The influence of temperature on chemical wastewater treatment by two-phase anaerobic digestion process was investigated.The pilot-scale experiment results show that temperature has a great influence on methanogenesis process.COD removal efficiency remains stable when the temperature is higher than 20 ℃,COD removal rate precipitously declines when the temperature is below 20 ℃.Methanogenesis function is lost when the temperature is equal to or less than 5 ℃ and products of acidogenesis are accumulated in methanogenesis stage with a decline of pH.Although temperature has a little influence on COD removal efficiency in acidogenesis,acidogenesis function is lost because microbial activity is greatly inhibited when the temperature drops to 0 ℃ or so.

Abstract:Sub-moderate temperature (ranging from low to moderate temperature) anaerobic digestion (AD) could balance fermentation efficiency and energy input. We investigated biogas production and the microbial community in wastewater AD at sub-moderate (15 °C, 20 °C, and 25 °C) and moderate (35 °C; control group) temperatures with the organic loading rate (OLR) incrementally increased over 200 days. The impact of temperature on biogas production was found to be minimal at a low OLR but became more significant at a high OLR. Notably, a temperature threshold ranging from 15 °C to 20 °C exerted a strong inhibitory effect on biogas production and disrupted the microbial community. And, SMT-AD is deemed by this study to be the optimal application strategy of wastewater with low temperature and low OLR. Bacterial richness was positively and linearly related to temperature. There is a relevance between methane production and archaeal diversity under the influence of temperature and OLR. Temperature and OLR shaped the ecological function of predominant bacteria. Anaerolineales, Thermotogales, and Lactobacillales were strongly influenced by temperature. Synergistales had a synergistic relationship with Desulfovibrionales. Clostridiales was responsible for acetate and butyrate production and closely related to Lactobacillales. Acetoclastic Methanosaetaceae was the predominant methanogen. Methanogens could survive and maintain their population even though methanogenesis was limited under high OLRs and low temperatures.

The Effect of Drying Temperature on Nitrogen Loss and Pathogen Removal in Laying Hen Manure

Abstract:Exhaust air from the poultry houses or ambient hot air are normally utilized to dry the laying hen manure extensively in the summer in China. The drying process can not only reduce the moisture of lay hen manure but can also have a degerming effect. However, the ammonia emission is considered as one of the major issues of laying hen manure drying and air pollution scouse. Then, it is not clear that whether the ammonia emission increased using the hot ambient air to dry laying hen manure in summer and whether increasing the temperature can inactivate more bacteria during low temperature drying process. Therefore, the main works of this study were to investigate the evolution of ammonium nitrogen (NH4-N) content, organic nitrogen (Org-N) content, and total bacteria count vs. time during the low-temperature drying process of laying hen manure at different drying temperatures. The results showed that increasing drying temperature can reduce the energy consumption of the manure drying system, but can increase the loss of NH4-N. The Org-N content among the three drying temperatures within same drying time was not significantly different (p > 0.05), which suggested that increasing the temperature did not accelerate the degradation of Org-N during low-temperature drying process. Low-temperature drying had weak destruction of bacteria in laying hen manure and the end dried manure still had a great number of bacteria.

Abstract:Synergistic effects among different chemical components under the anaerobic co-digestion (AcoD) process played an important role in improving its performance, which might be affected by the digesting temperature. The results showed that the actual methane production (AMP) and gasification rate (GR) of 50% lipid content were the highest, and the carbohydrate and protein content should be adjusted according to the temperature. Under mesophilic conditions, the M1 reactor with high protein content (carbohydrate–lipid–protein ratio, CLP = 20:50:30) had the highest AMP of 552.02 mL/g VS and GR of 74.72%. However, as the temperature increased, the high protein content produced high levels of ammonia nitrogen (AN) and free ammonia (FA), which formed a certain degree of ammonia inhibition, resulting in lower AMP and GR. Under thermophilic conditions, the low protein T2 reactor (CLP = 40:50:10) had the highest AMP and GR at 485.45 mL/g VS and 67.18%. In addition, the M1 and T2 reactors had the highest microbial diversity, which promoted substrate degradation and methane production. In the M1 reactor, acetoclastic metabolism is the main methanogenic pathway, while in the T2 reactor changes to hydrogenotrophic metabolism. Therefore, understanding the synergistic effect between temperature and chemical compositions was an effective way to improve the AcoD effect.

Abstract:The research and applications of anaerobic digestion (AD) in plateau areas are limited by the specific climatic conditions (e.g., low temperature, large temperature difference). To explore feasible and economical strategies of AD in these regions, the effects of different temperature modes (constant mode: 35, 25, 15 °C; stepwise changes mode that simulated daily environmental temperature changes: 35–25 °C, 25–15 °C) and substrate/inoculum (S/I) ratios (1: 1, 1: 2, 1: 4) on Tibetan food waste (FW) digestion performance and microbial community structure were analyzed. Besides this, the energy balance of AD under different conditions was estimated. The methane yields (335–463 mL g−1 VS) of FW at temperature changes mode with less energy consumption could reach the same level or even slightly higher than the methane yields at 35 °C. Kinetic analysis revealed that, compared with 25 °C, methane production performance was not greatly affected by temperature fluctuations around 25 °C. The relative abundance of acetoclastic methanogens (e.g., Methanosaeta) were dominant over hydrogenotrophic methanogens (e.g., Methanobacterium, Methanospirillum) at 35 °C. The opposite was true at 15 °C. The decrease in temperature caused the digestion pathway to shift from acetoclastic to the hydrogenotrophic pathway. The effect of temperature on the microbial community structure was more pronounced than the S/I ratio. Energy balance calculations showed that psychrophilic conditions and AD without temperature control were likely to be more economically competitive than mesophilic conditions for small‐scale AD in cold regions. © 2021 Society of Chemical Industry (SCI).

Abstract:Synthetic swine wastewater was treated by using anaerobic baffled reactor（ABR） with anaerobic granule sludge.The effects of low（15±1） ℃,medium（35±1） ℃,and high（50±1） ℃ temperature on the pollutant removal efficiency and the microbial community of ABR were investigated.The results showed that,at the medium temperature of 35 ℃,the removal efficiency of COD kept above 96%.While at the low and the high temperature,the COD removal efficiencies were about 70%.Temperature changes did not show obvious influence on the wastewater pH value in ABR.At medium temperature,the volatile fatty acid content was lower than that in other conditions,indicating the most stable condition.The results of fluorescence in situ hybridization（FISH） and scanning electron microscope showed that the total relative abundance of microbial community at 35 ℃ was 12% and 27%,respectively,higher than those at 15 ℃ and 50 ℃.

Abstract:Introduction: Aerobic composting is an effective method for utilizing chicken manure. However, its low carbon to nitrogen (C/N) ratio leads to slow heating and short high-temperature phases, which reduce composting efficiency and product quality. Methods: To address this issue, splinted mushroom cultivation residues were added to adjust the C/N ratio, and exogenous thermophilic composting strains were introduced to increase composting temperature. This study analyzed the relationship between physicochemical metabolites and microbial community structure during high-temperature chicken manure composting. Results and discussion: Based on metagenomic and physicochemical analyses, results showed that the exogenous microbial agents extended the thermophilic phase by three-times, reduced the heating phase duration by 75%, and increased nitrogen, phosphorus, potassium, and soluble organic carbon contents by 3.61, 21.63, 7.21, and 39.03%, respectively. Genes associated with amino acid metabolism were significantly enriched during the heating phase, while genes involved in the tricarboxylic acid cycle were more active in the thermophilic phase. During the thermophilic phase, bacterial diversity and richness decreased compared to the heating and cooling phases. Functional microbes such as Bacillus , Caldicoprobacter , and Virgibacillus showed a positive correlation with the key differential metabolites. While Actinomadura , Saccharomonospora , Paenibacillus , and Aneurinibacillus displayed an opposite correlation. Further experiments demonstrated that the increased temperature during the thermophilic phase triggered the upregulation of oleic acid metabolism and piperidine metabolism pathways in functional microorganisms, leading to the production of heat stabilizers and protective agents like oleic acid, gallic acid, and 2-piperidone. This phenomenon helped maintain microbial viability during the thermophilic phase and improved composting efficiency.

Abstract:Low temperature is one of the limiting factors for anaerobic digestion in cold regions. To improve the efficiency of anaerobic digestion for methane production in stationary reactors under low-temperature conditions, and to improve the structure of the microbial community for anaerobic digestion at low temperatures. We investigated the effects of different concentrations of exogenous Methanomicrobium (10, 20, 30%) and different volumes of carbon fiber carriers (0, 10, 20%) on gas production and microbial communities to improve the performance of low-temperature anaerobic digestion systems. The results show that the addition of 30% exogenous microorganisms and a 10% volume of carbon fiber carrier led to the highest daily (128.15 mL/g VS) and cumulative (576.62 mL/g VS) methane production. This treatment effectively reduced the concentrations of COD and organic acid, in addition to stabilizing the pH of the system. High-throughput sequencing analysis revealed that the dominant bacteria under these conditions were Acidobacteria and Firmicutes and the dominant archaea were Candidatus_Udaeobacter and Methanobacterium . While the abundance of microorganisms that metabolize organic acids was reduced, the functional abundance of hydrogenophilic methanogenic microorganisms was increased. Therefore, the synergistic effect of Methanomicrobium bioaugmentation with carbon fiber carriers can significantly improve the performance and efficiency of low-temperature anaerobic fermentation systems.

A cyclic shift-temperature operation method to train microbial communities of mesophilic anaerobic digestion

Abstract:Temperature is the critical factor affecting the efficiency and cost of anaerobic digestion (AD). The current work develops a shift-temperature AD (STAD) between 35 °C and 55 °C, intending to optimise microbial community and promote substrate conversion. The experimental results showed that severe inhibition of biogas production occurred when the temperature was firstly increased stepwise from 35 °C to 50 °C, whereas no inhibition was observed at the second warming cycle. When the organic load rate was increased to 6.37 g VS/L/d, the biogas yield of the STAD reached about 400 mL/g VS, nearly double that of the constant-temperature AD (CTAD). STAD promoted the proliferation of Methanosarcina (up to 57.32 %), while severely suppressed hydrogenophilic methanogens. However, when the temperature was shifted to 35 °C, most suppressed species recovered quickly and the excess propionic acid was quickly consumed. Metagenomic analysis showed that STAD also promoted gene enrichment related to pathways metabolism, membrane functions, and methyl-based methanogenesis.

Abstract:In this laboratory-scale study, the biological activities and vermicomposting potential of Eisenia fetida in pig manure were investigated to evaluate the influence of temperature (10°C–50 °C) on vermicomposting effect. The enzyme activities of catalase (CAT), peroxidase (POD), superoxide dismutase (SOD), and the content of malondialdehyde (MDA) in earthworm, macro-nutrients (N, P, K), total organic carbon (TOC), humification index (HIX) and C/N ratio in pig manure have been investigated simultaneously. Results revealed that CAT activity elevated initially and then declined with the increased temperature. The increased POD, SOD activity and MDA content indicated the antioxidant stress response of earthworm and the optimum temperature was found at 30 °C. After vermicomposting the pig manure for 60 d under temperature range of 10°C–50 °C, the vermicompost exhibited a significant increase in macro-nutrients, whereas a reduction was observed in TOC, C/N ratio and HIX, reflecting the efficient decomposition of organic waste driven by earthworms and microorganisms synergistically. The result of total microbial population showed the increase of actinomycetes and bacterial population. The enhancement percentage was in the order: 30 °C > 40 °C > 50 °C > 20 °C > 10 °C, revealing the impact of temperature. The results indicated the advantage of vermicomposting with Eisenia fetida and the regulation mechanism of temperature on the vermicomposting effect of pig manure. The present study will benefit to the utilization of agricultural wastes and achieving environmental sustainability.

Abstract:The application of anammox process in mainstream wastewater treatment process is still facing challenges especially at the low temperature. To resolve this problem, the coexistence system of anaerobic ammonia-oxidizing bacteria (AnAOB) and heterotrophic bacteria (HB) was built in this study. The nitrogen removal effidency mainly maintained at above 90% during the process of temperature reducing from 35 degrees C to 10 degrees C. The nitrogen removal rate were 0.30 g N.L-1.d(-1) at both 25 and 15 degrees C and 0.10 g N.L-1.d(-1) at 10 degrees C, respectively. Analysis of 165 rRNA genus sequencing revealed that as the temperature reduced to 10 degrees C, the Denutrotisoma genera presented a downward trend but Comamonadaceae genera showed an upward trend. At 10 degrees C, the contrast of anammox activities between granular and flocculent sludge in the system revealed that although the abundance of anammox genera was much lower in flocculent sludge than that in granular sludge, the anammox activities showed no significant discrepancy. And the abundance of Comamonadaceae and Chloroflexales genera were much higher in flocculent sludge than those in granular sludge, presenting their key roles to anammox activity at low temperature. The Circos diagram and Cluster of orthologous Group of protein functional predication showed that the functional abundance related to interaction among microbial communities were higher in flocculent sludge but those related to self-growth was higher in granular sludge. This result indicated the significance of the interactions based on the microbial diversity in the application of annamox process at low temperature. (C) 2020 Published by Elsevier B.V.

Abstract:The effects of thermal pretreatment at moderate temperatures (70, 80 and 90 degrees C) and high temperatures (120, 140 and 160 degrees C) over heating durations of 10-120 min on the physical and chemical properties of kitchen waste and on anaerobic digestion were investigated. The results show that thermal pretreatment significantly enhances the solubilisation of organic compounds (chemical oxygen demand, crude proteins, crude fats and volatile fatty acids) and their biodegradability during subsequent anaerobic digestion. High temperature and long heating duration are beneficial for the release and reduction of organic compounds, and the efficiency of subsequent anaerobic digestion is improved markedly under these conditions. Moreover, both the methane production rate and methane yield were observed to increase significantly at moderate treatment temperatures when the anaerobic digestion time was longer than 50 h. Abstract:Identifying the stability and phytotoxicity of liquid digestate (LD) is necessary for safe agricultural utilization. Storage temperature, method, and time are critical factors that affect the stability and phytotoxicity of LD. This study therefore aimed to explore the dynamics of stability, phytotoxicity, and microbial community of LD in cattle farms under different storage conditions. The results showed that the contents of solids, organic matter, nitrogen, and phosphorous decreased during storage and exhibited temperature dependency. Conversely, the seed germination index increased, which was negatively correlated with dissolved organic carbon and ammonium nitrogen and positively correlated with certain bacteria (Thermovirga and Fastidiosipila). Open storage and/or higher temperature were found to contribute to the stabilization efficiency and phytotoxicity disappearance of LD. Open storage of LD at 30 °C for 60 days and 20 °C for 90 days was safe for its agricultural utilization, while hermetic storage of LD at 30 °C for 120 days and 20 °C for 150 days was safe. However, for storage at 10 °C for 180 days, additional post-treatment is required.
Impact of Thermal Treatment on Biogas Production by Anaerobic Digestion of High-solid-content Swine Manure

Abstract:Livestock manure is a kind of waste with high organic content and sanitation risk. In order to investigate the impact of thermal treatment on the anaerobic digestion of high-solid-content swine manure, 70 degrees C thermal treatment was conducted to treat raw manure (solid content 27.6%) without any dilution. The results indicated that thermal treatment could reduce the organic matters and improve the performance of anaerobic digestion. When the thermal treatment time was 1d, 2d, 3d, 4d, the VS removal rates were 15.1%, 15.5%, 17.8% and 20.0%, respectively. The methane production rates (CH4/VSadd) were 284.4, 296.3, 309.2 and 264.4 mL x g(-1), which was enhanced by 49.7%, 55.9%, 62.7% and 39.2%, respectively. The highest methane production rate occurred when the thermal treatment time was 3d. The thermal treatment had an efficient impact on promoting the performance of methane production rate with a suitable energy consumption. On the other hand, thermal treatment could act as pasteurization. This showed that thermal treatment would be of great practical importance.

Abstract:Dissolved organic nitrogen (DON) formed by microbial metabolism in wastewater treatment processes adversely impacts wastewater reuse and receiving waters quality, and microbial metabolism is greatly influenced by temperatures. However, little is known about the effect of microorganisms on DON and bioavailable DON (ABDON) formation under low temperatures. In this study, six reactors were operated at low (8 °C and 15 °C) and room (25 °C) temperatures to evaluate the relationship between microbial activity, microbial communities, and DON and ABDON. Results showed that DON and ABDON concentrations significantly increased at low temperatures (p < 0.05, t-test). DON formation was significantly correlated to microbial activity only, with adenosine triphosphate (negative, r = -0.64) and polysaccharide (positive, r = 0.61) as key indicators; however, ABDON formation was influenced by both microbial activity (polysaccharide > triphenyltetrazolium chloride-dehydrogenases > adenosine triphosphate) and microbial community structure. Short-term tests using the biomass from six reactors were performed at room temperature to further validate the relationship. The distinct differences between these results provide a basis for different strategies on reducing effluent DON and ABDON under low temperatures.
